Neural Organization of Episodic Memory and Navigation in Children and Adults.

Episodic memory and spatial navigation are often viewed as interlinked, in part due to their shared reliance on medial-temporal lobe (MTL) structures. At the same time, there are important differences, for example, natural spatial navigation requires moving one's body through space while episodic memory does not. We investigated whether naturalistically encoded memories that were either episodic or additionally embedded in a navigational experience relied on overlapping or distinct neural networks. Children ages 8-13 years old and young adults viewed pictures of objects that they had previousl

High-Dimensional Multimodal Data Integration for Immune-Driven Diseases: A Practical Guide.

Recent advances in multiomics technologies have revolutionized the study of immune-driven diseases by enabling high-dimensional, single-cell resolution analyses. This chapter provides a practical guide for integrating CyTOF (mass cytometry) and single-cell RNA sequencing (scRNA-seq) data to address key challenges in this field. The integration of these modalities allows for consistent and reproducible cell type annotation, the transfer of annotations to assist in characterizing difficult-to-identify populations, and the transcriptional characterization of rare and heterogeneous subpopulations.

Naked-Eye Visual Loop-Mediated Isothermal Amplification Method for the Rapid Detection of Chikungunya Virus.

Chikungunya virus (CHIKV) represents a significant global health threat due to its recent resurgence. This protocol highlights the use of capillary-modified visual loop-mediated isothermal amplification (LAMP) technology for the rapid detection of CHIKV. We employed Vero cells for virus propagation and utilized the MolPure Viral DNA/RNA Kit for nucleic acid extraction, successfully amplifying extracted viral RNA at 63°C for 25 min. The presence of CHIKV was indicated by a distinct yellowish-green color change, demonstrating the effectiveness and simplicity of this protocol. Our protocol unders

Automated Counting of Chikungunya Virus-Formed Plaques.

The cytopathic effect (CPE) is a key to understanding the pathogenicity of the chikungunya virus (CHIKV) and is useful for evaluating viral infectivity in vitro. The plaque assay, based on the CPE induced by viral infection, provides an infectious virus titer for test samples. Additionally, the assay is simple, quantitative, and cost-effective. Thus, the plaque assay remains the gold standard for determining the infectivity of CHIKV. However, manually counting of virus-induced plaques with the naked eye or under the microscope is often time-consuming and labor-intensive. Therefore, automated p

Labeling and Detection of Peptidoglycan in Chlamydia and Chlamydia-Related Organisms Utilizing D-Alanine Analogs.

The incorporation and visualization of modified D-amino acids into the peptidoglycan (PG) of actively replicating microbes have made a tremendous impact on the field of bacterial physiology. Here, we describe the optimization of this technique for labeling PG in obligate, intracellular Chlamydia species as well as Chlamydia-like organisms. Fluorescent D-amino acids (FDAAs) are D-alanine analogs that readily incorporate into the stem peptides of living bacteria and can be visualized rapidly with a fluorescence microscope without the need for laborious postfixation procedures. By comparison, D-a

An Integrated Workflow for Culturing and AI-Based Profiling of Mouse Intestinal Epithelium.

Two-dimensional (2D) culture systems are powerful in vitro tools that have revolutionized the study of intestinal stem cell biology. These culture systems derived from mouse intestinal and colonic epithelia enable the modeling of tissue architecture, barrier function, and disease mechanisms under physiologically relevant conditions. This protocol details methods for isolating and maintaining mouse intestinal crypts as 2D monolayers. Monolayers are established via enzymatic dissociation and seeding onto ECM-functionalized hydrogels. This protocol also describes a deep learning pipeline that per

Beta Oscillations and Pupil-Linked Arousal Regulate the Updating of Cognitive Models.

The beta rhythm (neural oscillations in the range 13-30 Hz) is associated with the control of movement. Discrete movements (such as a button press) are bounded by a premovement dip in beta power and a postmovement beta rebound, which are thought to release and terminate the action, respectively. However, an additional or alternative role has been suggested for the beta rebound in regulating the extent to which sensory feedback updates the task model. Using EEG and pupillometry in human volunteers of both sexes, we developed a task that dissociates updating of a cognitive model from motor contr

The hormonal effects of owner-cat interactions on pet cats of different potential attachment styles.

OBJECTIVES: The aim of the present study was to investigate how different owner-cat interaction types influence stress and owner-bonding in pet cats with different potential attachment styles, using salivary cortisol and oxytocin as physiological indicators. METHODS: Twenty pet cats were classified into potential secure, insecure-anxious or insecure-avoidant attachment styles using the Secure Base Test. Owners engaged in four types of interactions with their cats at home on separate days. Saliva samples were collected before and after each interaction and analysed for cortisol and oxytocin co

Porcupine Quill Migration Into the Temporomandibular Joint Causing Trismus in a Dog.

Porcupine quill injuries have been reported in the veterinary literature as the source of a wide range of complications affecting separate body regions and organ systems, particularly in dogs from specific geographic locations. However, migration of porcupine quills involving the temporomandibular joint and its clinical consequences have not been previously reported. This report describes the first documented case of a dog presenting with trismus and a draining tract in the area of the left zygomatic arch several weeks following a porcupine encounter. The dog was referred as a consequence of t

Vessel Segmentation for Fundus Images Based on High-Resolution Networks with Gamma Correction and CLAHE Preprocessing.

Retinal vessel segmentation plays a crucial role in the early diagnosis and treatment of ophthalmic diseases, yet manual segmentation methods are time-consuming and prone to human error. The increasing demand for automated solutions has led to the exploration of deep learning-based approaches that improve segmentation accuracy and efficiency. However, existing methods often struggle with poor image resolution, a lack of robustness across datasets, and challenges in detecting fine vessel structures, limiting their applicability in large-scale clinical settings. To address these limitations, thi

The transposed-word effect persists across text orientations in Chinese.

In speeded grammaticality decision tasks, the transposed-word effect occurs when ungrammatical word sequences created by swapping adjacent words in an otherwise grammatical sentence are harder to reject than control sequences derived from ungrammatical base sentences, providing evidence for flexible word-order processing during reading. Previous research shows that this effect is often attenuated in atypical display formats, including serial word presentation and backward text, especially in RTs. Some accounts interpret this pattern as a shift from parallel processing, in which multiple words

Dimensional asymmetries in social attention: Dynamic head movements elicit opposite attentional biases in vertical versus horizontal dimensions.

When observing a person with direct gaze but a horizontally averted head posture, attention often shifts opposite to head direction (overshoot effect). We investigated whether this effect generalizes to vertical head movements and whether it depends on gaze direction information. Across four experiments, centrally presented faces shifted dynamically to averted head positions before a target appeared at a location congruent or incongruent with head direction. Vertical head movements (Experiments 1 and 2) produced head-congruency effects, which were stronger when the eyes were closed. In contras

Anterior segment optical coherence tomography-guided management of capsular contraction and anterior-posterior capsular adhesion after congenital cataract surgery: a case report.

BACKGROUND: Capsular changes after congenital cataract surgery may compromise the visual axis in infants and may necessitate secondary surgical intervention. In such situations, identifying a safe dissection plane can be challenging under conventional microscopic visualization, and adjunctive intraoperative imaging may be beneficial. CASE PRESENTATION: A 2-month-old boy diagnosed with bilateral congenital cataracts underwent phacoaspiration, posterior capsulotomy, and anterior vitrectomy with planned aphakia. Despite continued topical treatment, the patient developed progressive posterior syn
