Biochar-Straw Interaction Drives Soil Carbon Accrual by Modulating Microbial Metabolic Pathways.

Straw and biochar additions can enhance long-term carbon storage, yet how their interactions regulate carbon mineralization and allocation remains unclear. We conducted a 360-day microcosm incubation employing a reciprocal 13C-labeling design that paired 13C-labeled straw with unlabeled biochar and unlabeled straw with 13C-labeled biochar and used single-cell Raman spectroscopy and 13C-labeled cell metagenomics. At day 360, biochar reduced cumulative straw-derived 13CO2 from 1391 to 1199 mg C kg-1 soil, while increasing straw-C allocation to fulvic acid (FA) and humic acid (HA) fractions. This

Compartment-specific oxylipin-derived carbonyl profiles in spinach chloroplasts.

The first compartment-resolved map of chloroplast carbonyls was established, revealing distinct carbonyl compositions and patterns of accumulation in thylakoids, envelopes, and stroma. Oxylipin-derived carbonyls, diverse aldehydes and ketones generated from oxygenated lipids, function as key cytotoxic agents and signaling molecules in oxidative stress responses of plants. However, the intracellular and sub-organellar sites at which individual carbonyl species occur remain poorly defined. Here, we analyzed carbonyl compositions in spinach (Spinacia oleracea L.) leaves, isolated chloroplasts, an

Common Nitrification Inhibitors Exhibit Distinct Mechanisms on the Ammonia-Oxidizing Archaeon Nitrososphaera viennensis.

Microbial ammonia oxidation, the first and rate-limiting step of nitrification, plays a central role in soil nitrogen cycling. It is most relevant in agricultural soils as nitrifiers compete with crops for ammonia-based fertilizers. Therefore, synthetic nitrification inhibitors are widely used alongside fertilizers to reduce the activities of dominant drivers of this process, i.e., ammonia-oxidizing archaea (AOA) and bacteria (AOB). However, the physiological responses of ammonia oxidizers remain poorly resolved. Here, the response of the AOA Nitrososphaera viennensis to the nitrification inhi

Unveiling the resistance risk and molecular mechanism of Xanthomonas oryzae pv. oryzicola to zhongshengmycin.

Zhongshengmycin, an N-aminoglycoside agricultural antibiotic, has been widely used to control bacterial leaf streak (BLS) caused by Xanthomonas oryzae pv. oryzicola (Xoc) in China. However, the resistance risk and its underlying molecular mechanisms remain poorly understood. In this study, we determined the sensitivity of 101 Xoc isolates collected from major rice-growing regions in China, with a mean EC50 value of 0.7005 ± 0.2407 μg/mL; no naturally occurring resistant strains were detected. Five resistant mutants were obtained through laboratory adaptation, and these mutants exhibited signif

Synthesis, Fungicidal Activity, and Action Mode of Functionalized Siderophores.

Iron homeostasis influences fungal pathogenicity and is a promising target for crop protection. In this study, catechol moieties were incorporated into G2-class fungicidal scaffolds to prepare a series of siderophores. At 50 μg/mL, the compounds PP-3 and PPDM-2 achieved mean inhibition rates (MIRs) of 64.0% and 67.2%, respectively, against 11 plant pathogens, comparable to tridemorph (64.6%). However, the two compounds exhibited distinct activity spectra. PP-3 and PPDM-2 showed strong activity against two oomycete pathogens (Pythium aphanidermatum and Phytophthora capsici) and Fusarium gramine

High-Throughput Identification of PPARγ-Active Chemicals in Riverine Drinking Water Sources: Discover a Novel DEHP Biodegradation Product.

While exposure to PPAR active compounds can lead to various adverse health effects, studies on the environmental contaminants and their transformation products with PPAR activity remain limited. This study employed PPARγ protein affinity selection-mass spectrometry (AS-MS) to comprehensively identify and prioritize PPARγ-active contaminants in the Yangtze and Yellow Rivers, key drinking water sources in China. From wastewater treatment plant effluents, river, and source water samples, 61 PPARγ ligands were identified, including 18 antagonists not reported previously. Notably, a novel biodegrad

Energetic Controls on Biodegradation of Organic Matter from Diverse Environmental Sources.

Organic matter (OM) decomposition is a central process in the global carbon cycle, regulating whether OM accumulates as a long-term sink or is mineralized into greenhouse gases. Although multiple explanations have been proposed to what controls OM degradation, the energetic and thermodynamic properties of OM have received less attention, particularly in cross-system studies. We incubated a wide range of OM from soils, sediments, and plant material, simultaneously measuring CO2 production and O2 consumption to quantify decomposition rates and the respiratory quotient (RQ, i.e., CO2/O2 molar rat

Elucidation of the Hydroxylation Sequence in 1-DNJ Biosynthesis through Stable Isotope Tracing and Enzymatic Characterization.

1-Deoxynojirimycin (DNJ) is the primary bioactive polyhydroxy alkaloid responsible for the antidiabetic properties of mulberry leaves. While the upstream biosynthesis is characterized, the downstream hydroxylation logic and transient intermediates remain elusive. Here, we combined stable isotope tracing with in vivo and in vitro enzymatic assays to delineate this hydroxylation sequence. Stable isotope feeding experiments identified piperidine as a key precursor and pinpointed C-7 as a critical secondary hydroxylation site. We demonstrated that (2R,4R)-2-methylpiperidine-4-ol (2) and (2S,4R)-2-

Menaquinones in bacteria - occurrence, function and biosynthesis.

Menaquinones (MKs), known mainly as respiratory quinones in bacteria, are found in the majority of bacterial species and show great variety as regards to the structure, i.e. the length of the side-chain, its saturation, ring methylation and other modifications. MK function in bacteria is attributed mainly to its function in anaerobic respiration, but the number of potential electron donors and acceptors is very numerous. However, their function is far beyond contribution in respiration. For example, they take part in bacterial photosynthesis, mediate extracellular electron transport or partici

Latent Class Trajectory Modeling Identifies Cerebrospinal Fluid Protein Signatures Associated with Motor Progression in Parkinson's Disease: Evidence from the Parkinson's Progression Markers Initiative Cohort.

BACKGROUND: Motor progression in Parkinson's disease (PD) is highly heterogeneous, limiting prognosis and the efficiency of clinical trials that require accurate risk stratification. Traditional modeling approaches assume a single average trajectory and fail to capture meaningful divergence, whereas the biological determinants of trajectory-defined subtypes remain unclear. Multivariate proteomic biomarkers may improve early identification of rapid progressors. OBJECTIVES: We aimed to (1) delineate distinct motor progression subtypes in early PD using longitudinal Movement Disorder Society-Uni

[Construction and application of a universal detection system for crop variety identification based on universal tailed-sequences].

Universal tailed-sequences (UTS) are the tail sequences of universal primers for amplification. They can achieve fluorescence detection by combining common primers with fluorescent universal primers and have the potential to reduce the cost of primer synthesis. However, a universal identification scheme for multiple crops based on the UTS system has not been established. There is a lack of verification for the system has not yet been practically applied in standardized variety identification scenarios. In addition, the data result docking schemes with conventional fluorescence primer systems r

Genomic differentiation and conservation relevance of isolated marginal Primula farinosa populations in Central-Eastern Europe.

INTRODUCTION: Quaternary climatic oscillations produced heterogeneous range dynamics in European cold-adapted plants. Primula farinosa L. has been discussed in the context of the interglacial expansion model for moisture-demanding cold-adapted species. However, it remains unclear whether isolated low-elevation populations in Central European calcareous fens and wet meadows represent long-term relicts or recently established populations. METHODS: We used RADseq-derived SNPs from four sampling localities in the Balaton Region of Hungary and one low-elevation comparative locality at Lipce in the

Long-term peanut cultivation alters soil organic C stocks and aggregate stability: insights from coupled iron oxide, microbial, and lignin phenol analyses.

INTRODUCTION: The microbial and mineral C pumps play crucial roles in soil organic C (SOC) stabilization via complicated organo-mineral interactions. Yet, how both of them regulate SOC dynamics during peanut cultivation remains elusive. METHODS: The surface soils from five peanut fields of varying ages (0-, 2-, 5-, 10-, and 20-yr.) were selected to elucidate the temporal variations in SOC dynamics and its potential mineral-microbial synergistic regulation patterns, based on PLFAs, eco-enzymatic activity, amino sugars, lignin phenols and Fe oxides analysis, etc. RESULTS: With increasing peanu
