Influence of hormonal status in hereditary hemorrhagic telangiectasia - analysis of an online patient questionnaire.

BACKGROUND: Hereditary Hemorrhagic Telangiectasia (HHT) is a rare vascular disorder with mucocutaneous telangiectasia. Hormones are hypothesized to play a role in reducing bleeding in these patients; however, clear evidence is currently lacking. The aim of this study was to further investigate this hypothesis by assessing the impact of altered hormonal status on HHT severity. METHODS: This cross-sectional electronic questionnaire, available in English and German, was distributed with the support of various self-help groups. It included sections on the general medical history of HHT and the inf

Associations between reproductive hormonal milieus and binge eating: The roles of sex and hormonal contraceptive use.

Sex - and its associated gonadal hormones - matters for binge eating (BE). On average, females report higher BE than males. Estradiol generally suppresses appetite, whereas progesterone opposes estradiol's action such that their combined presence represents a high-risk hormonal milieu for BE. Testosterone is thought to be associated with increased BE in females but appears protective in males. Hormonal contraceptives (HCs) provide a naturalistic means to study the relation between gonadal hormones and BE, as HCs vary in formulations and delivery. Oral contraceptive (OC) use has been linked to

Generation and viability evaluation of human precision-cut placental slices for ex vivo studies.

INTRODUCTION: Exposure to medication during pregnancy can compromise placental function, impacting maternal and fetal health. This study aims to evaluate the viability of human precision-cut placental slices (hPCPSs) as a potential in vitro model, offering a new platform for experimental investigations in placenta. METHODS: Decidual and villous hPCPSs of healthy, full-term placentas were prepared with a Krumdieck slicer and were cultured and analyzed at 0 h, 24 h, 48 h, and 72 h. Slice viability was assessed using ATP/protein content, lactate dehydrogenase (LDH) leakage and PrestoBlue assay.

A detailed review of pharmacokinetics/pharmacodynamics of progestogens in oral contraception.

Many different progestogens are used by millions of women worldwide for oral contraception, either alone (Progestogen-only Pills [POPs]) or as combined oral contraceptives (COCs) comprising a synthetic estrogen (ethinyl estradiol [EE]), or a natural estrogen (estradiol and its valerate [E2; E2V] or estetrol [E4]), associated with a progestogen. This review describes first the three families of progestogens derived either from testosterone, 17 alpha-hydroxyprogesterone or spironolactone. Their pharmacokinetic parameters are largely differing, but also their metabolism, potency and efficacy via

Serum progesterone concentrations at frozen embryo transfer in women with adenomyosis and endometriosis: a retrospective cohort study.

BACKGROUND: Endometriosis and adenomyosis are estrogen-dependent inflammatory conditions frequently associated with impaired fertility. Although progesterone resistance at the tissue level is well documented, it remains unclear whether these conditions are associated with altered systemic progesterone exposure during frozen-thawed embryo transfer (FET) cycles and whether higher circulating progesterone levels are required to achieve successful reproductive outcomes. OBJECTIVE: To evaluate mean serum progesterone levels on the day of FET in women with endometriosis and/or adenomyosis compared

The Effect of Menstrual Cycle Phase, Symptoms, Motivation, and Readiness to Perform on Resistance Training Performance.

BACKGROUND: The menstrual cycle can influence a range of physiological and psychological processes that may affect physical performance. However, existing evidence is inconsistent and often based on isolated testing timepoints rather than typical training conditions. Monitoring kinematic outputs during resistance training allows quantification of day-to-day performance changes. OBJECTIVES: This study evaluated whether kinematic outputs during resistance training vary across menstrual cycle phases over two mesocycles and explored associations with symptoms, and perceived motivation and readine

A proteogenomic RNA processing mechanism drives sex differences in meningioma.

Meningiomas are the most common primary intracranial tumors and the only brain tumors that are more common in females compared with males1. Progestin hormonal therapies increase the risk of meningioma, and progestin-induced or pregnancy-associated meningiomas can regress as serum progestogen levels normalize2. The mechanisms that underlie sex differences and progestogen signaling in meningioma are unknown. Here we show that sex hormone interaction with PGRMC1, a transmembrane progesterone binding protein, regulates the activity of RNA processing proteins FXR1 and RBM39 to drive meningioma sex

Winter-associated downregulation of ovarian NR5A2 correlates with impaired follicle development in the striped hamster (Cricetulus barabensis).

To investigate the role of Nuclear receptor subfamily 5, group A, member 2 (NR5A2) in the seasonal regulation of follicle development in striped hamsters (Cricetulus barabensis), we collected 32 adult females, eight individuals from each season (spring, summer, autumn, and winter). Hematoxylin and eosin (H&E) staining was performed to quantify seasonal variations in follicle numbers across distinct developmental stages. Serum concentrations of gonadotropin-releasing hormone (GnRH), follicle-stimulating hormone (FSH), and luteinizing hormone (LH), as well as fecal concentrations of estradiol (E

Comparison of hormone profiles for singleton and twin births in a female giant panda.

The breeding of giant pandas is inherently difficult owing to the short oestrous cycles of the females. Determining the exact time of the oestrous cycle and when the females will attempt to mate may increase the chances of reproductive success. Because the female giant panda at the Everland zoo in Korea did not show signs of oestrus in 2019, her oestrous cycle could not be measured. In 2020, we induced oestrous behaviour in the same panda, used oestrogen peak and trough levels to predict her ovulation period, and two natural mating attempts were conducted in two days. We confirmed and monitore

Short- and Long-Term Impact of Exogenous Hormones on Thermoregulation in Women.

OBJECTIVES: How the body changes its methods of thermoregulation as part of the female ovarian cycle is a crucial aspect of human adaptive strategies to negotiate variable environments and tasks. The use of exogenous hormones to manage the ovarian cycle is a regular part of modern life but its long-term impact on thermoregulation, particularly while moving, continues to be understudied. METHODS: Here, we specifically track adults (N = 35) including men, women of known ovarian phases, and women on an estrogen-progesterone combination pill, doing the same walking and resting protocol while thei

Menopausal Hormone Therapy and Cardiovascular Risk in Midlife Women With Vasomotor Symptoms.

IMPORTANCE: Vasomotor symptoms in perimenopause are associated with increased future cardiovascular (CVD) risk. Most clinical trials on menopausal hormone therapy (MHT) and CVD risk have focused on postmenopausal women, with limited study of the perimenopausal period when hormonal fluctuations and symptoms are greatest. Moreover, these trials were not designed to evaluate CVD risk with MHT among women with vasomotor symptoms. OBJECTIVE: To estimate the effect of MHT on CVD risk among perimenopausal and recently postmenopausal women with vasomotor symptoms and assess the extent to which timing

Expanding the Horizons of Menopausal Hormone Therapy: A Narrative Review of Progestogen-Only and Testosterone-Only Strategies for Tailored Management.

Estrogen-progestogen therapy remains the clinical gold standard for managing menopausal symptoms; however, the growing emphasis on precision medicine underscores the need for individualized strategies for females with specific contraindications or distinct clinical profiles. This narrative review examines the physiological mechanisms, clinical efficacy, and safety profiles of progestogen-only and testosterone-only therapies. Evidence suggests that progestogen-only therapy, particularly oral micronized progesterone (oP4), offers a robust alternative for alleviating vasomotor symptoms and sleep

Effect of conazoles on steroidogenesis mapped by extended H295R-CALUX® assay and LC-MS/MS profiling.

The H295R assay (OECD TG456) is used to screen chemicals for interference with steroidogenesis, measuring testosterone and estradiol levels as endpoints. Focusing on these two hormones, however, may overlook informative changes in other steroid classes, particularly progestogens and glucocorticoids. Here, we extended the effect-based H295R-CALUX® read-out to include progestogens and glucocorticoids using PR- and GR CALUX® assays. First, we evaluated whether PR- and GR-CALUX® could be used as H295R read-outs by determining whether control samples produced adequate hormone levels and whether res

Medical Treatment for Endometriosis: One Size Does Not Fit All.

Endometriosis is associated with nociceptive pain, as well as peripheral and central sensitization. Evidence-based treatment suggestions for controlling endometriosis should be based on the convergence of the best scientific evidence, physicians' clinical expertise, and the values and priorities of individual patients. In this non-systematic, comprehensive narrative review, data from available randomized controlled trials and meta-analyses on hormonal treatment for symptomatic endometriosis are interpreted through the lens of clinical experience. The role of patients in defining therapeutic tr

Longitudinal changes in gut microbiota across reproductive states in wild baboons.

BACKGROUND: In humans and other mammals, female reproduction is linked to extensive changes in physiology, immunity, hormones, and behavior. These changes likely shape, and may be shaped by, the composition of gut microbial communities. Characterizing the dynamics of gut microbial change across reproductive states, including its relationship to female physiology, is important for understanding how the gut microbiota influences female and offspring health. RESULTS: Here we characterize longitudinal changes in gut microbiota across reproduction by combining 16S rRNA gene sequencing data from 4,

Literature review of estradiol/dydrogesterone in perimenopausal and postmenopausal women.

Menopausal hormone therapy (MHT) with 17β-estradiol/dydrogesterone (E/D) or Femoston® at a range of doses has been shown to provide effective relief from climacteric symptoms and prevention of osteoporosis in postmenopausal women. However, less is known about personalizing MHT for women who require lower doses, or those with obesity or overweight. A literature search was conducted using PubMed to evaluate the safety and efficacy of E/D in perimenopausal and postmenopausal women, with a focus on women with high body mass index (BMI). The results confirmed that ultra-low and standard doses of E/

The enigma of associations between reproductive health and development of osteoarthritis in midlife: a life-course approach.

Osteoarthritis (OA) is the most common form of arthritis. OA becomes more prevalent in women around the age of menopause, and it has been suggested that the decrease in estrogen at menopause may be important in the pathophysiology of OA development. The association between estrogen exposure and OA is complex; a recent meta-analysis suggested that menopause hormone therapy use may be associated with an increased risk of joint replacement, bringing this research area back into focus. This narrative review used a systematic process, adopting a life-course approach to consider relationships betwee
