OBJECTIVE: Genetic variation in the glucagon-like peptide 1 receptor (GLP1R) has been implicated in type 2 diabetes (T2D) risk and treatment response. We investigated whether GLP1R R131Q, which was previously reported as a T2D-protective variant in genome-wide association studies, alters receptor signaling, β-cell function, and response to glucagon-like peptide 1 receptor agonists (GLP-1RAs).
RESEARCH DESIGN AND METHODS: Trajectory of β-cell function was evaluated in a 20-year community-based prospective cohort in Korea (n = 6,373), where participants underwent biennial 2-h 75-g oral glucose

Oral insulin delivery is challenged by a cascade of gastrointestinal barriers, including mucus entrapment, poor apical endocytosis, extensive lysosomal degradation, and insufficient basolateral exocytosis. To overcome these obstacles in a coordinated fashion, we engineered a dual-ligand PLGA nanoparticle (PLGA-Tf/BAC-NPs) decorated with transferrin (Tf) and the sorting peptide BAC (ADDIDLLK). Tf facilitates efficient apical endocytosis via transferrin receptor (TfR) recognition, whereas BAC not only participates in AP-2-mediated internalization but, more critically, reroutes intracellular traf

Oral peptide delivery systems are conventionally engineered to improve intestinal absorption, but they rarely address the progressive and systemic nature of chronic inflammatory disorders such as type-2 diabetes (T2D), which requires both durable glycemic control and immunometabolic management. Here, we report an oral delivery system based on porous calcium carbonate/sodium alginate (PCC/SA) microparticles loaded with exenatide-chitosan oligosaccharide (EXE-COS). The PCC/SA carrier facilitates the efficient intestinal absorption and transepithelial transport of EXE-COS. Notably, in the lamina

BACKGROUND: Pregnant women in India are routinely prescribed oral iron supplements as part of the national Anemia Mukt Bharat (AMB) program for anemia control. In women with iron-deficiency anemia, the benefits of iron supplementation during pregnancy, both for the mother and child, are very clear. In non-anemic pregnant women, however, the benefits of iron supplementation vis-à-vis associated risks that have been reported have not been studied adequately. Studies have suggested that iron may impair insulin secretion, thus increasing the risk of gestational diabetes mellitus (GDM).

BACKGROUND: Diabetes mellitus constitutes a major global health burden. L. sativum has long been employed in traditional medicine for glycemic management, an effect attributed to its rich phytochemical constituents, including flavonoids, phenolic acids, and glucosinolates. This study investigated the ameliorative effects of L. sativum oil against streptozotocin-induced type 1 diabetes in rats, with particular emphasis on pancreatic and duodenal homeobox 1 (PDX-1) and glucokinase (GCK) gene expression.
METHODS: A total of 48 young male Wistar rats were randomly divided into four experimental g

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have rapidly expanded in use for the treatment of obesity and type 2 diabetes, including among women of reproductive age. Emerging evidence suggests that GLP-1 signaling may influence reproductive physiology through central effects on the hypothalamic-pituitary-ovarian axis and indirect metabolic pathways. In women with polyendocrine metabolic ovarian syndrome (PMOS), GLP-1 RAs improve insulin resistance and androgen profiles and may improve menstrual regularity. Most GLP-1 RAs do not significantly affect oral contraceptive pharmacokinetics

Imeglimin, a novel oral antidiabetic agent classified as a glimin, was developed for the treatment of type 2 diabetes (T2D). This single-center, single-arm interventional study evaluated the efficacy and safety of 24-week imeglimin treatment in patients with metabolic dysfunction-associated steatotic liver disease (MASLD) and T2D. Patients with MASLD and T2D were treated with imeglimin 1000 mg twice daily for 24 weeks. The primary endpoint was reduction in alanine aminotransferase (ALT) levels from baseline. Secondary endpoints included changes in metabolic parameters (including hemoglobin A1c

Major discoveries in the past have laid the foundation on which modern diabetes research has been built, and future findings can be anticipated. From this groundwork, we have pursued studies evaluating the role of β-cell function and β-cell mass in the pathogenesis of type 2 diabetes. In considering β-cell function, studies of β-cell responses to intravenous and oral glucose have highlighted the critical nature of interpreting these responses in the face of the prevailing insulin sensitivity, and not as isolated responses. The critical role of β-cell dysfunction in the pathogenesis of prediabe

Type 2 diabetes (T2D) is a major global health problem driven largely by insulin resistance, the impaired cellular response to insulin. Few current therapies directly address this underlying cause. Thiazolidinediones, potent peroxisome proliferator-activated receptor gamma (PPARγ) agonists, remain the only true small-molecule insulin sensitisers, but their clinical use is limited by adverse effects associated with non-selective activation of this target. Consequently, research has shifted toward alternative pathways that enhance insulin signalling without relying on PPARγ agonism. This review

Werner syndrome (WS) is a rare autosomal recessive progeroid disorder caused by mutations in the WRN gene and is characterized by distinctive physical features and metabolic complications, including insulin resistance, diabetes, hypertriglyceridemia, and hepatic steatosis. Although lipodystrophy is a recognized component of WS, clinical reports of metreleptin use in this setting remain very limited. We report the case of a 38-year-old woman with WS complicated by partial lipodystrophy, diabetes with suboptimal glycemic levels, hypertriglyceridemia and severe hepatic steatosis. She presented wi

The effects of oral contraceptive pills (OCPs) on metabolism and body composition remain unclear. This study aimed to compare fasting glucose and lipid metabolism between naturally cycling (NAT) females, second-(OCP2), and third-generation (OCP3) OCP users during low-(LH) and high-hormone (HH) phases. Forty-nine healthy females (age: 21 ± 2 years, BMI: 23.5 ± 3.8 kg/m2) were assessed. Participants were assessed in two randomized fasting blood draws during: (1) LH phase (NAT: early follicular, OCP: placebo pill week), and (2) HH phase (NAT: mid-luteal, OCP: third week of active pill use) to ass

Anthocyanins, as water-soluble natural pigments, have garnered increasing interest in the field of natural products due to their significant biological activities. While existing studies have verified the hypoglycemic potential of Lonicera caerulea anthocyanins, few investigations simultaneously cover in vitro enzyme activity, in vivo hypoglycemic efficacy and the underlying regulatory mechanisms relevant to Type 2 diabetes. This research investigates the extraction conditions, and hypoglycemic effects in vitro and in vivo of L. caerulea anthocyanin-rich extract (LCAE). The extract was prepare

Type 2 diabetes (T2D) is a growing global health challenge. With prevalence projected to rise significantly in the coming decades, T2D is expected to contribute to increased healthcare costs due to associated morbidity and mortality. Characterized by insulin resistance and impaired pancreatic insulin secretion, T2D could be mediated by the effects of glucagon-like peptide 1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). Incretin-based therapies have, therefore, emerged as effective treatments for improving glycemic management and weight reduction in people with T2D. Since the

Clinical evidence supports the safety and efficacy of imeglimin, a new oral antidiabetic medication that increases insulin secretion and reduces insulin resistance. However, comparative real-world safety data with dipeptidyl peptidase-4 inhibitors, widely used in Japan, remain limited. This retrospective, pharmacy-based cohort study compared the incidence of adverse events (gastrointestinal events and hypoglycemia) among patients newly prescribed imeglimin or sitagliptin in real-world settings. Using a new-user design, we analyzed the 2022-2024 community pharmacy data across Japan, including 1

Early life stress (ELS) significantly contributes to metabolic and psychological disorders by triggering oxidative stress and disrupting the hypothalamic-pituitary-adrenal (HPA) axis. This research examined ELS effects on redox status, along with anxiety-like behavior, in adolescent male Wistar rats, also assessing saffron (Crocus sativus L.) extract's potential benefits. Rats were exposed to foot shock stress from postnatal days 15-19, with treatment groups receiving saline or saffron extract (10 mg/kg) via intraperitoneal injection. Control groups were maintained without stress exposure. Ele

Concentrates are typically fed twice daily to supplement forages in horses to facilitate management, especially those with higher energy needs, but high-grain meals can contribute to digestive and metabolic challenges, including insulin dysregulation. This study investigated the effects of feeding frequency on glycemia, insulinemia, and insulin sensitivity measured by an oral sugar test in moderately exercised young horses. Thirty-two healthy Quarter Horses (2-3 years old) were assigned to either traditional feeding (TF; two grain meals/day) or fractioned feeding (FF; automated grain delivery

Vanadium compounds are among the most extensively investigated insulin-mimetic agents for diabetes mellitus (DM), with reported effects on glucose homeostasis, lipid metabolism, and insulin-related signaling. However, preclinical evidence remains highly heterogeneous due to differences in compound chemistry and experimental design. Then, this study mapped and synthesized preclinical evidence on the effects of vanadium-based compounds on hyperglycemia and dyslipidemia in animal models of DM. This scoping review followed Joanna Briggs Institute methodology and PRISMA-ScR guidelines. PubMed, Scop
