Functional vomiting disorders in infancy: innocent vomiting, nervous vomiting, and infant rumination syndrome.

Pediatric gastroenterologists have tended to view gastroesophageal reflux (GER) as a disease in and of itself--a disease that can be diagnosed "objectively" with use of numerical data from esophageal pH monitoring and cured with pharmacologic or surgical treatment. What is often forgotten is that the data derived from esophageal pH monitoring and other techniques may identify the presence of abnormal GER but tell nothing about its pathogenesis. The usual approach to infants who feed poorly, vomit, or fail to gain weight is to identify the presence of abnormal GER, rule out underlying organic c

Oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA inhibits D1 dopamine receptor-mediated behaviors in normal mice and in mice lesioned with 6-hydroxydopamine.

The effects of intracerebroventricular (i.c.v.) administration of a 20-mer phosphorothioate oligodeoxynucleotide antisense to the D1 dopamine receptor mRNA (D1 antisense) on D1 dopamine receptor-mediated behaviors were studied in normal mice and in mice with unilateral 6-hydroxydopamine-induced lesions of corpus striatum. Treating mice with D1 antisense inhibited grooming behavior induced by the D1 dopamine receptor agonist SKF 38393 [1-phenyl-2,3,4,5-tetrahydro-1H-3 benzazepine-7,8-diol hydrochloride], the reduction in grooming being related to the amount and length of time D1 antisense was g

Effect of NO synthase inhibition on behavioral changes induced by a single administration of methamphetamine.

The present study examined the effects of nitric oxide synthase inhibitor, N omega-nitro-L-arginine methyl ester (L-NAME; 10, 30, 60 mg/kg, i.p.) on behavioral changes induced by a single administration of methamphetamine (MA) (3.22 and 0.805 mg free base/kg, s.c.). MA increased locomotion-stereotypy rating scores and motor activity counts measured by an infrared sensor. L-NAME administered prior to MA significantly decreased the level of locomotion stereotypy rating and motor activity induced by MA. An inactive optical isomer, N omega-nitro-D-arginine methyl ester (D-NAME) had no effects on M

Dopamine receptor antagonist properties of S 14506, 8-OH-DPAT, raclopride and clozapine in rodents.

S 14506 (1-[-(4-fluorobenzoylamino)ethyl]-4-(7-methoxynaphthyl)piper azine hydrochloride), 8-OH-DPAT ((+/-)-8-hydroxydipropylaminotetralin hydrobromide), clozapine and raclopride were compared in some behavioural models able to characterize dopamine antagonist properties. In mice treated with apomorphine (0.75 mg/kg, s.c.), stereotyped climbing and sniffing were dose dependently antagonized by S 14506, by clozapine and by raclopride, but were virtually not modified by 8-OH-DPAT. Stereotyped climbing and sniffing induced by (+)-amphetamine (1.25 mg/kg, s.c.) in mice treated with L-DOPA (L-3,4-d

Early postnatal antidepressant treatment and juvenile stereotyped behaviour in rats.

The effect of antidepressants on stereotyped behaviour in rats during development was examined. Rats were injected s.c. once a day from the 6th to the 22th day after birth with 0.9% sodium chloride, with desipramine 5 mg/kg body weight, with nomifensine 10 mg/kg, or with zimelidine 25 mg/kg. Stereotyped behaviour was assessed when the rats were 1 month and 2 months of age. Desipramine intensified and prolonged apomorphine 0.5 mg/kg induced stereotyped behaviour at 1 month but not at 2 months of age. Nomifensine and zimelidine did not affect stereotyped behaviour at either 1 month or 2 months o

Unilateral deficits induced in rats by MPP+ are markedly reduced by an N-terminal peptide fragment of dopamine-releasing protein.

Dopamine-releasing protein (DARP) is a novel factor involved in the function and development of catecholaminergic systems. To test whether a peptide synthesized from the N-terminus of DARP (DARP-36aa) ameliorates deficits in nigrostriatal dopamine, rats were unilaterally lesioned with MPP+, 1 day before administration of DARP-36aa began. Striatal delivery of 1 microgram DARPP-36aa daily for 10 days elevated striatal dopamine (P < 0.01) and reduced amphetamine-induced rotations (P < 0.05) relative to controls, both indicating the protection or restoration of dopaminergic function.

Use of clomipramine to treat ritualistic stereotypic motor behavior in three dogs.

Atypical, stereotypic motor and locomotor behavior was observed in 3 dogs. In 1 dog, the behavior included circling, vocalization, and snapping; in another, circling associated with pica and aerophagia; and in a third, gradually intensifying locomotor behavior stimulated by bright light. This behavior interfered with normal interaction between their owners and the dogs, and has been associated with obsessive-compulsive disorders. Treatment with clomipramine, along with other psychotropic medications and behavioral modification, suppressed the inappropriate behavior in all dogs; in 2 of the 3 d

MK-801 does not prevent acute stimulatory effects of amphetamine or cocaine on locomotor activity or extracellular dopamine levels in rat nucleus accumbens.

Recent work has shown that the development of behavioral sensitization to cocaine, amphetamine, and morphine is prevented by coadministration of N-methyl-D-aspartate (NMDA) antagonists such as MK-801. This suggests that NMDA receptors mediate long-term changes in neuronal responsiveness essential for the development of behavioral sensitization, similar to their role in other forms of neuronal plasticity. However, other studies, suggesting that NMDA receptor antagonists interfere with acute behavioral effects of psychomotor stimulants, call this interpretation into question and suggest that the

NMDA receptor blockade reverses motor response alterations induced by levodopa.

Motor fluctuations that ultimately complicate the response of most parkinsonian patients to levodopa therapy might represent a form of behavioral or neuronal plasticity. Since various forms of neuronal plasticity appear to be mediated by glutamate transmission through the N-methyl-D-aspartate (NMDA) receptor, the effect of NMDA receptor blockade on the development of alterations in the motor response to chronic levodopa was evaluated in hemiparkinsonian rats. Repeated levodopa administration decreased rotational behavior induced by a D1 dopamine receptor agonist, increased D2 agonist-induced r

Lazaroids improve the survival of grafted rat embryonic dopamine neurons.

In rodent models of Parkinson disease in which transplants of dissociated rodent and human embryonic mesencephalic tissue, rich in dopamine neurons, have been studied, only 5-20% of the dopamine neurons survive the implantation procedure. We have investigated the effects of inhibiting free radical generation with two lazaroids, U-74389G and U-83836E, on the survival of embryonic rat dopamine neurons. U-74389G is a 21-aminosteroid, and U-83836E combines the piperazinyl pyrimidine portion of 21-aminosteroids with the antioxidant ring of alpha-tocopherol. In an initial study, we found that the la

Further localization of cardiovascular and behavioral actions of substance P in the rat brain.

Cardiovascular and behavioral actions of substance P (SP) were examined after microinjection into the medial preoptic area (MPO), anterior hypothalamic area (AH), and ventral tegmental area (VTA) in conscious unrestrained rats. SP elicited marked increases in mean arterial pressure and heart rate as well as stereotyped behaviors of excessive grooming and exploring when injected into the MPO or AH. In the MPO, the latencies to the cardiovascular responses were observed after SP injection into the VTA. These results, together with our previous results, suggest that SP acts as transmitter or modu

Direct intracerebral gene transfer of an adenoviral vector expressing tyrosine hydroxylase in a rat model of Parkinson's disease.

Direct intracerebral gene transfer to neural cells has been demonstrated with recombinant adenovirus encoding beta-galactosidase. To explore the potential of recombinant adenovirus for the therapy of neurological disease we constructed a recombinant adenovirus encoding tyrosine hydroxylase and optimized intracerebral injection to express the gene in the striatum of unilaterally denervated rats. These animals have dopamine depletion in their lesioned striatum, causing a rotation asymmetry induced by apomorphine. One and two weeks after intracerebral injection this sensorimotor asymmetry was dec

Possible genetic factors underlying the pathophysiology of tardive dyskinesia.

Rates of spontaneous and drug-induced repetitive jaw movements (RJM) in rats vary widely. Low and high RJM responders were isolated and genetically selected. At each generation mean RJM responses (spontaneous or SKF 38393-induced) of the two types of rats were found to differ significantly, whereas neither apomorphine-induced stereotypic responses nor D1 and D2 receptor numbers and affinities differed. A significant increase in cAMP production was evident in SKF 38393-stimulated striatal homogenates of high RJM responders as compared with low responders. Animals subjected to 8-months exposure

Chronic treatment with a classical neuroleptic alters excitatory amino acid and GABAergic neurotransmission in specific regions of the rat brain.

The purpose of the following experiments was to describe some of the neurochemical changes that occur in the basal ganglia of rats exposed chronically to a classical neuroleptic, fluphenazine, and to relate these changes to extrapyramidal motor dysfunction. For these studies a combination of behavioural, receptor autoradiographic and in situ hybridization methods were employed. Preliminary pharmacological studies on GABA receptors showed that incubation in Tris-acetate rather than Tris-citrate buffer increased the number of binding sites labelled by [3H]muscimol by over 120% without affecting

Spatial planning deficits in limb apraxia.

Geschwind (1975) proposed a disconnection model in which an apraxic subject is unable to carry out movements to command because the left hemisphere that comprehended the verbal command is disconnected from the right premotor and motor areas which controls the left hand. An alternate model, however, proposes that apraxia results from destruction of spatiotemporal representations of learned movement stored in the left hemisphere (Heilman, 1979). The disconnection hypothesis would predict that apraxic subjects should be able to correctly imitate gestures and correctly use actual tools since these

Effects of Tityus serrulatus crude venom on the GABAergic and dopaminergic systems of the rat brain.

This study was designed to investigate the effect of T. serrulatus scorpion venom on dopamine (DA) and gamma amino butyric acid (GABA) concentrations in different regions of the brain. The ratio of homovanillic acid (HVA) to DA, and the glutamic acid decarboxylase (GAD) activity were determined following intravenous or intracerebral venom injections. The increase in the HVA/DA ratio in the striatum after i.v. or intrastriatal injection could indicate an increase in DA turnover. One hour after i.v. injection of the venom GAD activity was shown to be decreased in the striatum and hypothalamus. A

Chronic neonatal blockade of N-methyl-D-aspartate receptor by CGP 39551 increases dopaminergic function in adult rat.

Following chronic neonatal treatment with the competitive N-methyl-D-aspartate antagonist CGP 39551, adult rats showed increased behavioral responses to the D2 dopamine receptor stimulation. In nucleus accumbens and in n. striatum of similarly treated rats increases in D2 dopamine receptor number were observed. CGP 39551 was administered daily to neonatal rats with increasing doses from postnatal day 1 to 22. At postnatal days 70-82, the rats were observed for hyperactivity induced by the selective D2 dopamine receptor agonist LY 171555, the grooming behavior elicited by the specific D1 dopami

Exposure assessment of biomechanical stress in repetitive manual work using frequency-weighted filters.

A quantitative exposure assessment strategy for physical stress associated with repetitive manual tasks is proposed using continuous biomechanical data measured directly from electrogoniometers or force sensors. This paper describes an efficient method for reducing large quantities of biomechanical data into a quantifiable metric that accounts for recognized musculoskeletal exposure factors, including repetitiveness, postural or forceful exertion stress, and duration. A frequency domain approach is used for averaging elemental data recorded for repetitive cycles. Parameters for frequency-weigh

Akathisia and pseudoakathisia: clinical observations and accelerometric recordings.

BACKGROUND: Akathisia is a common side effect of neuroleptic treatment that is diagnosed on the basis of clinical observations. Its definition is regarded as complex and controversial. Thus, for example, while some authorities see akathisia as strictly an abnormal subjective state, others require the inclusion of specific repetitive movements. The present study suggests that the addition of objective accelerometric recordings to standardized clinical observations may increase the validity of this diagnosis. METHOD: Sixteen chronic schizophrenic patients, treated with various neuroleptic medic
