Genomic evolution of pancreatic cancer at single-cell resolution.

Most evolutionary studies on pancreatic cancer rely on bulk sequencing, yet clonal evolution happens at the single-cell level. We used single-nucleus DNA sequencing to study 137,491 single nuclei from 24 pancreatic neoplasms reflecting various clinical scenarios. We found higher frequencies of somatic alterations to driver genes that bulk studies indicate; many manifest as copy number alterations and account for the majority of spatial heterogeneity. In pancreatic cancers with canonical KRAS oncogenic mutations, we found likely varied dependence on the genotype that may signify differential re

Roars, Rumbles, and Resonance: A Systematic Review and Meta-Analysis of Crocodylian Acoustic Signals.

Crocodylians are highly vocal reptiles, possessing a complex acoustic signalling system including vocal and non-vocal signals used for courtship, mating, mediating conflict, and providing maternal care. Despite this, research on crocodylian acoustic signalling remains infrequent, with methodologies and terminology varying widely across studies. Here, we systematically review the literature and standardise crocodylian acoustic signal descriptions, measurements, and terminology to establish a consistent foundation for future research. The compiled dataset included 22 publications, with 623 acous

Integrated analysis of lncRNA-miRNA-mRNA networks reveals stage-specific molecular regulation of oogenesis in Chinese alligator (Alligator sinensis).

BACKGROUND: Reptiles, as the earliest vertebrate group to fully adapt to terrestrial environments, have reproductive strategies that hold critical significance for understanding biological evolution and ecological adaptation. However, the mechanisms of oogenesis in reptiles, particularly in the endangered Chinese alligator (Alligator sinensis), remain poorly understood. RESULTS: Here we integrated transcriptomic and non-coding RNA analyses to elucidate stage-specific lncRNA/miRNA-mRNA regulatory networks during oogenesis. Gonadal tissue samples were collected at three critical developmental s

Functional Morphology of the Archosaur Neck Provides Evolutionary Insights Into the Avian Neck Flexibility.

The amniote neck has evolved extensive mobility and this has contributed to functional and ecological diversification across the clade. Among amniotes, extant birds exhibit an S-shaped neck with multiple flexion points and displays significant variation in vertebral counts and morphology, enabling diverse foraging and feeding behaviors. While avian neck flexibility has increasingly attracted attention in ethology and functional morphology in recent years, its evolution within the archosaur lineage leading to birds remains unclear. Here, we quantified the ratio of the lever lengths (RLL) of sel

Global Distribution and Dispersal Pathways of Riparian Invasives: Perspectives Using Alligator Weed (Alternanthera philoxeroides (Mart.) Griseb.) as a Model.

In struggling against invasive species ravaging riverscape ecosystems, gaps in dispersal pathway knowledge and fragmented approaches across scales have long stalled effective riparian management worldwide. To reduce these limitations and enhance invasion management strategies, selecting appropriate alien species as models for in-depth pathway analysis is essential. Alternanthera philoxeroides (Mart.) Griseb. (alligator weed) emerges as an exemplary model species, boasting an invasion record of around 120 years spanning five continents worldwide, supported by genetic evidence of repeated introd

Autocrine activity of engineered IL-33 mRNA enhances adoptive T-cell therapy for peritoneal carcinomatosis and synergizes with IL-12 mRNA.

Rationale: Peritoneal carcinomatosis (PC) remains a major clinical challenge with limited therapeutic options across tumor types. Adoptive cell therapy (ACT) with tumor-specific T cells offers promise, but its efficacy is often impaired by the immunosuppressive tumor microenvironment (TME). Intraperitoneal ACT is under investigation to improve its effectiveness against metastases within the peritoneal cavity. IL-33, a cytokine of the IL-1 family, plays dual roles in immunity and inflammation and may enhance antitumor responses. We evaluated whether IL-33 mRNA-engineered T cells improve ACT eff

The effect of hibernation on reproduction performance of Alligator sinensis, Chinese Alligator is not limited to maintenance of matural follicle, but also involves the fundamental resource of germ cells.

Hibernation, acknowledged as a survival strategy under low-temperatures, is regarded as exert a substantial impact on reproductive performance. However, numerous enigmas remain concerning the underlying mechanisms of this interaction. Ultrasound scanning was employed to quantify follicle numbers, and assay of plasma neurotransmitters and steroid hormones were conducted to elucidate the dynamic correlations between follicle development and endocrine fluctuations during the hibernation period, encompassing onset of hibernation (OH), deep hibernation (DH), and ending of hibernation (EH). On the Origin and Evolution of Sweet Taste Mediated by Tas1r2-Tas1r3 in Vertebrates.

Sweet taste is a crucial chemosensory modality for detecting natural sugar compounds, which are primarily derived from angiosperms. In vertebrates, excluding birds, sweet taste is typically mediated by the Tas1r2-Tas1r3 heterodimer, and the receptor function often reflects dietary adaptations to sugar-rich diets. To gain insight into early vertebrate dietary transitions, we identified Tas1r genes in 58 vertebrate species and one outgroup and conducted functional assays in 10 representative species spanning six major clades, including one coelacanth, two amphibians, one squamate, two turtles, t

Neoadjuvant modified FOLFIRINOX plus nivolumab in borderline-resectable pancreatic ductal adenocarcinoma: a pilot phase 1 trial.

Chemotherapy and immune checkpoint inhibitor combinations have failed to improve survival in pancreatic ductal adenocarcinoma (PDAC), except in rare microsatellite instability-high cases; most studies focused on advanced disease. Here, we present clinical and translational results from a single-arm, prospective phase 1b/2 investigator-initiated study (NCT03970252) evaluating neoadjuvant modified FOLFIRINOX (mFFX) plus nivolumab in patients with borderline-resectable PDAC. The co-primary endpoints of safety and pathological response rate were met, with 22 (79%) of 28 patients proceeding to surg

Sequential platinum and PARP Inhibition enhances PD1 immunotherapy efficacy in murine Brca2 mutated pancreatic cancer.

Pancreatic ductal adenocarcinoma (PDAC) is a challenging malignancy to treat, but emerging evidence suggests that specific subtypes may respond more favorably to certain therapies. BRCA-mutated PDAC represents a distinct subtype that is particularly sensitive to DNA-damaging therapies. The current standard of care for advanced BRCA-mutated PDAC involves induction platinum-based chemotherapy followed by maintenance therapy with a poly (ADP-ribose) polymerase inhibitor (PARPi). However, the randomized phase III POLO trial, upon which this standard is based, did not demonstrate an improved overal

The developing alligator tongue undergoes a soft form of cornification associated with intermediate filament keratins.

Little is known on the keratinisation and cornification in the alligator tongue. This process has been studied during late and pre-hatching embryonic stages, using histochemical, immunolabelling and electron microscopy. The study revealed that PAS-positive, Alcian blue and Blue Nile Sulphate reactive glycoprotein/glycolipids are produced and accumulated in the corneous layer of the lingual epithelium before hatching. Also, Intermediate Filament Keratins (IFKs) but not Corneous Beta Proteins (CBPs) are synthesised in the tongue epithelium. Alligator IFKs have variable composition and cysteine c

United by chewing: Hunter-Schreger band-like pattern and wavy enamel in a fossil crocodile suggest functional convergence with mammals and dinosaurs.

Tooth enamel of most mammals shows alternating light and dark bands, called Hunter-Schreger bands (HSB), in longitudinal sections caused by decussating bundles of prisms, the unit building blocks of mammalian enamel. HSB are thought to increase resistance to abrasive food and mitigate crack propagation and hence are considered a mammalian adaptation to high-efficiency mastication. Using traditional microscopy techniques as well as X-ray diffraction computed tomography (XRD-CT), here we report for the first time the presence of HSB-like features in the tooth enamel of a non-mammalian amniote, I

Convergence for Inactivation of TGFβ Signaling Is a Common Feature of Advanced Pancreatic Cancer.

UNLABELLED: We performed whole-exome sequencing of 250 unique tumor tissues from 30 multi-region sampled pancreatic cancer research autopsies from patients diagnosed with advanced-stage disease. Convergent evolution within the TGFβ pathway is a common feature of advanced-stage disease. However, SMAD4 inactivation is more common among de novo metastatic pancreatic ductal adenocarcinomas (PDAC), whereas inactivation of TGFβ surface receptors is more common among locally advanced nonmetastatic cancers. These differences in metastatic propensity were orthogonally validated in mice by orthotopic in

Dual pancreatic carcinomas: clonally related or independent primaries?

Dual pancreatic cancers, either synchronous or metachronous in presentation, are a rare occurrence. It remains unclear if these lesions are clonally related or independently occurring primary malignancies. Herein we present a cohort (N = 22) with dual pancreatic ductal adenocarcinoma (PDAC) tumors to resolve previously conflicting reports and interrogate the underlying biological and clinical characteristics of this patient population. Next-generation sequencing of paired lesions (N = 10) revealed that while most dual PDAC are clonally related, independently occurring lesions do occur, irrespe

Chemical armed allies promote plant resistance to pathogen: The contribution of arbuscular mycorrhizal fungi to an invasive weed.

Invasive weeds often possess strong resistance to biotic stresses, which causes huge ecological problems. Both plant growth regulators (PGRs) and arbuscular mycorrhizal (AM) fungi contribute to plant growth and resistance. However, their combined interactions in invasive plants' defense remain poorly understood. To address this knowledge gap, the invasive weed Alternanthera philoxeroides was treated with gibberellins (GA) and paclobutrazol (PAC), inoculated with Clariodeoglous etunicatum to test its response to pathogenic fungi. We found that these two PGRs suppressed AM fungi colonization. Bo

Acute and Seasonal Effects of Temperature on Bite Performance of Alligator Snapping Turtles (Macrochelys temminckii).

Environmental temperatures influence most aspects of ectotherm biology, especially when fluctuating on daily and seasonal scales. The Alligator Snapping Turtle (Macrochelys temminckii) is a species of conservation concern inhabiting temperate latitudes in the southeastern United States. To study the effect of temperature and season on the bite performance of M. temminckii, we used a force transducer and high-speed videography to measure bite force and kinematics, including bite duration, jaw velocity and acceleration, and lunge velocity and acceleration, for 21 captive subadult M. temminckii.

Comprehensive image dataset of flexible pavement: Alligator cracks and edge-breaks from national highway (N6) of urban areas.

This data article introduces a structured pavement surface image dataset developed to advance research in automated pavement condition assessment and data-driven road infrastructure monitoring. The dataset comprises three distinct pavement condition categories: (i) alligator cracking, (ii) edge-breaking distress, and (iii) undamaged (intact) pavement surfaces, each representing a prevalent form of pavement deterioration or intact condition typically observed in flexible pavements. The dataset consists of 12,000 raw images (4000 per class) collected under real-world conditions. These images rep

Regional variation in mercury bioaccumulation of Alligator Gar (Atractosteus spatula): A case study from inland and coastal reaches of the Brazos River, Texas, USA.

Alligator Gar (Atractosteus spatula) are a species of conservation concern due to habitat alteration and harvest-oriented fishing activities. An additional concern is contaminant exposure, particularly mercury (Hg), which may pose reproductive risks to this long-lived predator. We quantified total mercury (THg:mg/kg wet weight) in Alligator Gar populations in one coastal and one inland region of the Brazos River, Texas, USA to explore accumulation patterns, consumption risks, and potential reproductive impairment. Furthermore, we included an analysis of stable isotopes of nitrogen (δ15N) and c

Bridging the gap: A systematic approach to integrating serum and plasma proteomic datasets for biomarker studies.

Serum and plasma are widely used in proteomic biomarker discovery, but differences between their proteomes have hindered the integration of data from the two specimen types. Here, we describe a computational approach for bridging between serum and plasma proteomic measurements derived from the aptamer-based SomaScan assay. We aimed to enable cross-specimen data utilization in the context of the PROphet model designed to predict immunotherapy outcomes based on 388 plasma proteomic biomarkers. Proteomic profiling of 7289 proteins was performed on 177 matched serum-plasma sample pairs from cancer

Endocranial anatomy of the earliest Cretaceous European neosuchian crocodyliform Pholidosaurus purbeckensis provides new evidence for the ecological evolution of Pholidosauridae.

The neosuchian crocodyliform clade Pholidosauridae had a near-cosmopolitan distribution, spanning the Late Jurassic to the early Paleocene. Representatives of the group inhabited aquatic environments, ranging from freshwater to potentially fully marine forms. The phylogenetic placement of Pholidosauridae within Neosuchia remains debated: whereas most analyses place it as closely related to Dyrosauridae, some studies argue for a closer relationship with Goniopholididae. One skeletal region that could shed light on both the phylogenetic position of Pholidosauridae, as well as how it achieved its
