The temperature dependence of permeation across human red cell membranes has been determined for a series of hydrophilic and lipophilic solutes, including urea and two methyl substituted derivatives, all the straight-chain amides from formamide through valeramide and the two isomers, isobutyramide and isovaleramide. A marker-specific and strongly temperature-dependent reaction was observed to occur at a time during transformation in Diplococcus pneumoniae after the donor deoxyribonucleic acid (DNA) had acquired single-strand properties and immediately preceding the integration of these strands into the recipient chromosome. Operationally, it was observed as the prevention of an intracellular inactivation process, also described in this paper, which is specific for low molecular weight or for damaged DNA, and which occurs if the recipient cells are held at suboptimal temperatures after the DNA has entered.

Angular displacement from linear overlap of but a few degrees in the transition state of the enzyme-substrate complex has been postulated to be of great kinetic significance ("orbital steering"). Several antibiotic resistance markers in Bacillus subtilis have been mapped by three-factor transduction crosses using bacteriophage PBS1. These markers, which are thought to be located in genes coding for ribosomal proteins, are within a segment comprising about 5 per cent of the B. subtilis genetic map, close to the major group of rRNA cistrons. The order of genetic makers in this region was found to be cysA14 mic-1 dagger str-1 (ery-1, ole-2) spc-2 bry-2 lin-2. The positions of neo-2, nea-1, and kan-2 are uncertain, although they are located in this region to the right of cysA14.

The kinetics of initiation of germination and inactivation by hydrostatic pressure of phosphate-buffered Bacillus pumilus spores is shown to be a consecutive first-order process at 25 C. The rate of recovery of UV-irradiated Hemophilus influenzae from acriflavine-sensitized loss of colony-forming ability was studied at various acriflavine concentrations, UV doses, and temperatures. The osmotic permeability coefficient for water has been measured for the Ehrlich mouse ascites tumor cell. Measurements were made of the rate of cell shrinkage in hyperosmotic solutions of NaCI, a functionally impermeable solute. During the first 9 months of weekly serial transplantation the mean was 6.4 micro(3)/micro(3)/atm. +/- 0.8 (S.E.). Rhodopsin, the red photosensitive pigment of rod vision, is composed of a specific cis isomer of retinene, neo-b (11-cis), joined as chromophore to a colorless protein, opsin. We have investigated the thermal denaturation of cattle rhodopsin and opsin in aqueous digitonin solution, and in isolated rod outer limbs. Both rhodopsin and opsin are more stable in rods than in solution. In solution as well as in rods, moreover, rhodopsin is considerably more stable than opsin.
