INTRODUCTION: Insulin resistance increases the risk for gestational diabetes mellitus (GDM) and hyperglycemia-associated pregnancy complications. GDM is generally diagnosed at 24-28 weeks of gestation, leaving limited time to address adverse consequences. To explore metabolic markers that predict changes in insulin sensitivity and glycemic responses earlier in pregnancy, we performed a meal tolerance test (MTT) during the early and late second trimester in an at-risk population.
METHODS: We included 30 pregnant women with overweight or obesity in the Pregnancy Outcomes and Maternal Insulin Se

AIMS/HYPOTHESIS: Subcutaneous insulin delivery in individuals with insulin-deficient type 1 diabetes bypasses the portal circulation, disrupting the physiological porto-systemic insulin gradient and affecting postprandial hepatic glucose regulation. However, direct, non-invasive measurement of these liver-specific dynamics and their deviation from normal physiology in individuals with type 1 diabetes is challenging. To address this, we integrated metabolic imaging with whole-body tracer dilution to map postprandial glucose metabolism in both the liver and systemically in adults with type 1 dia

Metformin, a widely prescribed oral biguanide for type 2 diabetes mellitus, has gained significant attention as a potential anti-cancer agent. Beyond its established role in improving insulin sensitivity and lowering blood glucose, preclinical and epidemiological studies suggest that metformin exerts anti-tumor effects through both insulin-dependent and insulin-independent mechanisms. Central to its activity is the activation of AMP-activated protein kinase (AMPK), which disrupts the mammalian target of rapamycin (mTOR) pathway, leading to cell cycle arrest, apoptosis, and reduced tumor growth

Glucose-stimulated insulin secretion (GSIS) in β cells depends critically on membrane depolarization-induced Ca2+ influx. Accordingly, the electrophysiological assessment of intact islets, where β cells act as functional syncytia, is essential for evaluating insulin secretory dynamics. Imeglimin, an oral agent used in treating type 2 diabetes, increases insulin secretion; however, its effects on β-cell electroactivity remain unclear. Here, we investigated extracellular and membrane potentials in imeglimin-treated mouse islets using microelectrode array (MEA) recordings and a plasma membrane po

AIM: Intermittent fasting (IF) has emerged as a beneficial dietary strategy for managing type 2 diabetes mellitus (T2DM), with improvements in certain indicators of body composition and cardiometabolic health. However, limited research compares the different effects of IF in oral hypoglycemic agents (OHAs) versus insulin-treated patients.
METHODS: A comprehensive search was performed across PubMed, Scopus, and Cochrane Library (January 2010-January 2025). Randomized controlled trials (RCTs) involving adults (>18-years) with T2DM undergoing IF, time-restricted feeding (TRF), alternate-day fast

OBJECTIVE: It has been postulated that pregnancies may contribute to the lifetime risk of type 2 diabetes mellitus (T2DM) in women, although direct evidence is lacking. In this context, we hypothesized that pregnancy may impact the long-term natural history of the pathophysiologic determinants of T2DM: insulin sensitivity and β-cell function. We sought to evaluate 10-year trajectories of these features in parous women with and without an additional pregnancy.
RESEARCH DESIGN AND METHODS: Metabolic characterization was done in 303 women at 1 year postpartum and on three or more occasions over

Incretins, glucose-dependent insulinotropic polypeptide (GIP) and glucagon-like peptide-1 (GLP-1) have been shown to improve hyperglycemia in patients with type 2 diabetes, suggesting that an enhanced capacity of GIP and GLP-1 production could be beneficial in type 2 diabetes. We have recently found that dual-specificity tyrosine phosphorylation-regulated kinase (DYRK) inhibitors reduce glucose levels and increase the number of intestinal gip-expressing K-cells and glp-1-expressing L-cells in zebrafish. However, their effects on mammals require exploration in greater detail. In this study, we

INTRODUCTION: Diabetes mellitus (DM) is a global metabolic disorder, with type 2 diabetes mellitus (T2DM) being the most prevalent form. Recent research suggests that excessive sugar intake contributes to cognitive impairments, metabolic abnormalities, and neurodegenerative diseases through oxidative stress and inflammation. The hippocampus, crucial for memory and learning, is highly susceptible to oxidative damage. Linagliptin, a DPP-4 inhibitor, is known for its anti-diabetic properties, yet its potential neuroprotective effects remain unclear. This study investigates whether Linagliptin can

This study investigates the effects of atmospheric pressure plasma jet-activated gas (APPJ) treatment on pectin and its nanoparticles (APPJ-pectin NPs), and explores their applications in oral insulin delivery systems. The results showed that as APPJ treatment progressed, the molecular weight of pectin significantly decreased, while the degree of esterification increased and its hydrophilic/hydrophobic properties altered. The insulin encapsulation efficiency of APPJ-pectin NPs exhibited minimal variation during the initial stage of treatment. In vitro release studies showed that APPJ-pectin NP

BACKGROUND AND AIM: Sodium-glucose cotransporter-2 (SGLT2) inhibitors, a widely used class of oral antidiabetic agents provide additional cardioprotective and renoprotective benefits, but their use during pregnant remains limited. This study aimed to evaluate the association between SGLT2 inhibitor exposure during pregnancy and the risk of congenital malformations.
METHODS AND RESULTS: This population-based cohort study utilized data from the National Health Insurance Service Database (2016-2022). Pregnancies with known teratogen exposure were excluded. SGLT2 inhibitor or insulin (as active c

BACKGROUND: Patients with type2 diabetes frequently face prejudices that result in feelings of stigma, creating an adverse feedback loop that both degrades their quality of life and complicates the management of their illness. The aim of this study is to determine the relationship between stigma and illness acceptance in type 2 diabetes mellitus and the factors that determine it. METHODS: The research was conducted as a cross-sectional descriptive study and between January and June 2025 at the endocrinology clinic of an university hospital. The study included 96 patients. Data were collected u

Type 2 Diabetes (T2D), often preceded by reversible prediabetes, poses a major health challenge. Targeting early glucose dysregulation is a promising preventive strategy. Amino acids and their derivatives are emerging as key regulators of glucose homeostasis. Here, we investigate the role of O-acetyl-serine (OAS), a serine-derived metabolite produced by plants and microbes, including those of the gut microbiota, in glycaemia regulation. We developed a targeted method to quantify OAS in plasma and tissues, showing that it enters circulation and transiently accumulates in the pancreas. In vivo,

BACKGROUND: Higher circulating levels of the metabolite alpha-aminoadipic acid (2-AAA) associate with increased risk of diabetes and cardiometabolic disease. 2-AAA is metabolised from lysine, an essential dietary amino acid. However, the effects of lysine intake on plasma levels of 2-AAA were unclear. We measured post-prandial changes in plasma and urine levels of 2-AAA in healthy individuals in response to oral intake of 13C isotope-labelled lysine and assessed relationships with markers of glucose homeostasis.
METHODS: We recruited healthy individuals (N = 16) to an acute lysine challenge.

Diabetes mellitus affects over 500 million people globally, with current insulin therapies relying on subcutaneous injections that compromise the liver's natural role in glucose regulation and suffer from poor patient compliance. Oral insulin delivery offers a physiological alternative but faces challenges, including gastrointestinal degradation and poor absorption. Here, we engineered Saccharomyces boulardii, a probiotic yeast with established gastrointestinal survival capabilities, to secrete a long-acting Insulin-Fc fusion protein (Ins/Fc). Using strong constitutive promoters (TDH3 and TEF1

OBJECTIVE: Glucose tolerance (GT) is a major effector for adipose tissue (AT) remodeling in obesity, yet its molecular mechanisms remain incompletely defined. We hypothesized that the biophysical and molecular profiles of AT-derived small extracellular vesicles (sEVAT) change in response to glucose availability and differ by GT status.
METHODS: sEVAT were isolated from plasma of individuals with normal GT (NGT) and impaired GT (IGT) (n = 5/group) at fasting (0 h) and 1 h post glucose challenge during oral glucose tolerance test (OGTT). sEVAT were characterized for size, concentration, surface
