Calycosin extracted from Astragalus membranaceus root inhibits platelet activation and thrombus formation: Insights from in vivo and in vitro experiments.

Platelets, which mediate hemostasis, have been implicated in cardiovascular diseases such as myocardial infarction and stroke. Calycosin, a flavonoid extracted from the root of Astragalus membranaceus, has diverse biological effects, including anticancer, anti-inflammatory, and antidiabetic effects. Whether calycosin inhibits platelet activation and thrombus formation is unclear. The present study explored the mechanisms underlying the potential antiplatelet and antithrombotic effects of calycosin. Platelet aggregation assays, flow cytometry, and Western blotting were performed to analyze the

Mitochondrial Transplantation Therapy for Ischemic Stroke: Progress and Challenges.

Ischemic stroke is characterized by high morbidity, high disability, high mortality, high recurrence and high economic burden, which remains second leading cause of death and the third leading cause of disability worldwide. Although continuous progress has been made in vascular recanalization therapy and pharmacological treatment, the effect of these treatments for ischemic stroke has not yet met the expectations of patients and their families. Current clinical therapeutic regimens focus primarily on revascularization and antithrombotic, while neuroprotective strategies are still notably lacki

In vivo screening of Thai plant extracts for antithrombocyte activity using zebrafish: discovery of novel antiplatelet compounds.

Current antithrombotic therapies are associated with significant side effects, including gastrointestinal and intracranial bleeding, as well as interactions with diet and other medications. These limitations underscore the need for safer, more effective alternatives. Given the conservation of coagulation and platelet activation pathways between zebrafish and mammals, we investigated the utility of zebrafish as an in vivo model for screening plant-derived compounds with anti-platelet activity. Unlike conventional in vitro platelet aggregation assays, this approach allows simultaneous assessment

Prevention of In-Stent Restenosis After PCI by Saponin Natural Products: Inhibition of Platelet Activation.

Percutaneous coronary intervention (PCI) enables coronary revascularisation and restores haemodynamic stability but also carries risks of delayed complications including in-stent restenosis (ISR), a chronic progressive disease with endovascular damage after PCI, which compromises the long-term efficacy of PCI. Currently, the main treatment strategies for ISR include interventional and drug therapies. From the era of using aspirin as a single antiplatelet agent to the 'gold standard' era of dual antiplatelet therapy, the risk of ISR after PCI has been reduced. However, long-term use of antiplat

A Conotoxin-Derived Peptide Selectively Inhibits PAR-1 Mediated Thrombosis and Neutrophil Extracellular Traps Formation Without Affecting Hemostasis.

Thrombosis is the key driver for ischemic events, including stroke, which is the leading cause of global mortality. However, current antithrombotic therapies carry substantial bleeding risks. Targeting protease-activated receptor-1 (PAR-1), a thrombin-activated receptor central to thrombus growth, represents a promising antithrombotic strategy for safer intervention.we mined the venom gland transcriptome of Conus spp., applied in silico Furin cleavage prediction, and synthesized and screened the anti-PAR-1 activity of a series of conotoxin-derived peptides.Cb-26 exhibited the strongest but rev

Salvia miltiorrhiza (Danshen) and its extracts in the alternative treatment of diabetic nephropathy: mechanisms and clinical perspectives.

Diabetic nephropathy (DN), the most prevalent microvascular complication of diabetes, affects 20–40% of patients and remains a leading global cause of end-stage renal disease (ESRD). Current Western therapies, such as renin–angiotensin–aldosterone system (RAAS) inhibitors (ACEIs/ARBs etc.) and sodium-glucose cotransporter 2 (SGLT2) inhibitors, primarily target single pathways like glycemic control or hemodynamic stress in early DN. These approaches, while effective in reducing albuminuria, are limited by side effects like hyperkalemia in 10–20% of ACEI/ARB users and incomplete addressing of mu

Scutellarin-eluting stents attenuate coronary restenosis by inhibiting the abnormal proliferation and migration of vascular smooth muscle cells through suppression of NF-κB signaling.

BACKGROUND: Coronary restenosis remains a major challenge following percutaneous coronary intervention (PCI), necessitating the development of effective stent-eluting drugs. Previous studies indicate that scutellarin protects vascular endothelial cells and exhibits anti-thrombotic and anti-platelet effects. Notably, our prior research demonstrated that scutellarin specifically counteracts oxidative stress-driven endothelial dysfunction, a key initiating event in restenosis. This combined evidence strongly suggests its potential against in-stent restenosis (ISR). Therefore, this study explores

Prolonged QT interval and risk of recurrent stroke in the Atrial Cardiopathy and Antithrombotic Drugs in Prevention After Cryptogenic Stroke (ARCADIA) trial.

BACKGROUND: QT interval prolongation is associated with incident stroke in the general population. Whether QT prolongation predicts recurrent stroke is unknown. METHODS: The Atrial Cardiopathy and Antithrombotic Drugs in Prevention After Cryptogenic Stroke trial was conducted in North America from 2018 to 2023 and randomised patients with cryptogenic stroke and atrial cardiopathy to apixaban versus aspirin to assess the prevention of recurrent strokes. In this secondary analysis, we excluded patients with missing ECG data and those with prolonged QRS (≥120 ms). The poststroke QT interval was

Clinical relevance of circulating blood microaggregates and reactivation of Epstein Barr Virus in long-term Post-CoVID syndrome patients.

Chronic persistence of systemic symptoms after recovery from active CoVID-19 has become a significant disease burden, named post-CoVID syndrome. Among many pathophysiological hypotheses we focus on impaired hemostasis as well as reactivation of latent Epstein-Barr virus. We now introduce a novel diagnostic morphological approach for visualizing microaggregates circulating in peripheral venous blood, which are large enough to impede capillary blood flow. In addition, secretion of interferon gamma by mononuclear leukocytes in response to peptides of Epstein-Barr virus is increased in these patie

In Vitro Studies of the Effects of Antithrombotic Zn-Dipicolylamine-Harboring Liposomes (DPALs) on Serum Albumin and Human Umbilical Vein Endothelial Cells.

Thrombosis remains a leading cause of cardiovascular morbidity and mortality. During thrombosis, activated platelets and endothelial cells expose phosphatidylserine (PS) on their outer membranes, creating a surface that accelerates clot formation. Current antithrombotic therapies, such as heparin and warfarin, carry significant bleeding risks, highlighting the need for safer alternatives. In response, we developed a PS-targeting liposomal formulation composed of Zn-dipicolylamine (DPA)-cyanine-3[22,22] and 1-palmitoyl-2-oleoyl-sn-glycero-3-phosphocholine (molar ratio 3:97). This DPA-harboring

Recent advances of nanoparticles for ischemic stroke therapy.

Ischemic stroke (IS), as a cerebrovascular disease caused by interrupted blood supply to the neurons, results in a high prevalence of disability and mortality worldwide. Traditional treatment methods including antithrombotic and neuroprotective treatment have been limited by the blood-brain barrier. Nanoparticles (NPs)-based delivery systems explore new possibility for IS therapy because NPs could enhance the targeting ability of drugs the IS region, thus improving the treatment effect. In this review, we first described the pathophysiology of IS. Then, the state-of-the-art progress of NPs-bas

Nanocolloidal Hydrogel Coatings With Multiple Functions for Antithrombogenic Biointerfaces.

Hydrogel coatings offer a promising strategy for improving the biocompatibility and functionality of medical devices. However, conventional hydrogel coatings often suffer from swelling-induced delamination, poor durability, and limited resistance to thrombosis and biological adhesion. Here, we report a nanocolloidal hydrogel coating (NHC) that shows an important and highly impactful combination of properties, such as lubrication, antibioadhesion, and antithrombogenicity. The NHC is prepared by anchoring a layer of nanocolloidal hydrogel made from methacryloyl-modified hyaluronate nanoparticles

Antithrombotic effects and underpinning mechanisms of food-derived bioactive peptides: A review.

Thrombotic diseases are the primary cause of cardiovascular diseases. Existing clinical drugs are associated with side effects such as bleeding and gastrointestinal damage, making the development of safe alternatives a research hotspot. Food-derived antithrombotic peptides (FDAPs) have become promising candidates due to their strong antithrombotic activity, low immunogenicity, and excellent biocompatibility. This paper systematically reviews the latest research on FDAPs from 2016 to 2025, summarizes and discusses their sources, preparation, separation and purification, and structural identific

Advancements in nanomaterials for the treatment and management of vascular surgeries: from drug delivery to biomedical implants.

Vascular surgery is an important procedure that is carried out to treat diseases of the entire vascular system. Intraoperative hemostasis, precise vision of intricate vascular anatomy, restenosis, implant-associated infection, and inadequate endothelium regeneration are among the enduring difficulties that continue to limit vascular surgery. Advanced biomaterial techniques are required since conventional materials and imaging technologies often fail to meet these multifactorial limitations. By enabling synthetic nanomaterials with programmable physicochemical qualities that directly interact w

Selective Inhibition of Integrin β3 Topology Provides a Safer Antithrombotic Strategy.

Cardiovascular and cerebrovascular diseases, the leading causes of death worldwide, are primarily driven by pathological thrombus formation resulting from uncontrolled platelet aggregation at sites of atherosclerotic plaque rupture. Current antiplatelet drugs are designed either to indirectly inhibit intracellular signaling pathways that activate integrin αIIbβ3 or to directly block the binding of the activated integrin to fibrinogen. However, because this interaction is also essential for normal hemostasis, these drugs inevitably increase the risk of serious spontaneous bleeding. In this stud

Significance of 8-iso-PGF2α in cardiovascular diseases.

Oxidative stress (OS), derived from an imbalance between reactive oxygen species (ROS) accumulation and impaired antioxidant defense, is a recognized cause of atherothrombosis, through a complex interaction between low-grade inflammation and platelet activation. Lipid peroxidation, as reflected by the urinary excretion of 8-iso-Prostaglandin F2α (8-iso-PGF2α), is central in the pathogenesis of atherosclerosis. This biochemical abnormality has been observed in patients with cardiovascular risk factors, including diabetes mellitus, obesity, cigarette smoking, hypercholesterolemia, hypertension,
