Assessing Aerobic Capacity in Rat and Mouse Models: A Critical Narrative Review.

Aerobic capacity reflects pulmonary ventilation, oxygen transport, cardiovascular performance, skeletal-muscle perfusion, and mitochondrial oxygen utilization. In rodents, V̇O2max, V̇O2peak, running speed, time to exhaustion, critical speed, lactate-derived thresholds, and voluntary activity are often grouped as aerobic capacity despite representing distinct constructs. This review aims to critically examine the physiological foundations, experimental modalities, outcome measures, methodological determinants, and translational challenges of aerobic-capacity assessment in rats and mice. Introducing GABLE: Lightweight infrastructure for longitudinal experiments using Google Apps Script.

Longitudinal studies are essential for understanding how behaviors, skills, and psychological states evolve over time. While online platforms have made study development easier, researchers still face major logistical challenges when managing multi-session studies, especially when coordinating large numbers of participants, session timing, reminders, and compensation. In this paper, we introduce GABLE (Google Apps script-Based Longitudinal Experimentation), a lightweight infrastructure built using Google Apps Script. GABLE automates participant scheduling, e-mail communication, session trackin

The IFN-γ response cycle: decoupling tumor killing from evolutionary selection in cancer immunotherapy.

Interferon-γ (IFN-γ) confronts the field with an apparent contradiction that is better understood as a causal consequence: the cytokine most central to antitumor immunity is also a principal driver of immune escape, and recombinant IFN-γ has both helped and harmed patients. We argue that this duality is not paradoxical but expected, escape is, in large part, the predictable evolutionary outcome of effective IFN-γ-mediated pressure. The prevailing "two faces" framing, which separates antitumor and protumor effects by dose and duration, names this discrepancy without explaining what couples its

Baseline Performance and the 72-Hour Performance Window Following Plyometric Withdrawal in Collegiate Sprinters.

Plyometric studies emphasise pre-post change, but coaches need to know what remains after the block ends. This single-cohort study followed 26 collegiate sprinters through 12 weeks of Monday plyometric training and 4 weeks of withdrawal, without a parallel control group. During intervention, 30 m sprint time and arm-assisted countermovement jump (CMJ) height were measured at 0, 24, 48, 72, and 96 h; 0 and 72 h were retained during withdrawal. Mixed models showed large week effects for sprint, F(1, 1534) = 2272.01, partial η2 = 0.597, 95% CI [0.570, 0.622], and CMJ, F(1, 1534) = 2537.82, partia

The Biomechanical Signature of Elite Distance Running: A Matched Pairs Analysis Across Multiple Speeds.

This study compared the biomechanical profiles of elite and recreational distance runners across five measurement domains using wearable inertial sensors across treadmill speeds of 10-15 km/h. A total of 104 runners (52 elite; 52 recreational) were assessed using RunScribe sensors during an incremental treadmill protocol. Twenty-three variables spanning spatiotemporal parameters, stiffness and force, impact loading, foot kinematics and vertical dynamics were recorded at each speed. Elite runners were matched 1:1 with recreational runners by sex, age, height, and body mass. For each variable, a

Effectiveness of exercise training on hippocampal mitochondrial regulation in Alzheimer's disease: A mechanistic systematic review.

Alzheimer's disease (AD) is associated with hippocampal mitochondrial dysfunction, including impaired mitophagy, biogenesis, dynamics, bioenergetics, and redox homeostasis, which may contribute to amyloid-β (Aβ) accumulation, tau pathology, and cognitive decline. This systematic review examined whether exercise training is associated with changes in hippocampal mitochondrial regulation and cognitive function in transgenic AD rodent models. Following PRISMA 2020 guidelines, PubMed, EMBASE, and Web of Science were searched up to July 2, 2026. Twenty-one studies met inclusion criteria. Exercise m

Clinical similarity, economic divergence: cost-effectiveness and population health implications of dietary, pharmacological, and exercise interventions in older adults with type 2 diabetes: a secondary analysis of a randomized controlled trial.

BACKGROUND: Population ageing is increasing the burden of chronic disease and frailty, raising important questions regarding how health systems define and allocate value. While pharmacological treatments dominate clinical care, lifestyle-based interventions may offer broader benefits for healthy ageing. However, integrated clinical and economic evidence in older populations remains limited. METHODS: We conducted a secondary analysis of the CEV-65 randomized controlled trial (n = 100 older adults aged 65 years and older with type 2 diabetes), comparing a Mediterranean diet (VegMedD), pharmacol

Spatio-temporal machine learning for multi-horizon prediction of bluetongue outbreaks.

Reliable early warning of infectious disease outbreaks remains a major challenge for surveillance systems, particularly for vector-borne pathogens whose transmission depends on interactions among hosts, vectors, and climate-sensitive environmental conditions. Data-driven forecasting offers a promising approach for predicting outbreak risk using surveillance and environmental data. This study develops a logit-weighted ensemble (LWE), a machine-learning framework that predicts outbreak occurrence 1-6 months ahead at the administrative unit-month scale using routinely available outbreak notificat

Running in reverse: Tau redirects mitochondrial electron flow toward neurodegeneration.

In this issue of Neuron, Li et al.1 show that tau enters mitochondria and binds NDUFS3 to regulate reverse electron transport (RET) in a phosphorylation-dependent manner. RET promotes tau phosphorylation, forming a self-perpetuating loop that is exacerbated with stress and tauopathy.

Parietal Structural and Functional Differentiation Associated with Visual Tracking Across Motor Participation Profiles.

PURPOSE: This study aimed to examine whether structural and resting-state functional measures of parietal subregions differ across predefined motor participation groups with comparable physical activity levels, and whether these neural measures are associated with visual tracking performance. METHODS: We adopted an a priori motor-cognitive grouping framework and recruited athletes accordingly, using two representative sports as operational exemplars for each profile: (1) externally driven spatial updating and action planning (ESU; basketball and taekwondo), (2) body-environment calibration an

Third-Body Dynamics and Tribo-Oxidation Mechanism in Fretting Degradation of an Inconel 718/304 Stainless Steel O-Ring Seal.

Fretting wear at metal O-ring sealing interfaces involves the coupled processes of debris generation, entrapment, and oxidation. Existing accelerated life testing relies on similarity theory, yet it overlooks the dominant role of third-body behavior in governing wear regime transitions, so equivalence between test and service conditions lacks a physical basis. This study investigates reciprocating fretting wear of an Inconel 718/304 stainless steel pair and reinterprets wear evolution using third-body dynamics. Age-dependent effects of TRPA1 deficiency on pain and osteoarthritis pathology following joint injury in mice.

INTRODUCTION: Transient receptor potential ankyrin 1 (TRPA1) is a calcium-permeable cation channel expressed in multiple cell types, including nociceptive C-fibers innervating the knee joint, and is activated by mechanical, inflammatory, and oxidative stimuli. While TRPA1 inhibition reduces pain-related behavior in chemically-induced arthritis, its role in mechanically induced osteoarthritis (OA), and how this relationship changes with age, remains unclear. Here we tested the hypothesis that TRPA1 deficiency alters mechanical sensitivity and OA pathology after joint injury in an age-dependent

Protocol for simulating cell-contact dynamics of proliferating epithelia using the vertex model.

Vertex models simulate epithelial mechanics at single-cell resolution but represent cell division as a single in-plane split, agnostic to the differential apicobasal geometry of mitotic cells. Here, we present a protocol that simulates basal mitosis as two sequential changes: basal-cell diminution followed by daughter-cell reinsertion. Apical-side simulations can be performed with minimal code modification. We provide instructions for generating initial configurations, running simulations, and quantifying mechanical and topological rearrangements using command-line operations accessible to exp

Reproducible analytical pipelines for healthcare discrete‑event simulation: An open guide and worked examples.

Discrete-event simulation (DES) is a popular technique for exploring problems in healthcare. For these models to be reused and have lasting impact, they need to be reproducible, transparent and well structured. Reproducible analytical pipeline (RAP) approaches are structured robust workflows that ensure analyses can be reproduced. They have emerged as best practice, but modellers struggle to implement them due to gaps in accessible guidance, skills, and time. This paper presents an integrated set of resources designed to help modellers bridge this implementation gap: an open-access e-book and

Predicting the Risk of Unplanned Hospital Encounters in Hemodialysis Patients: A Comparison Between Clinicians and a Machine Learning Prediction Algorithm.

RATIONALE & OBJECTIVE: Dialysis patients have a higher risk of hospitalization than the age-matched general population. Unplanned hospital encounters (UHEs), including emergency department visits and hospitalizations, may be preventable but are difficult to anticipate. We aimed to stratify dialysis patients' risk of UHE, using a machine learning (ML)-based prediction model. STUDY DESIGN: We created an ML-based prediction model to stratify hemodialysis (HD) patients' risk of UHE in the subsequent 7 and 30 days. We compared the performance of this prediction model with that of clinicians. SETT
