Links between cannabinoid hyperemesis syndrome symptoms and drug use, mental health problems, antisocial behavior, and personality in a national survey of adults in the United States.

BACKGROUND: Cannabinoid hyperemesis syndrome (CHS) is characterized by episodes of severe nausea, vomiting, and abdominal pain among those with heavy cannabis use. We estimated differences between those reporting CHS symptoms and those who use cannabis daily and less frequently on drug use, psychiatric symptoms, other health aliments, antisocial behavior, and personality. METHODS: The National Firearms, Alcohol, Cannabis, and Suicide survey was administered to 7034 US adults (3427 male, 3607 female) in 2025. Survey items assessed substance use, common psychiatric symptoms, personality traits,

Dynamic view of an allosteric intermediate in a positively cooperative dimer.

UNLABELLED: Positive cooperativity in ligand binding is a hallmark of allosteric oligomers, yet how the first binding event enhances the second remains obscure, because the pivotal singly-bound intermediate (lig₁) is thermodynamically disfavored and rarely accumulates. Distinguishing concerted (MWC) from sequential (KNF) mechanisms turns on one question: when a ligand binds one protomer, does its empty partner change conformation? Here we resolve this for the allosteric homodimer chorismate mutase (CM) using mixed-labeled dimers (MLDs), in which a single maleimide crosslink stabilizes a hetero

Whole-exome sequencing in individuals with obsessive-compulsive disorder and chronic tic disorders identifies 36 large-effect risk genes.

Obsessive-compulsive disorder (OCD) and chronic tic disorders (CTDs) are highly heritable. Rare mutations confer large risks for OCD and CTDs but only four high-confidence (hc) genes have been identified. Here we analyzed whole-exome sequencing data from 3,964 individuals with OCD, CTDs or both, including 2,418 trios. We found an excess in cases of de novo and rare protein-damaging mutations and identified 36 hc genes (false discovery rate < 0.1), including four previously identified hc genes (CELSR3, CHD8, SCUBE1 and WWC1) and four genes that overlap with OCD genome-wide association study loc

Evoke (+) trials of semaglutide for early alzheimer's disease: Innovations, lessons, and implications.

The evoke and evoke+ trials for the treatment of early Alzheimer's disease (AD) with semaglutide were founded on the observation that semaglutide reduced the incidence of dementia in patients with type 2 diabetes. Non-clinical investigations support an effect of semaglutide on AD pathology. The evoke and evoke+ trials were large, well conducted, and innovative. These trials were negative, showing no clinical benefit of treatment with 14 milligrams of semaglutide for two years. Semaglutide produced a statistically significant and robust reduction in high sensitivity C-reactive protein consisten

Addressing evidence gaps in haemodialysis care: a critical review of clinical trials, technological innovations, and implementation priorities.

Kidney failure treated with maintenance haemodialysis (HD) carries high risks of premature mortality, hospitalisation and impaired quality of life, yet important evidence gaps persist across routine HD care. The Dialysis Research Innovation Network, part of the UK Kidney Research Consortium, convened the inaugural UK Haemodialysis Research Nexus in Birmingham on 11th May 2026. There were fifty-six delegates: 22 doctors, 13 nurses, six dietitians, one renal scientist, seven people in other roles (including one patient), and seven whose role was not recorded. The meeting reviewed recent trials,

Properdin deficiency or anti-properdin treatment ameliorates disease in the C3 gain-of-function mouse model of atypical haemolytic uraemic syndrome.

INTRODUCTION: We have previously shown that the C3D1115N mouse, engineered around a single point mutation in C3 associated with atypical haemolytic uraemic syndrome (aHUS) in man, fully recapitulates the clinical disease. In this study, we investigated the role of properdin in aHUS. METHODS: C3D1115N mice were crossed onto properdin-deficient mice or C3D1115N mice were treated with anti-properdin monoclonal antibody therapy and survival tracked. Kidney function, serum biomarkers and kidney pathology was carefully assessed at set end points. C3 and Fibrin deposition was assessed using immuno-f

Better Conversations With Parkinson's: Feasibility and Acceptability of a Novel Communication Partner Training Telehealth Intervention.

BACKGROUND: Communication Partner Training (CPT) has an established evidence base in aphasia, dementia and traumatic brain injury. It has been shown to improve the skills, attitudes and knowledge of the communication partner, and improve communicative participation and confidence of the person with communication difficulties. CPT therefore holds potential value for people living with Parkinson's and their everyday conversation partners who report significant impact of communication changes on their day-to-day lives. However, the CPT evidence base is less well established in this population. Pe

CCL2 as a microvascular "tone breaker": inflammatory signaling and the failure of kidney autoregulation in salt-sensitive hypertension.

Hypertension-associated kidney injury is strongly shaped by the ability of the kidney to buffer systemic pressure at the afferent arteriole. Feng et al. identify C-C motif chemokine ligand 2 as a direct regulator of kidney microvascular smooth muscle contractility in Dahl salt-sensitive rats. Loss of C-C motif chemokine ligand 2 preserves Notch3-Mylk (myosin light-chain kinase) signaling, maintains myogenic tone, protects autoregulation, and prevents kidney injury. These findings position the C-C motif chemokine ligand 2/C-C motif chemokine receptor 2 axis as a mechanistic bridge between infla

Self-rated health and mental health before and during the early phase of the COVID-19 pandemic in Germany: the population-based German National Cohort (NAKO) study.

BACKGROUND: The COVID-19 pandemic and accompanying social distancing measures might have caused adverse health consequences. We aimed to describe changes in participants' self-rated health and mental health (depression, anxiety, and stress), and investigate factors associated with them. METHODS: We collected data from the German National Cohort (NAKO). We first described changes in participants' self-rated health and mental health from the baseline examination (1 to 6 years earlier) to the early phase of the COVID-19 pandemic. We then applied the multinomial logistic regression model (self-ra

Patient blood management in general intensive care patients.

PURPOSE: Critically ill and high-risk perioperative patients requiring intensive care are often multimorbid and depend on rapid, highly specialized management. While most comorbidities are difficult to modify in the acute setting, anemia, particularly iron-defi ciency anemia, represents a potentially modifiable risk factor. Clinicians are also often confronted with complex alterations in hemostasis that require rapid assessment and targeted therapeutic interventions, including the optimal use of blood products. This narrative review summarizes the current evidence on Patient Blood Management s

Longitudinal evaluation of the psychometric stability of the somatic symptom scale-8 (SSS-8) in a large German general population sample.

The Somatic Symptom Scale (SSS-8) is a brief 8-item tool for screening common somatic symptoms. This study is the first to examine its temporal invariance and stability across three COVID-19 pandemic time points, while also analyzing changes in SSS-8 scores in the general population and across groups (sex, age, relationship status, poverty status), and testing temporal stability and measurement invariance. Longitudinal data from three pandemic time points in a large population-based sample were used. Temporal stability was assessed using Pearson correlations, and confirmatory factor analysis w

Simultaneous quantification of anti-Spike IgG subclasses against various SARS-CoV-2 variants using a multiplex serological assay.

Recent research indicates that the composition of serum anti-SARS-CoV-2 Spike IgG subclass antibodies changes upon repeated exposure to viral Spike. These antibody subclass dynamics were suggested to impact adaptive immune responses and protection against infection. To characterize the role of the IgG subclass composition in the immune response to SARS-CoV-2, assays that measure virus variant-specific anti-Spike IgG subclass concentrations are needed. Here, we describe an easy-to-use serological method for quantifying anti-Spike IgG subclass antibodies against various SARS-CoV-2 variants in mu
