Development of a TaqMan-based triplex qPCR assay for the simultaneous detection of Muscovy reovirus, novel duck reovirus, and Riemerella anatipestifer.

INTRODUCTION: In duck breeding production, the incidence of infection of Riemerella anatipestifer (RA), Muscovy duck reovirus (MDRV), and novel duck reovirus (NDRV) is gradually increasing. Because the clinical symptoms caused by these three pathogens are relatively similar, it is quite difficult to distinguish and diagnose. METHODS: This study aims to establish a TaqMan-based triplex qPCR detection assay that can detect the above three pathogens at the same time. Specifically designed, optimized and verified specific primers and probes, targeting the dtxr gene of RA and the σB protein coding

Prevalences of respiratory viruses and bacteria in Western Canadian commercial feedlot calves detected using a single metagenomic sequencing protocol vary during the first two weeks of arrival and by age group.

INTRODUCTION: Detection of pathogens associated with bovine respiratory disease (BRD) typically involves several laboratory tools, with results limited to a defined list of targets. This study adapted a previously reported method for metagenomic sequencing of nasal swabs to describe sequencing data from BRD associated viruses. Changes in virus composition were identified between arrival to a feedlot and 14 days on feed (DOF). These data were also assessed for the simultaneous characterization of bacteria and antimicrobial resistance genes (ARGs). METHODS: Nasal swabs were obtained from fall-p

Overexpression of Escherichia coli yaiX Confers Multidrug Resistance and Enhances Virulence in the Silkworm Infection Model.

The emergence of bacteria with both antimicrobial resistance and high virulence has become a global health concern, underscoring the urgent need to elucidate the molecular basis underlying these traits. Here, we employed the silkworm (Bombyx mori) infection model, which is suitable for high-throughput screening, together with an Escherichia coli library containing plasmid clones of all genes from strain W3110, to identify genes whose overexpression enhances virulence. We found that overexpression of the uncharacterized protein YaiX promoted bacterial proliferation in silkworms and increased ho

Spatial distribution and drivers of antibiotics and resistance genes in a key Yellow River tributary.

Antibiotics, various environmental factors, and suspended particulate matter exert a significant influence on the occurrence and dissemination of antibiotic resistance genes (ARGs). However, current research on the mechanisms underlying environmental factor-driven ARG dissemination in sediment-laden rivers, particularly the role of suspended particulate matter, remains incomplete. This study systematically examined the distribution and interaction mechanisms among environmental factors, antibiotics, and ARGs in the Fenhe River Basin, a vital river traversing urban and agricultural areas of Sha

An Easy-To-Use and Field-Deployable Cell-Free Bioreporter for Environmental Detection of Tetracyclines and Macrolides.

The widespread use of antibiotics in agriculture, livestock production, and clinical medicine has resulted in their continuous release into the environment, posing substantial ecological and public health risks. Although chromatographic approaches remain the gold standard for antibiotic detection due to their high accuracy and sensitivity, they require expensive instrumentation, labor-intensive workflows, and trained personnel, limiting their suitability for rapid or on-site monitoring. Here, we report a synthetic biology-based cell-free bioreporter that integrates antibiotic-responsive transc

Manganese thresholds govern antagonism-synergy switching in tetracycline phytotoxicity in Lemna minor.

Tetracycline antibiotics frequently co-occur with manganese (Mn) in freshwater environments, yet how Mn background gradients reshape mixture phytotoxicity remains insufficiently resolved. The objective of this study was to systematically investigate how varying Mn background levels modulate the phytotoxicity and interaction patterns of tetracycline antibiotics. Using Lemna minor as a model species, we employed full-factorial Mn × tetracycline concentration matrices (tetracycline, TC; oxytetracycline, OTC; and chlortetracycline, CTC) to quantify Mn-dependent modulation of tetracycline toxicity

Distinct Osteogenic Profiles of Tetracyclines from Different Generations in an Ex Vivo Embryonic Chick Femur Model.

Tetracyclines are broad-spectrum bacteriostatic agents with well-established antimicrobial efficacy and a shared core chemical structure, differentiated by distinct functional substitutions across generations. Beyond their antibacterial action, tetracyclines also exhibit pleiotropic biological effects, including modulation of bone metabolism. Nevertheless, the selection of agents and dosing for local bone applications remains largely empirical. Therefore, this study compares the tissue-level osteogenic potential of four tetracyclines from distinct generations using a translational ex vivo embr

Structure-Based Drug Design Targeting the Substrate-Binding Pocket of MexB.

The multidrug-resistant (MDR) phenotype of Pseudomonas aeruginosa poses a significant clinical challenge and frequently causes severe and potentially lethal infections. The activity of efflux proteins, which are membrane transporters that use the electrochemical gradient across the bacterial membrane to extrude antimicrobials and reduce their intracellular concentrations, is a significant factor in this resistance. One of the most well-known mechanisms of multidrug resistance in Pseudomonas aeruginosa is the Resistance-Nodulation-Division (RND) efflux pump system. MexB, the inner membrane tran

Embedding mediator function into laccase-like nanozymes: HAT@BPD-Cu as a self-contained laccase-mediator platform to enhance nanozyme performance.

HBT is a common laccase mediator whose N-OH group can be oxidized to generate nitroxyl radicals, thereby enhancing laccase catalytic activity. This enhancement effect has also been demonstrated in laccase-like nanozymes. However, the poor recoverability of HBT may lead to secondary pollution, which limits its environmental applications. Based on the structural homology of nitrogen-containing heterocycles, this study innovatively incorporated 1-hydroxy-7-azabenzotriazole that simultaneously features a nitrogen-containing heterocycle and an N-OH active site into the 4,4'-bipyridine-Cu-based lacc

Levels of tetracycline and sulphonamide residues in freshwater fish: A systematic review and meta-analysis.

The widespread use of antibiotics in freshwater aquaculture has raised significant global concerns because of the accumulation of antibiotic residues in fish and the associated risks to human and environmental health. Tetracyclines and sulphonamides, two of the most applied antimicrobial classes, are of particular interest owing to their broad-spectrum activity, environmental persistence, and ability to select for resistant bacterial populations. This systematic review and meta-analysis conducted using the Population-Exposure-Outcome (PEO) framework, synthesised global evidence on the occurren

Enhancing the Fenton-like activity of molybdenite through Fe(III) ions impregnation.

The Fenton-like application potential of MoS2 has recently attracted widespread interest in advanced oxidation processes. This study utilized natural molybdenite ore as the source of MoS2, and the Fenton-like reactivity of molybdenite was further enhanced by dropping Fe(III) ions on the basal planes of MoS2. EDS elemental analysis and XPS results suggest that Fe species were successfully anchored on the molybdenite surface, which exhibited exceptional efficiency in peracetic acid (PAA) activation. The Fe-molybdenite/PAA system achieved 83.2% degradation of sulfamethazine (SMT) within 30 min, a

Global in silico genomic epidemiology of tetracycline resistance in Staphylococcus aureus: one health dynamics and genotype-phenotype associations.

Tetracyclines remain widely used in human and veterinary medicine for the treatment of Staphylococcus aureus, including methicillin-resistant strains, and are key agents at the human–animal–environment interface. However, the global genomic epidemiology of tetracycline resistance mechanisms, their temporal stability, host structuring, and genotype–phenotype relationships across the tetracycline class remain incompletely resolved. We performed a large-scale in silico analysis to characterize tetracycline resistance determinants, minimum inhibitory concentration (MIC) patterns, and One Health dy
