Creating the first synthetic plastid genome in plants.

Over the last decade, there has been a shift in biotechnology from genome reading (sequencing) to genome writing (engineering). While significant progress has been made in prokaryotes and yeast, genome writing remains in its infancy in plants. Unlike their complex nuclear genomes, the compact and biotechnologically attractive plastid genome represents an ideal target to launch plant genome writing at scale.

Genome-wide characterization and drought-responsive expression profiling of plasma membrane intrinsic proteins (PIPs) in Caragana korshinskii.

Plasma membrane intrinsic proteins (PIPs) are an important subfamily of aquaporins that function as membrane channels that facilitate water transport across the plasma membrane in plants. Although the molecular mechanisms of PIPs have been extensively studied in model species, their characteristics and functions remain poorly characterized in Caragana korshinskii. Here, 13 CkPIP genes were identified through genome-wide analysis. CkPIPs were clustered into PIP1 (seven members) and PIP2 (six members) subfamilies. They exhibited conserved physicochemical properties, contained cis-acting elements

Visualization and quantitative analysis of endosperm cavities in maize kernels via X-ray micro-computed tomography.

BACKGROUND: Endosperm cavities within maize kernels influence quality traits such as kernel plumpness and hardness, serving as a key phenotypic indicator for assessing maize yield and quality. Research on endosperm cavities remains relatively scarce due to the small size of maize kernels and limitations in technical approaches. This study employed X-ray micro-computed tomography (μCT) three-dimensional reconstruction technology to extract morphological parameters and spatial configurations of endosperm cavities in multiple maize varieties, enabling visualisation and quantification of endosperm

Chromosome-level phylogenomics and genome evolution of the grape family.

Grapevines (Vitis) are pivotal fruit crops for human nutrition and civilization. The evolutionary history of grapevines in the grape family (Vitaceae) remains mysterious, requiring family-wide and whole-genome phylogenomic insights. Here we conduct evolutionary genomic analyses of Vitaceae by generating chromosome-level genome assemblies, placing Ampelopsis closer to Parthenocissus and Vitis. Karyotype evolution analysis delineates major chromosome rearrangements, highlighting overall conservation within Vitis but extensive reorganization for Tetrastigma and Cissus. Pan-3D genome analysis reve

A chromosome-level genome assembly of an elite indica breeding line resolves the genomic architecture of drought-yield QTLs qDTY2.2 and qDTY4.1 in rice.

Drought-yield quantitative trait loci (QTLs) qDTY2.2 and qDTY4.1 improve grain yield under reproductive-stage drought in rice (Oryza sativa L.), yet their genomic architecture remains poorly resolved in elite indica breeding backgrounds. We generated a chromosome-level assembly of IR87707-446-B-B-B (IR64-qDTY2.2 + qDTY4.1), which carries both QTLs in the IR64 background, and delineated donor-derived introgressions using informative parental single-nucleotide polymorphisms (SNPs). The 395.5-Mb assembly comprised 12 chromosomes, showed 99.2% BUSCO completeness, and contained 40,189 predicted pro

Explainable fine-grained feature enhancement framework for strawberry diagnostic decision making.

As the number of smart agricultural automation systems increases, accurate disease diagnosis is essential to maximize production efficiency. However, early-stage symptoms of strawberry diseases (e.g., Tipburn, Phytophthora blight, and Fusarium wilt) often manifest as low-contrast micro-symptoms that are visually similar to the inherent complex texture of strawberry leaves, leading to high technical difficulties in achieving an accurate diagnosis. Furthermore, providing an explanation for the diagnosis remains challenging, which hinders trust in field applications. We present EN-FDEA, a deep le

Contrasting physiological-biochemical responses and elemental homeostasis in Pinus sylvestris families exposed to As, Cd and Pb.

Contamination with As, Cd and Pb can limit tree establishment on degraded substrates, but family-level variation in Scots pine responses remains poorly resolved. This study evaluated three open-pollinated maternal half-sib families of Pinus sylvestris L. under controlled hydroponic exposure to As, Cd and Pb. Growth, photosynthetic pigments, soluble sugars, phenolic and flavonoid contents, malondialdehyde, antioxidant enzyme activities, elemental composition and metal(loid) accumulation indices were analysed, with PCA and Random Forest used as exploratory integrative tools. The Membrane Continuum Governing Cadmium Uptake, Partitioning, and Detoxification in Plants.

Cadmium (Cd) contamination threatens agro-ecological safety and food security; thus, elucidating mechanisms of Cd uptake and accumulation in plants is key to breeding low-Cd crops. Within the defensive network plants deploy against Cd stress, the plasma membrane and endomembrane system act as physical barriers to metal influx and as the core regulatory hub governing signal perception, transmembrane transport, and vacuolar compartmentalization for detoxification. This review examines the physiological and molecular mechanisms by which the cell membrane system influences Cd transport and accumul

Ligand-Driven Secondary Speciation of Silver Sulfide Nanoparticles Governs Fertilizer Nitrogen Fate in Paddy Soils.

Although silver sulfide nanoparticles (Ag2S-NPs) are presumed stable, their interactions with soil ligands remain poorly understood. Combining 15N tracing, synchrotron-based spectroscopy, and microbial profiling, we investigated how chloride (Cl-) and natural organic matter (NOM) regulate Ag speciation and nitrogen (N) cycling in paddy soils. We found that Cl- promoted the transformation of Ag2S-NPs toward labile Ag-Cl species, increasing Ag bioavailability by approximately 3-fold. This Ag mobilization was associated with enhanced denitrification potential, a greater unaccounted 15N fraction (

Deciphering the Enzyme-Mediated Oxidation of Dehydroabietic Acid and Other Diterpenoid Resin Acids.

Cytochrome P450 enzymes are versatile monooxygenase biocatalysts. Members of the bacterial CYP226 family are hypothesized to catalyze the oxidation of diterpenoid resin acids as the first step in their catabolism. Two CYP226 enzymes (CYP226A3 and CYP226A31) from the thermophilic bacterium Zestomonas thermotolerans were characterized. Both could bind the diterpenoids in gum rosin. Oxidation metabolites were formed using peroxide-driven reactions with the wild-type enzymes and variants with modifications at the dioxygen-binding groove. The CYP226A31 enzyme was used to enantioselectively hydroxyl

Phenyl-O-β-d-ribofuranoside Derivatives from the Plant Endophytic Fungus Neofusicoccum sp. LZUC-S1 and Their Herbicidal Activity.

Seven novel phenyl-O-β-d-ribofuranoside derivatives (1-7), along with four known naphthalenones (8-11), were isolated from the endophytic fungus Neofusicoccum sp. LZUC-S1 using a bioactivity-guided isolation. Their chemical structures were elucidated through an integrated analysis of IR, NMR, and HRESIMS data, and verified by crystallography and degradative experiments. Compounds 1-7 represent the first reported instance of phenyl ribosides. The phytotoxic potency of the purified metabolites was assessed against seedlings of Chloris virgata and Portulaca oleracea. Molecular insights into Acinetobacter calcoaceticus JK40-mediated zinc availability and transporter gene modulation in rice.

Zinc (Zn) deficiency is a critical edaphic constraint that limits the productivity of rice (Oryza sativa L.). This study examined the mechanistic effectiveness of Acinetobacter calcoaceticus JK40 (Ac-NAU-JK40), a zinc-solubilizing bacterium (ZSB) isolated from the rice rhizosphere. Notably, JK40 was selected for this study due to its dual ability to solubilize both zinc (Zn) and phosphate (P), a trait believed to prevent P-induced Zn precipitation in the rhizosphere. In hydroponic assays, co-inoculation of Ac-NAU-JK40 with insoluble ZnO significantly improved morphological traits (shoot length

Antagonistic activity and biocontrol potential of Pseudomonas sp. T19G isolated from the tomato rhizosphere against Ralstonia solanacearum.

Tomato bacterial wilt, caused by the soilborne bacterium Ralstonia solanacearum, remains difficult to manage, creating a need for biologically derived control agents. Here, we isolated Pseudomonas sp. T19G from the rhizosphere soil of healthy tomato plants and evaluated its antagonistic activity, protective effects, genomic basis, and influence on rhizosphere function. T19G fermentation broth inhibited R. solanacearum, and sterile cell-free supernatant caused a dose-dependent decline in viable pathogen populations. T19G significantly reduced viable root-associated pathogen populations and path

[Development of a rapid detection system for tomato spotted wilt orthotospovirus based on multienzyme isothermal rapid amplification (MIRA)].

Tomato spotted wilt virus (TSWV) is characterized by rapid transmission and a broad host range, creating an urgent need for detection methods applicable on-site at ports, in greenhouses, and in open fields. This study aims to develop a sensitive, specific, rapid, and field-applicable TSWV detection technique to provide technical support for the early diagnosis and precise monitoring of this virus. It targeted the conserved region of the TSWV L gene and designed multiple MIRA primers and probes. Through orthogonal-reciprocal cross screening, the optimal primer-probe combination was identified,
