Rhizoplane microbiome: niche-specific recruitment and plant defense priming against bacterial wilt disease.

The plant microbiome plays a pivotal role in host adaptation and disease suppression, yet niche-specific microbial responses to biotic stress, particularly within distinct plant compartments, remain poorly understood. Here, we revealed that bacterial wilt disease (BWD) induced pronounced niche-specific microbiome alterations in tobacco, with the rhizoplane (RP) emerging as a critical hub for beneficial microbial recruitment and defense coordination. Utilizing 16S and ITS amplicon sequencing across 6 distinct plant niches, we observed significantly enhanced bacterial diversity and a striking en

Current perspectives on natural and pharmacological interventions for combating drug-induced pulmonary toxicity.

Drug-induced pulmonary toxicity (DIPT) is a serious and often underappreciated complication that can emerge in patients receiving a wide variety of prescribed drugs, from cytotoxic chemotherapy agents and antiarrhythmics to immunomodulatory compounds. Four agents, in particular methotrexate (MTX), cyclophosphamide (CPA), bleomycin (BLM), and amiodarone (AMI) have the most extensive body of evidence documenting their pulmonary adverse effects, with each capable of producing distinct injury patterns that nonetheless share overlapping features. The mechanisms driving DIPT are far from simple; mul

Parental Social Environment Has no Effect on Offspring Development in the Dung Beetle: A Test of Adult Sex Ratio Effects.

The adult sex ratio (ASR) is a key demographic parameter that shapes sexual selection and social interactions. While ASR variation drives profound behavioral plasticity within a generation, it remains unclear whether parental experience of skewed ASRs influences offspring development via transgenerational plasticity (TGP), and if so, which of the two core nongenetic pathways: pre-zygotic germline-mediated information transfer and post-zygotic parental investment, predominates. To disentangle these mechanistically distinct pathways, we conducted a controlled egg-transplantation experiment in th

Scaffold hopping yields novel furo[3,2-d]pyrimidine NNRTIs with optimized antiviral potency, enhanced selectivity, and reduced hERG liability.

Considering the exceptional anti-HIV-1 potency of rilpivirine (RPV) against diverse mutant strains and its remarkable human ether-a-go-go related gene (hERG) potassium channel inhibition (IC50 = 0.50 μmol/L) as well as low selectivity (SI = 3989), a series of novel furo-[3,2-d]pyrimidine derivatives were rationally designed through a scaffold hopping strategy. Encouragingly, compound 10 revealed a striking reduction in hERG channel inhibition (IC50 > 30 μmol/L) and significant increase in selectivity (SI = 161580). Notably, 10 exhibited excellent antiviral activity against various HIV-1 strain

Are different populations fairly represented in single-cell omic atlases?

Single-cell omic atlases are transforming biology and medicine, yet their demographic representativeness has not been systematically evaluated. We analyzed >13,500 samples from the Human Cell Atlas (HCA), Human Tumor Atlas Network (HTAN), and PsychAD Consortium. Benchmarking against global and US general and disease-prevalence data, we found a striking, pervasive European overrepresentation and underrepresentation of Asian and Latino individuals. Nearly 70% of HCA samples lacked ancestry annotation, and among annotated samples, Europeans were overrepresented 6-fold. PsychAD was nearly two-thir

Rhizosphere Microbiome Dynamics Associated with Root Rot in Polygonatum kingianum Coll.

Polygonatum kingianum Coll. (PKC) is a valuable medicinal herb native to Yunnan, China, but its yield and quality are severely threatened by root rot. To elucidate the rhizosphere microbial dynamics associated with this disease and to identify location-transcending patterns, we collected rhizosphere soils from healthy and diseased PKC plants at three planting bases in Lincang, Qujing, and Kunming. Soil properties were measured, and bacterial (16S rRNA) and fungal (ITS) communities were characterized by amplicon sequencing. Geographic origin emerged as the dominant factor associated with microb

The efficient plant genetic transformation system mediated by FELBs and RTBs respectively via novel somatic embryogenesis in Allium sativum.

INTRODUCTION: Somatic embryogenesis represents an indirect but highly efficient biotechnological method for plant regeneration, genetic transformation, and virus-free propagation. METHODS: Herein, we investigated the effects of NAA, 2,4-D, and TDZ on the morphogenic capacity of Allium sativum. This is evaluated in terms of the formation of frog egg-like body and rhizoid tuber morphological structures, somatic embryo development, shoot regeneration, and the genetic transformation method. RESULTS: Two novel high-efficiency SE systems in garlic were characterized, operating through the FELB and

Structure-activity of outer membrane proteins in native bacterial membrane vesicles by solid-state NMR.

The outer membranes (OMs) of bacterial pathogens are potent virulence factors and serve as the first line of defense against host immunity. Their striking bilayer asymmetry is essential for function but poses an exceptional challenge for reconstitution in vitro, limiting structure-activity analysis to artificial non-native platforms that can interfere with structure and function. Here, we describe bacterial OM vesicles (OMVs), natively secreted from the cellular OM during bacterial cell growth and development, as an effective vehicle for structure-activity analysis in situ based on solid-state

Integrated transcriptomic and metabolomic profiling reveals coordinated regulatory networks associated with mosaic disease resistance in sugarcane.

INTRODUCTION: Sugarcane mosaic disease (SCMD) poses a severe threat to global sugarcane yield. Since conventional field management is insufficient to restrict viral transmission, unraveling the underlying defense mechanisms is imperative for targeted breeding. The primary objective of this study was to delineate the molecular and metabolic networks governing SCMD resistance by comparing highly resistant (XIDAZHE10-19, YT94-128) and susceptible (HP, XTT22) cultivars. METHODS: We employed metabolomic profiling and integrated transcriptomic data to investigate the genetic basis driving host resp

Potential involvement of persistently infected cells in protective immunity of grouper fish convalesced from nervous necrosis virus infection.

Nervous necrosis virus (NNV) is a simple RNA virus with high pathogenicity across over 170 fish species. Two experimental models have been developed to efficiently induce convalescent fish using live viruses: (1) pre-administering polyinosinic:polycytidylic acid (poly(I:C)) to induce a non-specific antiviral state, and (2) maintaining fish at suboptimal temperatures for disease onset. Notably, these convalescent fish exhibit robust long-term protective immunity despite having undetectable levels of neutralizing antibodies. This study synthesizes established in vivo and in vitro data on the kin

Embryonic Torsion and Axial Rotation in Developing Amniotes: Pattern Distribution, Functional Aspects, and Evolutionary Significance.

Early amniote development includes a striking but underappreciated event: rightward torsion ("twisting") of the head and trunk followed by axial rotation that positions the embryo's left side against the yolk. Although well documented in the domestic chicken, the evolutionary distribution and biological significance of this rotational pattern have never been evaluated systematically. Reports of supposed "leftward" rotation in some reptiles and birds, as well as a reported absence of rotation in some reptilian species, have further suggested interspecific variability. Here we present a comprehe

Non-invasive brain stimulation to prevent aging-related diseases: What does photobiomodulation offer compared with other methods.

The issue of the maintenance of cognitive and brain health across ageing is a striking concern in our ageing societies. Non-invasive brain stimulation methods have been proposed as tools to fight against various neurological pathologies and to this aim, could also be used against age-related brain changes. In this chapter, we examine the potential of transcranial photobiomodulation (tPBM), in comparison with other neurostimulation methods, to improve brain function and protect neurons. In particular, it is hypothesised that the double action of tPBM - both on neuronal function and at a more sy

Leaf anatomical traits support classification of Galphimioid clades in Malpighiaceae.

BACKGROUND AND AIMS: Laticifers represent one of the most functionally striking secretory systems in angiosperms, characterizing entire families and being the latex considered a key element in defense against herbivores in plants. However, they are still understudied, especially in clades where the secretions are not so copious, which is the case of the Malpighiaceae. Here, we undertake a study combining anatomy, phylogeny, ancestral state reconstruction to unravel the structure, distribution, and evolution of laticifers and other leaf anatomical traits in Galphimioid clade (Malpighiaceae), sa

Striking two targets with one scaffold via benzo[d]imidazole-isatin conjugates as potent dual VEGFR-2/c-MET antagonists.

A series of benzo[d]imidazole-isatin hybrids (4a-d, 6a-d, and 8a-d) was developed and synthesized as prospective dual inhibitors of VEGFR-2 and c-MET to address tumor growth and resistance-related signaling pathways. The biological assessment demonstrated encouraging dual kinase inhibitory action, with compound 8b identified as the most balanced inhibitor, displaying IC50 values of 92 nM and 63 nM against VEGFR-2 and c-MET, respectively. In-vitro antiproliferative evaluation against MDA-MB-231 and A549 cancer cell lines revealed notable cytotoxicity for compound 8b, yielding IC50 values of 2.3

Deciphering podophyllotoxin-mediated defense against downy blight in Litchi chinensis Sonn. via integrated metabolomics and transcriptomics.

Lychee (Litchi chinensis Sonn.) is a commercially important subtropical fruit. Lychee downy blight (LDB), caused by Peronophythora litchii, poses a major threat to lychee production. Here, we identified the LDB-resistant cultivar Yurong (YR) and the susceptible cultivar Guiwei (GW) from 288 global lychee germplasms. Widely targeted metabolomics analysis revealed a striking accumulation of podophyllotoxin (PTOX) in YR. Both in vitro and in vivo assays demonstrated that PTOX effectively inhibited the growth of P. litchii. Further investigation identified LcOMT1 as a key gene encoding an O-methyl

Instagram v. Reality: The Case for Regulating Adolescent Social Media Use.

In the span of just two decades, social media has transformed from a novel communication platform into an omnipresent force in modern life, fundamentally altering how we connect, communicate, and consume information. For adolescents born into this digital landscape, social media is not merely a tool but an integral component of identity formation, social development, and daily existence. Most teens ages thirteen to seventeen use social media daily, and over forty percent of Instagram's user base is under the age of twenty-two. Yet as platforms have evolved to capture and maintain user attentio

Genetic control of a multifunctional trait: The central role of a fatty acid synthase gene in regulating survival and sexual communication.

The genetic basis of multifunctional traits shaped by both natural and sexual selection remains poorly understood. In insects, cuticular hydrocarbons (CHCs) are an excellent example of such traits, providing protection against different micro-climatic conditions while simultaneously encoding cues predominantly used in sexual signaling. The fatty acid synthase (fas) gene family has been implied as an important cornerstone in initiating and maintaining CHC functionality, whereas their exact biosynthetic and regulatory mechanisms have remained largely elusive. Here, we characterize a single fatty

GWAS With a Global Population of Wheat Stripe Rust Identified High-Confidence Avirulence Gene Candidates.

Wheat stripe rust, caused by Puccinia striiformis f. sp. tritici (Pst), is a globally destructive disease characterised by rapid virulence evolution. Despite its significance, the genetic basis of avirulence variation at a global scale remains poorly understood. In this study, we integrated virulence phenotypes against 18 Yr resistance genes with genome-wide variation from a worldwide collection of Pst isolates to resolve the population-genomic architecture of avirulence. We identified multiple avirulence-associated genomic regions and defined 69 candidate effectors, with 11 high-confidence ca

Understanding Autistic Adults' Psychological Experiences of Anxiety Problems: A Qualitative Interview Study.

Our understanding of psychological experiences of anxiety in autism could be improved if it were shaped by autistic individuals. However, anxiety research in autism often begins with cognitive conceptualisations drawn from neurotypical or non-autistic individuals. This qualitative study aimed to explore psychological experiences of anxiety using semi-structured one-to-one interviews with a diverse sample of 20 U.K.-based, autistic adults, aged 18-55 years. Transcripts were analysed using two complementary methods: conceptual content analysis, where existing theories were tested against the dat
