Effects of loss of CO2-Responsive CCT Protein interaction with 14-3-3 proteins and its ectopic expression on starch synthesis in rice.

CO2-Responsive CCT Protein (CRCT) is a transcription factor that positively regulates the expression of starch synthesis-related genes in the vegetative organs of rice. CRCT interacts with 14-3-3 proteins. Yeast two-hybrid analysis showed that the CRCT with the S217A mutation was unable to interact with CRCT. This mutated CRCT was introduced into the CRCT knockout rice. The transcript of CRCT-S217A was detected significantly by qRT-PCR, but not by immunoblotting. Nevertheless, starch accumulation and the expression of starch synthesis-related genes in the leaf sheath of the transformants were

FairFlow: A transparency-first framework for verifiable and reproducible bioinformatics.

Bioinformatics analyses depend on external software that itself relies on further programs, or dependencies, each with its own release cycle. Because tools are updated to fix bugs and add features, identical code can produce different results depending on which versions are installed, and incomplete documentation of dependencies and versions therefore undermines reproducibility. Containerization packages software with all its dependencies, but demands technical expertise and lacks a standard operating procedure. Existing workflow managers and container tools have improved reproducibility, yet

Investigating the outcomes of pregnancies with atypical results following SNP-based cfDNA screening: a population-based cohort study.

BACKGROUND: Cell-free DNA (cfDNA) screening has transformed prenatal screening by offering superior performance for detecting common aneuploidies, though it occasionally yields nonreportable or uninterpretable results. In 2020, a SNP-based cfDNA platform introduced a new category of nonreportable results termed "atypical," indicating additional or unexpected findings during DNA analysis. The clinical significance of these atypical results remains unclear, leaving clinicians without evidence-based guidance for counseling of pregnant individuals in the context of such results. OBJECTIVES: This

Enrolment criteria for surgical plication in pediatric diaphragmatic eventration: a systematic review of the literature.

PURPOSE: Diaphragmatic eventration (DE) is an uncommon but clinically significant condition in children. Although diaphragmatic plication is widely performed in symptomatic patients, standardized criteria guiding surgical indication remain poorly defined. This review aimed to identify the clinical-radiological parameters most consistently used to select pediatric surgical candidates. METHODS: We performed a literature search for English-language studies (2000-2025) reporting pediatric DE cases. Studies limited to radiology, genetics, or unrelated diaphragmatic disorders were excluded. Extract

Establishing meaningful score differences for the observer-reported communication ability (ORCA) measure using the scale judgment approach with caregivers of individuals with Angelman syndrome.

PURPOSE: The Observer-Reported Communication Ability (ORCA) measure assesses communication ability in individuals living with Angelman syndrome (AS). As gene therapies progress through clinical trials, we need to identify what within-person changes in communication are meaningful to families. This study used the Scale Judgment approach to estimate meaningful score differences (MSDs) on the ORCA measure for AS. METHODS: Caregivers (n = 156) of individuals living with AS completed Scale Judgment tasks showing vignette pairs depicting an individual's communication ability before and after a hypo

First-Aid Practices and Knowledge Regarding Snake and Scorpion Bites Among Patients Attending Onandjokwe State Hospital, Namibia: A Hospital-Based Cross-Sectional Study.

BACKGROUND: Snake and scorpion bites pose a significant public health burden, with an estimated global annual morbidity rate of 5.4 million and 1.2 million, respectively. Various first-aid methods are commonly practiced in communities, aiming to improve the chances of survival after bites. This study evaluated community practices for first aid on snake and scorpion bites and examined the alignment of these practices with WHO recommendations. This will determine the need for targeted educational interventions to promote evidence-based and safe first-aid practices. METHODS: A cross-sectional st

Child Trauma and Family Adversity Predict Treatment Completion Among High-Risk Youth in Intensive Home-Based Treatment: A Latent Class Analysis of the Intensive In-Home Child & Adolescent Psychiatric Service (IICAPS).

Over the past three decades, rigorous empirical research has highlighted both cumulative and non-additive effects of childhood trauma, which are intricately intertwined with the broader developmental and psychosocial context. Latent class analysis has proven useful in identifying at-risk groups, thereby informing the design of targeted prevention and early-intervention efforts. Extending prior research to highly complex, high-risk children and families receiving intensive home-based treatment (IHBT), this study analyzed archival data from 10,301 Connecticut families enrolled in the Intensive I

The relationship between childhood trauma and adult neuroticism: A systematic review and meta-analysis.

Childhood trauma has been consistently associated with elevated levels of neuroticism in adulthood, a transdiagnostic trait marked by emotional instability, heightened negative affect, and stress sensitivity. This systematic review and meta-analysis aimed to synthesise evidence examining the association between childhood trauma and adult neuroticism, both overall and by specific trauma subtypes. A comprehensive search of four electronic databases identified 136 eligible studies, encompassing a total of 526,371 individuals. Using a random-effects meta-analysis, results revealed a significant po

Association between cold weather and emergency department utilization among patients experiencing houselessness in a northern Canadian city.

INTRODUCTION: Lack of secure shelter is a contributing factor for healthcare utilization. Weather extremes, specifically extreme cold, is common in Canada. Because of the safety-net function of emergency departments (EDs), their use as shelters or warming centers is common in cold weather. Timely diversion resources may help mitigate ED crowding. METHODS: We conducted a cohort study using linked administrative data between April 2018 and March 2024 of all adult (> 17 years old) patients presenting to any EDs in the greater Edmonton area with an International Classification of Diseases, 10th r

Safety Assessment of Glucagon-Like Peptide 1 Receptor Agonists Based on the FAERS Database: Focus on Tumorigenic Risk in Subpopulations.

GLP-1 receptor agonists (GLP-1RAs) are extensively used in treating Type 2 diabetes due to their therapeutic benefits. However, clinical reports have highlighted adverse events, particularly the underexplored association with tumor development. This study investigates GLP-1RA-related adverse events, focusing on tumorigenic risks, using data from the FDA Adverse Event Reporting System (FAERS). FAERS data from Q1 2005 to Q4 2023 were analyzed using the proportional reporting ratio (PRR) method to identify GLP-1RA-associated adverse events. Detailed analyses examined event distribution across gen

Coasting effect of n-hexane-induced neuropathy evidenced by electroneuromyography and clinical symptom scales: a 12-month follow-up study.

To the best of our knowledge, no study so far has investigated the coasting effect of neuropathy caused by occupational exposure to n-hexane through more detailed clinical evaluations than nerve conduction measurements, such that would also include pain assessment and functional status. The aim of our study was therefore to see if our measurements, which include all three elements, would support the coasting effect and reveal associations between electrophysiological measurements and clinical symptom assessments over a 12-month follow-up. Our study included eighteen patients working in the sam

Evidence-based behaviour change techniques to aid smoking cessation for pregnant women with mental illness: a systematic review.

BACKGROUND: Smoking during pregnancy is the leading preventable cause of maternal and infant morbidity and mortality in high income countries, disproportionately affecting women with mental illness. While behavioural smoking cessation interventions are effective for pregnant women and non-pregnant people with mental illness separately, evidence for specific smoking cessation interventions for pregnant women with mental illness is lacking. This review aimed to identify and map BCTs from effective general mental health smoking cessation trials to determine which may be helpful for pregnant women

Examining adverse effects in a large clinical trial of rTMS application as a treatment for Alzheimer's disease.

BACKGROUND: Repetitive transcranial magnetic stimulation (rTMS) has several advantages compared to other interventions for neurological and psychological disorders. However, various adverse effects have been reported in rTMS research, and little is known about who is most susceptible to rTMS adverse effects, or how they can be minimized. AIMS: We aimed to identify risk factors for adverse effects reported in a recent clinical trial examining rTMS as a treatment for Alzheimer's disease (AD). We hypothesized that higher stimulation intensity would be associated with experiencing unspecified pai
