Development of a real-time multiple cross displacement amplification assay for rapid and highly sensitive detection of Streptococcus pyogenes.

The aim of this study was to develop a method based on multiple cross displacement amplification (MCDA) and real-time fluorescence technique for rapid, highly sensitive and specific detection of Streptococcus pyogenes (Group A Streptococcus, GAS). A set of 10 primers targeting the speB gene of GAS was designed for the MCDA reaction. According to the principle of real-time MCDA detection, the core primer was further modified with a restriction endonuclease recognition sequence, a fluorophore, and a quencher. The optimal reaction temperature for the assay was determined based on the performance

Advanced serial analysis of the diaphragm surface EMG: insights into the effect of pressure support on the neuro-ventilatory response during the ICU stay.

BACKGROUND: Ventilatory support levels in ICU patients should be tailored to both optimal gas exchange and respiratory muscle loading, as over- and underassistance may cause diaphragm dysfunction. The diaphragm's capacity to overcome mechanical load and deliver ventilatory output is reflected by the patient's neural respiratory drive (NRD), tidal volume (TV) and respiratory rate (RR). Surface electromyography of the diaphragm (sEMGdi) offers a continuous, non-invasive measure of NRD. We investigated the effect of pressure support (PS) level on the coupling of diaphragm electrical activity (sEA

Multi-criteria decision analysis of gas enrichment risk and land degradation in goaf areas.

Gas enrichment in goaf areas significantly threatens mine safety and surface environmental stability, particularly under top-cutting and pressure-relief mining methods. This study proposes a comprehensive multi-criteria decision-making framework that configures the Analytic Hierarchy Process (AHP) and the Technique for Order Preference by Similarity to Ideal Solution (TOPSIS) to evaluate a coupled gas enrichment-land degradation risk in goaf areas under top-cutting and pressure-relief mining. Five main criteria, geological conditions, mining parameters, ventilation characteristics, gas emissio

The Criterion Validity of a Newly Developed Ballroom Aerobic Test (BAT) Protocol Against Objective Methods.

Although laboratory testing to assess aerobic capacity has been a 'gold standard' in sports science, its high costs and time-consuming protocols may not be feasible for monitoring and tracking progress in limited conditions. In dancesport athletes, several field-based aerobic tests have been proposed, but the majority of them have been developed for ballet or contemporary dancers at the individual level, while the data among dance couples engaging in standard dance styles is lacking. Therefore, the main purpose of this study was to validate a newly developed Ballroom Aerobic Test (BAT) protoco

An alternative method to differentiate pleural effusion after leakage of artificial enteral nutrition formula.

PURPOSE: This case report highlights a rare but fatal complication of artificial enteral nutrition due to feeding tube misplacement, leading to pleural effusion and respiratory failure. The primary objective was to determine whether enteral nutrition formula leakage contributed to the patient's death and to differentiate the pleural effusion from other possible etiologies, such as chylothorax. METHODS: A 59-year-old male patient with severe lung disease underwent left-sided partial lung resection and subsequently received enteral nutrition via a nasogastric feeding tube. After suspected feedi

Multi-ancestry genome-wide meta-analysis of 56,241 individuals identifies known and novel cross-population and ancestry-specific associations as novel risk loci for Alzheimer's disease.

BACKGROUND: Limited ancestral diversity has impaired our ability to detect risk variants more prevalent in ancestry groups of predominantly non-European ancestral background in genome-wide association studies (GWAS). We construct and analyze a multi-ancestry GWAS dataset in the Alzheimer's Disease Genetics Consortium (ADGC) to test for novel shared and population-specific late-onset Alzheimer's disease (LOAD) susceptibility loci and evaluate underlying genetic architecture in 37,382 non-Hispanic White (NHW), 6728 African American, 8899 Hispanic (HIS), and 3232 East Asian individuals, performin

Consensus meta-analysis of genome-wide association studies for Alzheimer's disease and related dementia.

To better characterize the genetic architecture underlying Alzheimer's disease (AD) and related dementia (ADRD), we performed a meta-analysis of European ancestry genome-wide association studies in 128,681 cases or proxy cases of ADRD and 849,833 (proxy) controls. We identified 91 genetic loci associated with ADRD risk, including 16 which are novel, and 56 which are specifically detected in clinically diagnosed AD cases. We also provide a list of 18 loci (15 novel) requiring further external validation. A polygenic score combining the effect of the ADRD loci except APOE was primarily associate

Validation of a direct injection-GC-MS method for the quantification of volatile alcohols in blood and vitreous humor.

Volatile compounds such as ethanol, methanol, isopropanol, and acetone are frequently encountered in forensic toxicology, particularly in postmortem investigations, poisoning cases, and incidents involving impaired driving. While headspace gas chromatography (HS-GC) remains the gold standard for their analysis, its limited availability in many laboratories highlights the need for more accessible alternatives. This study aimed to develop and validate a direct injection gas chromatography-mass spectrometry (DI-GC-MS) method for the simultaneous quantification of ethanol, methanol, isopropanol, a

Use of cold atmospheric plasma (CAP) for the control of gingival inflammation in a rabbit (Oryctolagus cuniculus) with progressive syndrome of acquired dental disease (PSADD): case report.

Progressive syndrome of acquired dental disease (PSADD) is common in domestic rabbits and is often associated with chronic gingivitis, which compromises quality of life. Conventional treatment involves prolonged use of antimicrobials and anti-inflammatories, which may cause resistance and adverse effects. Cold atmospheric plasma (CAP) has antimicrobial, anti-inflammatory and regenerative properties, suggesting its therapeutic potential. This pilot study evaluated the effects of CAP as a complementary treatment for gingivitis associated with PSADD in a domestic rabbit. Two applications of the p

Use of Hyperbaric Oxygen in Patients with Necrotizing Soft Tissue Infections: A Scandinavian Multicenter, Prospective, Observational Cohort.

INTRODUCTION: Hyperbaric oxygen (HBO2) treatment is regularly used as adjuvant treatment in patients with necrotizing soft tissue infections (NSTI). Several observational studies have suggested an association between hyperbaric oxygen (HBO2) treatment and improved survival in patients with necrotizing soft tissue infections (NSTIs); however, the evidence remains inconclusive. Most patients with NSTI are severely ill, having sepsis or septic shock, and requiring mechanical ventilation and inotropic support in an intensive care unit (ICU). Although recent data suggest that the beneficial effect

Innovative disposable in-tip cellulose paper (DICP) device for facile determination of pesticides in postmortem blood samples: A proof-of-concept study.

Accurately identifying and quantifying toxicants is crucial for medico-legal investigations in forensic toxicology; however, low analyte concentrations and the complex samples matrix make this work difficult. Therefore, a simplified sample preparation procedure is crucial to streamline the analysis to minimize sample handling errors, reduce cost and improve the overall efficiency of analysis of toxicants. To address these challenges, an innovative disposable in-tip cellulose paper (DICP) device has been developed for the extraction of three pesticides viz. Chlorpyrifos, Quinalphos and Carbofur

Age estimation on post-mortem CT based on pelvic bone mineral density measurement and the state of putrefaction: a multivariate method.

Age-at-death estimation is an important issue in forensic medicine and anthropology. Initially, methods relied on morphological criteria, but with the advancement of radiology, new techniques such as morphological studies on multi-slice computed tomography (CT) reconstructions have emerged. Recent studies have shown promising results by investigating the correlation between age and bone mineral density (BMD). However, there is currently a lack of data on post-mortem CTs (PMCT) involving decomposed bodies, and limited information exists regarding changes in Hounsfield Units measurement in a pos

Comparative transcriptomic analysis provides novel insights into mescaline biosynthesis by Lophophora williamsii.

Lophophora williamsii, known for mescaline synthesis, has raised legal and ethical considerations. However, L. williamsii specimens that do not contain mescaline have been recently identified, necessitating the development of techniques to differentiate between mescaline-positive and mescaline-negative groups. Genetic markers have been explored to differentiate these specimens, complementing gas chromatography/mass spectrometry analysis. Here, we used a single-molecule real-time polymerase chain reaction to generate a full-length L. williamsii transcriptome and elucidate the mechanisms underly
