A Tocopherol-Related Methyltransferase SlVTE3(2) Regulates Tomato Fruit Ripening and Tocopherol Accumulation.

Tomato fruit ripening is a critical developmental process that determines the fruit quality. However, the regulatory mechanisms underlying the ripening initiation remain incompletely understood. Here, we characterized SlVTE3(2), a tocopherol-biosynthesis-related methyltransferase, and investigated its role in tomato fruit ripening. CRISPR/Cas9-mediated mutation of SlVTE3(2) accelerated ripening initiation, resulting in earlier ethylene production, faster softening, and an earlier color transition, whereas overexpression delayed these processes. Transcriptome analysis showed that SlVTE3(2) affe

Characterization of a Novel Amidase and Catabolic Pathway in Paraburkholderia sp. AP1-9 Involved in Acetaminophen Biodegradation.

Acetaminophen (APAP) is a ubiquitous pharmaceutical contaminant frequently detected in agricultural environments and crop produce due to its extensive consumption and continuous release. Although microbial degradation is a major pathway for APAP removal, the molecular mechanisms underlying APAP catabolism in Paraburkholderia remain unexplored. In this study, an efficient APAP-degrading bacterium, Paraburkholderia sp. AP1-9, was isolated from activated sludge and characterized through genomic, biochemical, and metabolite analyses. Genome analysis identified acdH, encoding a previously uncharact

Mechanisms Underlying Differential Flusulfinam Sensitivity between Oryza sativa and Echinochloa crus-galli.

Flusulfinam is a novel postemergence HPPD-inhibiting herbicide with excellent safety on direct-seeded indica rice, but the mechanism underlying its selectivity between rice and Echinochloa crus-galli remains unclear. Rice exhibited 41-141-fold higher GR50 values than E. crus-galli, with cultivar-dependent tolerance. Residue analysis showed higher flusulfinam absorption in E. crus-galli, whereas metabolism was significantly enhanced in rice and inhibited by P450 and GST inhibitors. Transcriptomic and qRT-PCR analyses identified OsGSTU16 as highly expressed in rice compared with E. crus-galli. F

Silica Aerogel Microcompartments with On-Demand Reactivation Enable Mimicry of Natural Resurrection.

Mimicking natural resurrection remains extraordinarily challenging. Herein, we report dormant aerogel-based synthetic microcompartments fabricated via a water-in-oil emulsion strategy to produce polyelectrolyte-surfactant complex membrane-coated mesoporous silica aerogel microcompartments loaded with various bioactive enzymes. The dormant aerogel microcompartments feature a high specific surface area (>500 m2/g), mesoporosity, and a superhydrophilic aerogel framework composed of SiO2 nanoparticles. Upon rehydration, the nanoscale capillarity of the aerogel framework, combined with its superhyd

Bridging Catalytic Worlds: A Coassembly Platform for Unified Chemoenzymatic Cascades.

The integration of biocatalytic and chemocatalytic cascades offers a promising route toward sustainable and efficient chemical synthesis. However, inherent incompatibilities between these catalytic systems have impeded progress in this area, particularly regarding their efficient coimmobilization within a single carrier. Herein, we report a general Pickering emulsion-driven interfacial coassembly strategy to construct modular micro/nanoreactors that enable the precise spatial integration of metal nanoparticles and multiple enzymes within a porous crystalline framework, including diverse metal-

Split Cas12a Protospacer Engineering Enables Ultraspecific Recognition of a PAM-Less DNA Strand via an External PAM-Proximal Duplex.

CRISPR-Cas12a is a programmable RNA-guided endonuclease whose activation by double-stranded DNA normally depends on the recognition of an adjacent protospacer-adjacent motif (PAM), which constrains target selection in diagnostic applications. Here, we divide the DNA activator into PAM-proximal (Pp) and PAM-distal (Pd) duplexes, spatially separating the PAM requirement from the interrogation of a PAM-less target sequence. FRET, electrophoretic mobility shift, fluorescence polarization, and functional assays indicate that either fragment can associate with Cas12a, but that robust productive acti

Galvanic Replacement-Driven Structural Evolution of Pd-Pt Alloy Nanoflowers with Modulated d-Band Center for Electrochemiluminescence Biosensing of Organophosphates.

Organophosphorus pesticides (OPs) are widely used agrochemicals that pose health risks through irreversible acetylcholinesterase (AChE) inhibition, necessitating reliable detection. Herein, we report an electrochemiluminescence (ECL) biosensor based on Pd-Pt nanoflowers (Pd-Pt NFs) for diazinon detection, powered by oxygen reduction reaction (ORR). Driven by galvanic replacement, cubic Pd templates evolve into Pd@Pt nanodendrites and finally fully alloyed Pd-Pt NFs. Benefiting from the nanoflower geometry and the ligand effect-modulated Pt d-band center, the catalyst delivers exceptional ORR a

Fe(III)-Superoxo σ-Addition and PCET-Driven L-DOPA 4,5-Cleavage by the Nonheme Enzyme BvDOD in the Biosynthesis Pathway of Betalamic Acid.

4,5-DOPA extradiol dioxygenase (BvDOD) catalyzes the ring cleavage of L-DOPA to generate betalamic acid. Although BvDOD shows a similar fold as those of other extradiol dioxygenases, it utilizes an atypical Fe-coordinate active site. Here, we constructed a reactant model and performed MD and QM/MM calculations to explore the reaction mechanism. Our results reveal that L-DOPA adopts a monodentate binding mode, and the two low-spin states (OS and triplet) correspond to the activated O2. During the reaction, the Fe(III)-superoxo first triggers σ-addition to C4 of L-DOPA, and then a proton-coupled

Monomer Quaternization Enables Antibiofouling and Superior Perm-selectivity of Nanofiltration Membranes for Sustainable Water Purification.

Biofouling poses a substantial challenge to advanced wastewater treatment and reuse via nanofiltration (NF) membranes, while current strategies for enhancing antibiofouling performance often compromise membrane perm-selectivity. Herein, quaternized NF (QPIP-NF) membranes were fabricated via piperazine (PIP) quaternization to simultaneously enhance antibiofouling performance and perm-selectivity. Compared with PIP-NF membranes, QPIP-NF membranes exhibited 1.4-fold higher pure water permeance, slightly increased Na2SO4 rejection, and 1.4-fold higher water/Na2SO4 selectivity, owing to increased s

Enzymatic Ligation Strategy to Enhance Electrospray Ionization Efficiency and Liquid Chromatography─Mass Spectrometry of DNA and RNA Oligonucleotides.

Mass spectrometry (MS) is a powerful technique for characterizing modified RNA as it directly maps and quantifies all mass-altering modifications simultaneously. However, the physicochemical properties of RNA result in poor ionization efficiencies during electrospray ionization, presenting a major barrier to sensitive MS measurements necessary for low-abundance RNA samples and RNAs with low modification stoichiometries. Here, we report a ligation-based approach to increase ionization efficiencies of RNA oligonucleotides. We show that short (∼5 nt), chemically modified DNA oligonucleotides can

A Novel "On-Off-On" Electrochemiluminescence Biosensor from Multifunctional AuPt Triangular Nanoprisms/Carbon Nitride Nanosheets.

In this study, a novel electrochemiluminescence (ECL) biosensor is constructed by employing multifunctional gold-platinum triangular nanoprisms decorated on carbon nitride nanosheets (AuPt/CN) and an exonuclease III (Exo III)-assisted target recycling amplification strategy for the ultrasensitive detection of miRNA-499 via an "on-off-on" readout. CN nanosheets work as an excellent ECL luminophore, while AuPt TNPs enhance catalytic activity and electron-transfer capability and boost ECL emission. The AuPt/CN-modified glassy carbon electrode (AuPt/CN/GCE) produces a strong initial ECL signal, i.

DNA Nanotube-Confined 3D Walker Enables Endogenous Dual-Activated AND Gate Imaging of Tumor-Associated piRNA.

Intracellular molecular imaging for tumor biomarkers faces significant challenges in achieving high specificity and sensitivity, primarily due to signal leakage and the low abundance of target molecules. Herein, we develop an endogenous dual-activation sensing platform (termed E-DWRB) that integrates an AND logic gate driven by endogenous apurinic/apyrimidinic endonuclease 1 (APE1) with the signal amplification of a 3D DNA walker and the spatial confinement of DNA nanotubes for highly sensitive detection of piRNA in living tumor cells. In this design, DNA nanotubes serve as multidirectional tr

Catalytic Network states Govern Programmable Analytical Functions in a Multifunctional Nanozyme.

Although multifunctional nanozymes enable diverse analytical applications, existing activity-based models cannot adequately explain how a single nanozyme is programmably reconfigured to perform distinct analytical functions under different reaction conditions. Here, we show that programmable analytical functions emerge from catalytic network states rather than isolated enzyme-like activities. Using hyaluronic acid-confined platinum nanozymes (HA-Pt) as a model system, we identify an autonomous chromogenic oxidation (ACO) process that proceeds without externally supplied H2O2. Biosynthesis of 3-Hydroxy-3-methylbutyrate by Probiotic Escherichia coli Nissle 1917.

3-Hydroxy-3-methylbutyrate (HMB) is an important nutritional supplement for managing sarcopenia. This study engineered an HMB biosynthetic pathway in the probiotic Escherichia coli Nissle 1917 (EcN). The initial engineered strain exhibited low HMB titers, as carbon flux was primarily diverted toward acetate formation. Deleting the pyruvate oxidase gene (poxB) reduced acetate but caused significant pyruvate accumulation, revealing a severe kinetic bottleneck in endogenous pyruvate dehydrogenase (PDH) activity. Supplementing pantothenic acid (Vitamin B5) profoundly expanded the intracellular coe

A Novel Alkaline-Stable Chitin Deacetylase from Chitiniphilus eburneus for Eco-Friendly Preparation of Specific Deacetylated Products.

Chitin deacetylase is the key enzyme responsible for the biological deacetylation of chitin. However, its limited sources and low catalytic efficiency toward α-chitin present significant barriers to the bioproduction of chitosan. Herein, a chitin deacetylase Ce1425 sourced from Chitiniphilus eburneus was identified, which displayed optimal catalytic efficiency at pH 10.0 and 55 °C, maintaining 91.92% of its activity at pH 12.0. The presence of Co2+ enhanced its activity by 167.38%. Ce1425 exhibited the highest specific activity toward (GlcNAc)3 (8.45 U/mg) and was capable of catalyzing the dea

Synergistic Inhibition of α-Glucosidase by Hyperin, Isoquercitrin, and Quercetin: Mechanistic Insights from Multi-spectroscopic, Calorimetric, Computational, and Caco-2 Cell Assays.

Naturally abundant flavonoids inhibit α-glucosidase (α-GLU), but the mechanisms underlying the synergistic effects among key active compounds remain poorly understood. In this study, the synergistic inhibition of α-GLU by hyperin, isoquercitrin, and quercetin was systematically investigated using fluorescence quenching, isothermal titration calorimetry, circular dichroism, differential scanning calorimetry, computational models, and Caco-2 cell assays. The ternary combination synergistically inhibited α-GLU (IC50 = 0.022 mg/mL, combination index <1) and showed the greatest α-helix reduction an
