Antibiotics at subinhibitory concentrations are key drivers of resistance gene dissemination in the environment; however, the mechanisms operating with recipient bacteria remain poorly understood. Here, we propose a Physical-Physiological Coupling hypothesis to explain how tetracyclines promote plasmid transfer. Using a combination of molecular simulations and biological assays, we show that oxytetracycline exerts the strongest effect, increasing conjugation frequency by 4.1-fold relative to controls. Mechanistically, oxytetracycline acts as a specific modulator that binds tightly to the TolC

Klebsiella pneumoniae is an important emerging pathogen leading to serious clinical outcomes as the multidrug-resistant (MDR) isolates arise. While K. pneumoniae is a well-known pathogen in human clinical settings, its risks in nonhuman hosts and potential for cross-species transmission remain poorly understood. In this study, we performed whole-genome sequencing on 112 K. pneumoniae isolates from four host sources in Northeast China to elucidate their phylogenetic relationships and characterize their antimicrobial resistance (AMR) and virulence profiles. Our analysis identified 53 distinct se

Pseudomonas aeruginosa is a widely prevalent multidrug-resistant pathogen in clinical environments, capable of forming biofilms and evading antibiotic clearance, which poses a significant threat to public health. Targeting the multidrug resistance and biofilm formation mechanisms of P. aeruginosa, this study designed and synthesized a series of novel N-acyl homoserine lactone (AHL)-based quorum sensing inhibitors (QSIs). Through structure-activity relationship optimization and activity screening, compound 5-6 was identified as a lead molecule. By competitively binding to the LasR protein recep

Antimicrobial resistance (AMR) in captive wildlife remains poorly characterized, particularly in zoological collections within the Middle East. This study investigated the occurrence, phenotypic resistance patterns, and genomic characteristics of extended-spectrum β-lactamase (ESBL)-producing Escherichia coli in clinically healthy captive ungulates at Al Ain Zoo, one of the largest zoological institutions located in an urban area in the United Arab Emirates (UAE). Fecal samples were collected non-invasively from 101 ungulates during routine husbandry and screened for ESBL-producing E. coli. An

Colistin, a “last-resort” antibiotic for treatment of human infections caused by multidrug-resistant (MDR) Gram-negative bacteria, has been widely used in veterinary medicine for decades. Farm animals have been identified as a significant reservoir of mcr-1-carrying bacteria with potential zoonotic transmission. This study aimed to characterize mcr-1-positive Escherichia coli isolates obtained from calves in Iran. Rectal swab samples were collected from both diarrhetic and healthy calves and screened for colistin-resistant bacteria. The presence of colistin resistance genes mcr-1 to 5 was exam

The use of antimicrobials in livestock farming drives selection and dissemination of antimicrobial resistance (AMR), prompting implementation of veterinary stewardship programs to reduce antimicrobial usage (AMU). We evaluated changes in AMR on 45 Dutch pig farms before and after tailored, coaching-based interventions using phenotypic testing of Escherichia coli and metagenomic profiling of pooled faeces. Post-weaning pig farms, including nursery and fattening units, entered the intervention in a stepped-wedge design, with intervention periods ranging from 10 to 27 months. Across farms, AMU an

The gut microbiota is a crucial component of a tiger's health and plays a significant role in adapting to changes in food and the environment. Although extensive studies have been carried out on the gut microbiota of tigers, investigating the responses of gut microbial composition and function to preadaptation to wild predation patterns under captive conditions is particularly significant for South China tigers, given that it is the only tiger subspecies existing solely in captive settings at present. Here, we performed shotgun metagenomic sequencing for a comprehensive analysis of the gut mic

This study aimed to analyse the microbiome of chronic infected diabetic foot ulcers (DFUs) using parallel methods: traditional culture (the culturome), 16S rRNA gene sequencing (the microbiome) as well as the Antibiotic Resistance Gene (ARG) profile of isolated strains. Swab samples were collected in parallel from affected ulcers. The microbiome sequencing results identified that all patients had a polymicrobial flora with the five most frequent genus level OTUs as Escherichia, Staphylococcus, Streptococcus, Pseudomonas and the anaerobe Anaerococcus. Microbiological culture from the same swabs

Several species of the genus Acinetobacter are nosocomial pathogens with a well-documented ability to acquire resistance to multiple antibiotics. Although Acinetobacter is one of the most abundant genera in meat processing environments, data on this genus outside of clinical environments remains limited. The objective of this study was to ascertain the prevalence, diversity and antimicrobial resistance profile of Acinetobacter spp. in 200 samples collected from food contact surfaces, non-food contact surfaces, carcasses and final meat cuts across five pork, chicken and beef processing faciliti

Sediment is the sink of antibiotic resistance genes (ARGs) in aquatic systems. The pollution of ARGs in sediments is becoming severe and complicated. However, the relationships between heavy metals/antibiotics with ARGs in sediments are unclear. Thus, we collected river sediment samples to investigate the effects of 18 antibiotics/8 heavy metal (loid)s on ARGs' contamination. Results showed that tetracycline, lipopeptide and sulfonamide were 100% detection, where tetracyclines were the most abundant, ranging from 44 ng/g to 408 ng/g dw. Meanwhile, arsenic, zinc, and cadmium were the most serio

We profiled antibiotic resistance genes, bacterial communities, and mobile genetic elements in untreated hospital wastewater from three tertiary hospitals in Metro Manila using shotgun metagenomic sequencing. The resistome analysis revealed high abundances of genes known to confer resistance against sulfonamides (sul1, sul2), aminoglycosides (aadS), and macrolides/streptogramins (msrE, mphE). High-risk resistance genes were also detected, including those known to confer resistance to β-lactams (blaOXA, blaTEM, blaGES, blaNDM, blaKPC), colistins (mcr-5), and tetracyclines [tet(C), tet(A), tet(L

Salmonella spp. represents a leading cause of foodborne disease globally. Wild aquatic birds inhabiting ecosystems impacted by human activities may serve as reservoirs and dispersers of Salmonella and antimicrobial resistance genes (ARGs), posing significant public health risks. This study evaluated the prevalence, serovars, resistance genes, and genomic relationships of Salmonella in fecal samples from wild aquatic birds across three high-Andean lakes in Ecuador. Of 134 samples collected from 10 species, five (3.73%) tested positive, all from Yahuarcocha Lake, isolated from Fulica ardesiaca a

Klebsiella oxytoca is an opportunistic pathogen linked to hospital-acquired infections, with extensively drug-resistant (XDR) strains posing serious treatment challenges. Here, an XDR K. oxytoca 611 isolated from hospital wastewater was found to carry 59 antimicrobial resistance genes (ARGs) compared to the average of six ARGs for K. oxytoca. Twenty-eight of these ARGs were on a mega hybrid plasmid (pKO611.1) carrying C, HI2 and HI2A replicons. Among the resistance genes carried on pKO611.1 was a new tetracycline efflux pump tet(E) variant and an uncharacterised AmpC β-lactamase gene. Cloning

Comprehensive characterization of mammalian gut viromes is essential for early detection of commensal and potentially zoonotic viruses and for reducing the risk of cross-species transmission. Viral metagenomics was applied to profile gut viral communities from zoo mammals maintained across multiple zoological institutions in China. Viral communities differed markedly among host dietary guilds, with herbivores exhibiting the highest viral species diversity. In total, 1 027 viral sequences representing five major viral groups were recovered, including multiple mammal-associated astroviruses, pic

Reservoirs are vital drinking-water sources, yet research has largely overlooked antibiotic accumulation in surrounding soils, which can act as terrestrial sinks and secondary sources that reintroduce contaminants into surface waters, pose latent ecological and public-health risks. This study investigated the occurrence, spatial heterogeneity, and ecological risks of soil antibiotics across three land-use types (cropland, forest land, and construction land) surrounding the Jinze Reservoir, Shanghai. A mixed-effects model and a Positive Matrix Factorization (PMF) model were applied to identify
