Oncolytic reovirus: tumor-microenvironment remodeling, engineering constraints, and translational strategies.

Oncolytic reovirus (ReoV) has emerged as a promising cancer immunotherapy platform because of its natural tumor tropism and immunostimulatory properties. Reovirus enters susceptible cells through receptors including junctional adhesion molecule-A and exploits oncogenic signaling and impaired antiviral responses to preferentially replicate in tumor cells. Viral replication induces apoptosis and other context-dependent forms of programmed cell death, while tumor lysis promotes antigen release and immune activation. However, reovirus monotherapy has shown limited clinical efficacy because of circ

Pituitary adenomas (PitNETs): molecular pathophysiology, targeted drug delivery strategies and clinical therapeutic outcomes.

Pituitary neuroendocrine tumors (PitNETs), historically referred to as pituitary adenomas, comprise a heterogeneous group of neuroendocrine tumors with diverse hormonal phenotypes, molecular characteristics, and clinical behavior. Their management includes surgery, pharmacological therapy, radiotherapy, and, in selected refractory cases, systemic or molecularly targeted approaches. Advances in molecular endocrinology have identified recurrent alterations and dysregulated signaling networks, including GNAS, USP8, PI3K/AKT/mTOR, and MAPK-associated pathways, that contribute to the biological het

The role of immune aging in rheumatoid arthritis and its clinical and therapeutic significance.

Rheumatoid arthritis (RA) is a chronic autoimmune disease, and its onset and progression are closely related to age-related remodeling of the immune system. Immunosenescence refers to age-related changes in the overall structure and function of the immune system; inflammaging is a persistent, low-grade, systemic inflammatory state; cellular senescence refers to a relatively stable cell cycle arrest and functional remodeling of cells under continuous stress. These three are interrelated but correspond to the immune system, systemic inflammatory environment, and cellular state, respectively; the

Anticancer Drugs and Cancer-Associated Thrombosis.

The rapid development of novel cancer therapies has considerably improved the care of cancer patients and cancer-related survival; however, these therapies are associated with a broad range of adverse events that have become a major burden in this population. Alongside other cancer-related factors, vascular toxicity from anticancer therapies contributes to an increased risk of venous thromboembolism. Among anticancer drugs, chemotherapeutic agents (eg, anthracyclines, platinum-based drugs, antimetabolites), hormonal therapies (eg, selective estrogen receptor modulators), agents used for multip

JAK Inhibitors for Alopecia Areata: Approved Therapies, Efficacy, and Unanswered Questions.

Alopecia areata (AA) is a common autoimmune disorder characterized by non-scarring hair loss. While traditionally viewed as a T-cell-mediated disease, recent advances have elucidated the central role of the Janus kinase/signal transducer and activator of transcription (JAK/STAT) signaling pathway in its pathogenesis, establishing a self-sustaining inflammatory loop. The identification of this pathway has transformed AA from a condition with limited therapeutic options to one with targeted biological interventions. Over the past five years, multiple JAK inhibitors have received regulatory appro

MIL-101 (Cr) metal-organic frameworks for systemic delivery of rifampicin antibiotic to lungs.

Respiratory pulmonary infections are a serious threat to human health. Their therapy is primarily based on the use of antibiotics. However, non-specific accumulation and low concentration in the target tissue reduce therapeutic effectiveness, cause side effects, and promote the development of antibiotic resistance. In this study, metal-organic frameworks (MOFs) with MIL-101 (Cr) structure were used for delivery of rifampicin to the lungs. The nanoparticles (NPs) showed high antibiotic loading by mass, namely (127 ± 8)% for MIL-101 (Cr) and (82 ± 7)% for amino-modified NH2-MIL-101 (Cr). The kin

Quercetin rejuvenates aged adipose progenitor cells by attenuating inflammatory pathways.

The mass of inguinal white adipose tissue (iWAT) decreases with age, and its dysfunction contributes to systemic effects, including chronic inflammation, ectopic lipid deposition, and insulin resistance. However, the molecular and functional characteristics of aged adipose progenitor cells (APCs), as well as effective strategies to rejuvenate their adipogenic potential, remain poorly understood. In this study, we found that aged mice exhibited a reduced frequency of APCs, increased inflammatory activity, and impaired adipogenic differentiation capacity. Strikingly, while quercetin exerted conc

A Critical Analysis of the Impact of Etoposide as a Topoisomerase II Inhibitor in Cervical Cancer Treatment: A Review.

BACKGROUND: Etoposide is a semisynthetic derivative of podophyllotoxin and an FDA-approved topoisomerase II inhibitor that induces DNA strand breaks leading to cancer cell death. Cervical cancer remains the fourth most common cancer among women worldwide, with platinum-based chemotherapy constituting the standard of care. Although etoposide has demonstrated broad anticancer activity, its role in cervical cancer therapy is considered secondary and is mainly explored in combination regimens. METHODS: This review summarizes published clinical and preclinical studies evaluating the use of etoposi

Low-dose metformin in the brain: A focused review of its novel mechanism of glucose lowering in type 2 diabetes.

Metformin remains the first-line pharmacological therapy for type 2 diabetes mellitus (T2DM). While its glucose-lowering effects have primarily been attributed to peripheral actions, evidence indicates that it also crosses the blood-brain barrier (BBB) and can exert anti-diabetic effects from within the central nervous system (CNS). This focused review discusses metformin's central actions and how they might integrate with established peripheral mechanisms of glucose-lowering. We synthesized recent mechanistic studies conducted in murine models using systemic and intracerebroventricular metfor

Methylated tirilazad alleviates DSS-induced colitis in mice through reciprocal microbiome-metabolome.

Ulcerative colitis (UC) pathogenesis involves complex interactions between inflammatory reactions, gut dysbiosis, metabolic disruption, and barrier dysfunction. Current therapies primarily target inflammation but fail to correct the underlying dysbiotic ecosystem. We hypothesized that methylated tirilazad (MT), a synthetic 21-aminosteroid with the antioxidant properties of tirilazad, would alleviate colitis by orchestrating a beneficial restructuring of the gut microbiome and its metabolic output. Using a DSS-induced murine colitis model in C57BL/6 mice, we evaluated MT via integrated multi-om

Mn(II) MRI contrast agents supported by unsymmetric pyridinophane-picolinate ligands.

Gadolinium-based contrast agents are commonly utilized in magnetic resonance imaging (MRI). However, manganese-based agents have attracted increasing interest due to their favorable relaxation properties and enhanced biocompatibility, particularly in light of concerns regarding nephrogenic systemic fibrosis associated with the administration of acyclic Gd3+ chelates. In this study, we developed a series of hexadentate and heptadentate chelators based on the 2,11-diaza[3.3](2,6)pyridinophane macrocycle to improve the relaxivity of the corresponding Mn(II) complexes. We evaluated how coordinatio

Biomaterial-Focused Strategies Targeting Dendritic Cells for Autoimmune Disease Treatment.

Autoimmune diseases encompass over 100 distinct diseases where the immune cells betray our body by attacking the tissues they are meant to protect. Even though rare, 15 million Americans are collectively affected by autoimmune diseases, a number that continues to rise every year. Current treatment modalities primarily focus on alleviating symptoms rather than providing prevention or a cure. Tolerogenic dendritic cells (tolDCs) have gained popularity for autoimmune disease treatment as an alternative to traditional systemic immunosuppressive therapies due to their ability to restore immune home

Exploring the potential of HVJ-E for cancer immunotherapy.

HVJ-Envelope (HVJ-E), an inactivated particle derived from the Hemagglutinating virus of Japan, represents a unique virus-based approach for cancer immunotherapy. Unlike replication-competent oncolytic viruses, which exert antitumor effects through viral propagation and tumor cell lysis, HVJ-E lacks replicative capacity and pathogenicity because its viral genome is fragmented. Conversely, although most non-replicative viral vectors are designed primarily as delivery vehicles, HVJ-E itself possesses intrinsic antitumor activity. By preserving the native viral structure and membrane-fusion capac

Stewardship configuration and medicines governance under fiscal stress: lessons from Botswana's 2025 health emergency.

BACKGROUND: Botswana's health system has been characterized in terms of its handling of HIV/AIDS and the comparability of public-sector delivery. These characterizations have pretty good bedrock. The 2025 declaration of an official public health emergency, after disruptions of the supply of almost all essential medicines, threw a spanner in the works. Public discourse changed in response. The conversation now became primarily about budgetary and supply chain procurement challenges. While these were concerns, there were several other issues, the most troubling being the absence of a systemic re

Case Report: Lenalidomide as a novel maintenance therapy for metastatic follicular dendritic cell sarcoma.

Follicular dendritic cell sarcoma (FDCS) is a rare, low- to intermediate-grade malignancy arising from mesenchymal-derived follicular dendritic cells, which can also present with high-grade pathological features and may rapidly become fatal. Conventional therapeutic strategies, primarily consisting of surgical resection with adjunctive systemic chemotherapy, such as gemcitabine-docetaxel, ifosfamide, doxorubicin, or other regimens, remain inconsistently effective, with median survival notably reduced in advanced or recurrent disease. Given the limited therapeutic options and frequent relapse,

Mechanistic insights into selenium nanoparticles: Comparative effects in plant and animal systems.

Selenium nanoparticles (SeNPs), characterized by high bioavailability and low toxicity, have emerged as a class of redox-active nanomaterials that can modulate cellular oxidative states. However, previous reviews have largely considered the agricultural and animal-health applications of SeNPs separately, whereas an integrated comparison of their uptake, biotransformation, and redox-regulatory mechanisms across plants and animals has remained lacking. SeNPs uptake and transformation are influenced by species characteristics, nanoparticle properties, and application strategies. In plants, SeNPs

Inflammation Levels and the Health Impact of Anti-Inflammatory Diets on Chronic Obstructive Pulmonary Disease: An NHANES Study.

BACKGROUND: Chronic obstructive pulmonary disease (COPD), the third leading cause of death globally, is closely associated with systemic inflammation in its pathophysiological mechanisms. Previous studies have primarily focused on localized airway inflammation, while the relationship between systemic inflammation markers such as the systemic immune-inflammation index (SII) and the systemic inflammation response index (SIRI)and COPD remains controversial. Additionally, the role of the composite dietary antioxidant index (CDAI) in modulating COPD risk requires further investigation. METHODS: Th
