Model Systems for Investigating Root Responses to Localized Water Availability.

Water is distributed unevenly in soil, creating fine-scale patterns that shift with soil structure and drying. To navigate this complexity, roots deploy localized adaptive responses that allow them to sense and respond to water availability with high spatial precision. These include alterations in root branching (hydropatterning and xerobranching) and directional growth responses (hydrotropism and xerotropism), all of which collectively optimize water capture. Despite their importance, reproducing these heterogeneous water cues in the laboratory is challenging because soil systems are structur

Mapping Labile Fe2+ and Fe3+ in Living Arabidopsis thaliana Roots: Imaging and Quantification.

Iron (Fe) is involved in numerous key physiological processes due to its transfer or gain electron capacity. However, in excess, Fe is toxic to cells. The understanding of mechanisms governing its homeostasis requires imaging techniques capable of localizing and quantifying Fe and its redox state in situ. The Perls-DAB histochemical staining is a widely used method for visualizing Fe at a cellular level. Other advanced approaches, such as micro X-ray fluorescence and X-ray absorption near-edge structure spectroscopy, also provide information on the spatial distribution and oxidation state of F

Fluorescence and Ultrastructural Microscopy for Botrytis cinerea.

Botrytis cinerea, a notorious pathogenic fungus, leads to postharvest decay in horticultural crops and substantial economic losses, while the pathogenicity of B. cinerea is closely related to its structural characteristics and cellular homeostasis. Among the currently available techniques, fluorescence and ultrastructural observations serve as crucial tools for investigating subcellular structures and cellular events during B. cinerea growth. These techniques enable the monitoring of cell structural integrity, such as variations in the cell membrane and cell wall, as well as intracellular chan

Phospholipases D: An Update on Structure-Function Relationships.

Phospholipases D (PLD) are lipolytic and interfacial enzymes that hydrolyze the distal phosphodiester bond of phospholipid substrates. PLD are widespread in prokaryotes and eukaryotes and have various functions in bacteria, yeast, plants, and mammals. The PLD reaction frees phosphatidic acid (PA), which is an intermediate in the lipid pathway but also a signaling molecule involved in metabolic, cellular, and physiological processes. Depending on their primary structure, PLD belong to two different groups: the HKD family, characterized by an HXKX4D sequence that is often duplicated, and the non

Lipase-Catalyzed Synthesis of Nutritional Structured Lipids at Laboratory-Scale Using Crude or Refined Oils.

In this chapter, some examples of laboratory protocols to produce nutritional structured lipids, namely human milk fat substitutes, dietetic triacylglycerols, and interesterified fat blends with improved functional and rheological properties, catalyzed by either immobilized commercial or noncommercial lipase preparations, are presented. The use of crude oils instead of refined counterparts to reduce oil purification costs is also addressed. In addition to batch synthesis, the continuous production in packed- or fluidized-bed bioreactors is addressed, as well as the evaluation of operational st

Lipase-Catalyzed Pathways to the Synthesis of Pharmacologically Active Amides.

In this chapter, we describe the application of lipases as catalysts in the synthesis of structurally different amides with pharmacological activities. Numerous products, many of them new compounds, were obtained in very good to excellent yield and a highly regioselective manner. The influence of various reaction parameters in the enzymatic reactions, such as enzyme source, nucleophile/substrate ratio, enzyme/substrate ratio, solvent, and temperature, was examined for each case.

Bio-Inspired Silica Lipase Nanobiocatalysts for the Synthesis of Fatty Acid Methyl Esters.

Lipase immobilization strategies are key to the potential use of biological catalysts in reactions of interest. The bioinspired synthesis of silica nanoparticles uses biological or environmentally friendly processes to create highly functionalized supports with desirable properties such as increased surface area and stability. Here, we describe the preparation of two immobilization methods for Thermomyces lanuginosus lipase on biomimetic silica supports: in situ entrapment and covalent attachment to the hetero-functionalized surface of the nanoparticles, and their application in the synthesis

Viable Storage of Botrytis Conidia and Sclerotia.

Long-term storage of microorganisms is essential for maintaining a consistent laboratory strain collection. To prevent a reduction in viability of Botrytis stocks, we can make use of the naturally formed survival structures of this fungus, conidiospores and sclerotia. Conidiospore stocks are the preferred method of storage and can be stored in 20% glycerol at -80°C for up to 30 years. When conidiospore storage is not feasible, sclerotia can be stored at 0°C, and agar plugs containing mycelium can be stored at 4°C. Sclerotia and mycelium stocks will only last several years and thus require more

Biopolymers Derived from Solanum lycopersicum: Synthesis with CAL-B and Their Structural Analysis by Mass Spectrometry (DI-ESI and MALDI-TOF).

The main component of tomato cutin, 10,16-dihydroxyhexadecanoic acid, was obtained from agro-industrial waste and used to perform the polymerization reaction catalyzed by CAL-B, which is the best enzymatic reaction for producing linear oligomers; the oligomers were analyzed using direct infusion electrospray mass spectrometry (DI-ESI) and matrix-assisted laser desorption/ionization mass spectrometry (MALDI-TOF) techniques. The characterization of aliphatic biopolymers is challenging due to their chemical characteristics, which make them poorly accessible for mass spectrometry analysis. During

Phospholipases: Where Do We Stand?

Phospholipases are a diverse group of lipolytic enzymes that hydrolyze membrane glycerophospholipid substrates at specific ester bonds at the lipid-water interface. They are involved in numerous functions such as the production of various intracellular signaling molecules, inflammation, and human digestion. These highly widespread enzymes in nature vary considerably in terms of structure, mode of action, and regulation. Research on phospholipases has progressed greatly in recent decades. This review aims to provide a general framework of the current understanding of phospholipases and a discus

High-Resolution Detection of Sphingolipids During Chlamydia trachomatis Infection via Expansion Microscopy.

The obligate intracellular bacterial pathogen Chlamydia trachomatis establishes a membrane-bound compartment called the inclusion, which serves as a replicative niche, within host cells. Sphingolipids, a major class of eukaryotic membrane lipids, are acquired from the host and incorporated into chlamydial membranes. Visualization of individual bacteria within inclusions or sub-bacterial structures by fluorescence microscopy remains challenging due to the limited resolution of conventional light microscopy. Expansion microscopy (ExM), a super-resolution imaging technique, enables nanoscale reso

Near-Theoretical-Density Zinc Plating Enabled by an Ordered Janus Interphase for High-Energy Zinc Pouch Cells.

Achieving ultradense zinc plating at the theoretical limit is crucial for high-energy aqueous zinc metal batteries (AZMBs), but is hindered by severe interfacial instabilities such as dendrite growth and hydrogen evolution. Here we construct a nanometer-thin, vertically stratified organic-inorganic Janus bilayer (CF-LaF3) by ion layer epitaxy to pre-engineer the zinc anode interface. A hydrophobic perfluoroalkyl top layer accelerates Zn2+ desolvation, blocks active water and suppresses parasitic reactions, while a crystalline LaF3 bottom layer with high modulus, zincophilicity and continuous i

Graphene-Enhanced Magnetic Anisotropies in Fe/Ni (100) Superlattices with Monolayer Periodicity.

Fe/Ni(100) superlattices grown on graphene/Ir(100) by intercalation of alternate monolayers of Ni and Fe under graphene show magnetic properties severely modified. The graphene-iron interaction at the external interface produces a surface anisotropy Kint(gr/Fe) = 0.89 ± 0.08 mJ/m2, large enough to drive the uniaxial, out-of-plane magnetic anisotropy of the whole superlattice to KU = 0.87 ± 0.03 MJ/m3 and compensate the in-plane shape anisotropy characteristic of thin magnetic layers. Graphene also introduces an additional trigonal in-plane anisotropy superimposed to the biaxial anisotropy of t

Vicinal Halothiolcarbonates as Pseudo-Ambident Electrophiles: Carbamate Synthesis, Unified Thiol Reactivity, and Architecture-Dependent Antibacterial Activity of Ciprofloxacin Glycoconjugates.

Vicinal bis-electrophilic halothiolcarbonates (vic-HTCs) are introduced as a platform for the chemoselective synthesis of carbohydrate carbamates under mild conditions. Their aminolysis accommodates a broad range of nucleophiles, including complex bioactive amines, providing access to glycoconjugates that remain difficult to obtain by conventional methods. Beyond their synthetic utility, vic-HTCs display an unusual thiol reactivity in which exchange at a C(sp2) center and substitution at a remote C(sp3) site arise from a common tetrahedral intermediate. Combined experimental and DFT (Density F

Line-Graph Node Centrality in Retrosynthesis.

Molecular structures are naturally represented as graphs in which nodes correspond to atoms and edges to bonds, enabling the application of graph-theoretical and linear algebraic tools to chemical problems. In retrosynthetic analysis, the central challenge is to identify strategic bonds whose disconnection simplifies a target molecule into accessible precursors. While computational approaches to synthesis planning have evolved from rule-based systems to artificial intelligence platforms, there remains a need for interpretable, quantitative, structure-based metrics that guide bond disconnection

Detailed Insight into the Chignolin Folding Process from Maximally Informative Low-Resolution Representations of Its Isocommittor Hypersurfaces.

The synthetic miniprotein chignolin features a small size, a well-defined native structure, and a rather paradigmatic two-state folding transition. Nonetheless, this deceivingly simple molecule showcases a nontrivial folding pathway, whose detailed characterization has been the subject of several studies and still presents some open questions. In this work, we investigate chignolin by employing, for the first time in a combined and integrated pipeline, transition-path theory (TPT) and the mapping entropy optimization workflow (MEOW). The former describes the folding process in terms of its nat

Translating Spin-Adapted RPA to Spin-Adapted TDDFT.

Linear-response TDDFT based on open-shell references may produce spin-mixed excited states, particularly when the response space is not spin complete. Spin-adapted RPA was developed using tensor equation-of-motion and applying the Wigner-Eckart theorem with tensor decoupling. Casting the RPA Fock matrix and kernels as energy derivatives gives a TDDFT extension. To restore spin-component degeneracy and eliminate multicounting of spin correlation in pure exchange-correlation functionals, we use hybrids combining HF exchange with spin-unpolarized pure XC parts. Given that the mapping from spin-ad
