Occurrence, source apportionment, and risk assessment of antibiotics in the Beiyun River system: a highly urbanized watershed in Beijing, China.

Antibiotic residues in receiving water bodies of densely populated urban areas have attracted widespread concern. In this study, 35 antibiotics, including tetracyclines, macrolides, sulfonamides, and quinolones, were identified in the Beiyun River system of Beijing, a megacity in China. The spatial distribution patterns and ecological risks related to the target antibiotics in different surface water types were investigated, while their main sources were quantitatively identified using principal component analysis-multiple linear regression (PCA-MLR). The results indicated that all 35 antibiot

Characterization of a Fungal Modular Polyketide Synthase System Reveals an Unusual Strategy for Polyketide Chain Initiation.

Polyketides constitute a major class of natural products with remarkable structural diversity and biological activities. While bacteria predominantly produce polyketides via modular type I polyketide synthases (PKSs), eukaryotic modular PKSs remain largely unexplored. Here, we report the functional characterization of a fungal modular PKS system from Lasiodiplodia theobromae through heterologous expression in Aspergillus oryzae, leading to the production of the antimalarial macrolide strasseriolide C (1). Intriguingly, biosynthesis of 1 involves C-methylation of the starter malonyl unit despit

Unsupervised machine learning for multi-system adverse event pattern detection in pharmacovigilance: A proof-of-concept analysis of fluoroquinolone reports in FAERS.

AIMS: Traditional pharmacovigilance detects single drug-event associations but cannot readily characterize multi-system adverse-event patterns such as fluoroquinolone-associated disability. We evaluated unsupervised machine learning for this purpose, using fluoroquinolones as an exemplar. METHODS: We analysed 40 127 adult fluoroquinolone primary-suspect reports from the FDA Adverse Event Reporting System (FAERS, 2004-2025). Events were encoded as binary MedDRA Preferred Term (PT) and System Organ Class matrices; seriousness and outcome were excluded from clustering and used only post hoc. Mul

In silico design of an epitope-based vaccine for Rocio virus using phage display and E. coli expression system.

The incidence of heart failure with concomitant cardiac fibrosis is very high in developed countries. Fibroblast activation in heart is causal to cardiac fibrosis as they convert to hypersynthetic cardiac myofibroblasts. There is no known treatment for cardiac fibrosis. Myofibroblasts contribute to the inappropriate remodeling of the myocardial interstitium, which leads to reduced cardiac function and ultimately heart failure. Elevated levels of autophagy have been linked to stress-induced ventricular remodeling and other cardiac diseases. Previously, we had shown that TGF-β1 treatment of huma

Activity of MMV Pandemic Response Box compounds against Mycobacterium ulcerans viability and mycolactone production.

Mycobacterium ulcerans causes Buruli ulcer (BU), a skin neglected tropical disease, largely through the expression of a potent cytotoxin known as mycolactone. As mycolactone is central for BU pathogenesis, it follows that targeting its production with drugs could be one means of controlling the infection. However, this has not yet been attempted due to the difficulty of applying typical mycolactone-detecting assays, such as mass spectrometry, to routine drug screening to identify suitable drugs for this purpose. In this repurposing study, we screened the Pandemic Response Box, an open-source c

Development and Optimization of Extraction Methodologies for the Determination of Antibiotics in Honey by UHPLC-ToF-MS.

BACKGROUND: The administration of antibiotics to food-producing animals, including bees, is a common practice employed to prevent or treat bacterial diseases. Such practices may result in the presence of veterinary residues in honey, potentially compromising the safety and health of consumers. In the European Union, maximum residue limits (MRLs) have not been established for pharmacological substances in honey. In this context, the development of accurate and robust analytical methodologies is of paramount importance for appropriate risk assessment. OBJECTIVE: This study aimed to develop and

Compatibility of behavioral disruptors in attract-and-kill formulations for sustainable control of Drosophila suzukii (Diptera: Drosophilidae).

Drosophila suzukii (Diptera: Drosophilidae) is a major pest of soft-skinned fruits, inflicting economic losses through pre-harvest infestation. Among reduced-risk strategies, Attract-and-Kill (A&K) presents an alternative to full-cover insecticide sprays by concentrating exposure to insecticide-laced lures. However, compatibility of insecticides and attractants, as well as application volume, requires optimization to improve efficacy. We evaluated A&K formulations that combined organic Entrust SC (spinosad), Pyganic (pyrethrin), and conventional Gowan Malathion-8 Flowable (malathion), Mustang

Effect of Contact Insecticides on the Transmission of 'Candidatus Liberibacter asiaticus' by Diaphorina citri During Sweet Orange Shoot Development.

Insecticide applications are commonly recommended for managing Diaphorina citri, the vector of huanglongbing (HLB), but their effectiveness in reducing transmission of 'Candidatus Liberibacter asiaticus' (CLas), especially during continuous psyllid influx and shoot growth, remains unclear. This study evaluated the efficacy of foliar application of thiamethoxam and spinetoram in reducing CLas transmission in sweet orange seedlings. Two experiments were conducted up to 13 days after first spray. In experiment 1, CLas-positive psyllids were confined in seedlings every 2 days. Treatments were: con

Comprehensive methodological evaluation of V-ATPase assembly in the context of cardiac lipid overload: implications for (endo)lysosomal function and autophagy.

The vacuolar-type H+-translocating ATPase (V-ATPase) plays a pivotal role in cellular homeostasis by acidifying endosomes and lysosomes, regulating key processes such as autophagy and membrane trafficking. While the importance of V-ATPase in these functions is well-established, the methodologies for studying its assembly and function remain varied and under-characterized. In this study, we systematically validated and compared methodologies for assessing V-ATPase assembly and endo/lysosomal acidification under physiological and high-fat conditions, both in vitro and in vivo. Various techniques
