Microcapillary-assisted acoustoelectric platform for selective single-cell isolation and in situ transfection.

Single-cell transfection technologies hold tremendous promise for precision cell and immunotherapies. However, monolithic integration of cell isolation with targeted intracellular delivery remains technically challenging, especially within complex biological matrices. Herein, we present a microcapillary-assisted acoustoelectric single-cell selective isolation and sonoporative transfection platform (μC-ASSIST), which seamlessly sequences microcapillary-assisted dielectrophoresis (μC-DEP) for deterministic single-cell trapping with high-frequency surface acoustic waves (SAWs) for transient membr

Development of an effective direct qPCR method for cell therapy product quantification in non-clinical biodistribution studies.

BACKGROUND: Evaluating the non-clinical biodistribution (BD) of cell therapy products (CTPs) is essential to predicting their efficacy and safety in clinical trials. Although BD is typically quantified by qPCR following DNA extraction, this process is labor-intensive and time-consuming, and subject to variability in extraction yield, which complicates normalization to organ weight or blood volume. In contrast, direct qPCR enables the quantification of lysed samples without DNA extraction. Here, we systematically developed an effective direct qPCR method applicable to BD studies of CTPs. METHO

Changes in the microbial spectrum and antimicrobial resistance of bloodstream infections in intensive care units before and during the COVID-19 pandemic.

BACKGROUND: Bloodstream infections (BSIs) are major infectious conditions associated with high morbidity and mortality, particularly in intensive care units (ICUs). In critically ill patients, early and appropriate empirical antimicrobial therapy plays an important role in preventing adverse clinical outcomes. However, the increasing burden of antimicrobial resistance necessitates that empirical treatment strategies be continuously reassessed based on up-to-date local resistance data. This study aimed to compare the distribution of bacterial pathogens and antimicrobial resistance profiles of B

Eugenol alleviates dityrosine-triggered insulin resistance and LPS-related inflammatory response by modulating gut microbiota homeostasis.

Insulin resistance (IR) is a key pathological basis of metabolic diseases. Dityrosine (Dityr), a characteristic protein oxidation product commonly present in over-processed foods, can induce IR by disrupting metabolic homeostasis. Eugenol (EUG), a natural phenolic compound with anti-inflammatory and antioxidant activities, has been shown to improve insulin sensitivity. However, its protective effect and underlying mechanism against Dityr-induced IR remain unclear. The present study aimed to investigate the effect of EUG on Dityr-induced IR in C57BL/6 mice and explore its potential mechanisms.

Stage-dependent signaling networks and therapeutic strategies in diabetic retinopathy: from neurovascular dysfunction to angiogenesis and fibrosis.

Diabetic retinopathy (DR) is a consequence of multiple metabolic, neuronal, inflammatory, vascular and fibrotic changes, not a single cause. Early hyperglycaemia, oxidative stress, lipid disturbance, and mitochondrial dysfunction weaken the retinal neurovascular unit before clinically apparent vascular changes develop. With the accumulation of damage, nuclear factor kappa B, high-mobility group box 1, Toll-like receptor 4, receptor for advanced glycation end products, cyclic GMP-AMP synthase-stimulator of interferon genes, and the NOD-like receptor protein 3 inflammasome enhance inflammation,

Targeting glial modulation: molecular mechanisms of Polygonatum in neurological disorders.

Neurological disorders arise from interconnected pathological processes, including oxidative stress, neuroinflammation, mitochondrial dysfunction, impaired proteostasis, synaptic injury and blood brain barrier disruption. Increasing evidence indicates that glial cells are not passive bystanders in these diseases, but central regulators of neuroimmune balance, metabolic support, myelin integrity and tissue repair. Polygonatum spp., a traditional medicinal and edible plant, contains diverse bioactive constituents, including polysaccharides, steroidal saponins and flavonoids, which exhibit multi-

Potential Mechanisms of N-Methyltyramine in Multitarget Regulation of Mental Disorders via the GPCR-cAMP Pathway.

N-Methyltyramine (NMT) is a tyramine derivative widely distributed in citrus plants and the traditional Chinese medicine Zhishi. Pharmacological studies have confirmed that NMT directly binds to the dopamine D₂ receptor (DRD2; Ki = 31.3µM), inhibits forskolin-induced cAMP accumulation (EC₅₀ ≈ 4µM, Emax = 89%), and dose-dependently antagonizes quinpirole-mediated β-arrestin2 recruitment, exhibiting G protein-biased agonistic properties. Meanwhile, NMT activates the human trace amine-associated receptor 1 (hTAAR1; EC₅₀ ≈ 2µM) to increase cAMP, possesses α₂-adrenergic receptor antagonistic activi

A Chiral Virus-Like Nanoplatform That Broadly Mediates Antibiotic Delivery to the Brain for Bacterial Meningitis Therapy.

Blood-brain barrier (BBB) preserves brain homeostasis yet restricts therapeutics; in bacterial meningitis, antibiotics' limited penetration impairs delivery to the parenchyma and cerebrospinal fluid, constraining efficacy and motivating strategies to overcome the barrier. To address this, we developed a chiral virus-mimicking mesoporous silica nanostructure (CVSN) functionalized with L-alanine for antibiotic delivery. It offers several distinct advantages: (1) nanoscale dimensions facilitating BBB transport; (2) virus-like morphology enhancing vascular accumulation and penetration, providing a

Integrating graph attention networks and deep learning with multi-stage computational screening: Bisdemethoxycurcumin as a potent A₂A receptor inhibitor for Parkinson's disease.

Parkinson's disease (PD) remains a progressive neurodegenerative disorder with limited disease modifying therapies, as current treatments such as L-DOPA offer only symptomatic relief and are associated with severe adverse effects. This study presents an integrated multi-stage computational pipeline that combines quantitative structure activity relationship (QSAR) modelling enhanced by graph attention networks (GATs), molecular docking, density functional theory (DFT), molecular dynamics (MD) simulations, and ADMET profiling to identify natural inhibitors targeting four key PD related proteins:

Influence of ecological and management factors on Toxoplasma gondii infection in small ruminants in Algeria.

Toxoplasma gondii is a zoonotic protozoan of major concern for small-ruminant production and public health. This study is among the first to simultaneously compare T. gondii seroprevalence across contrasting Algerian bioclimatic zones. In summer 2024, a cross-sectional study was conducted across 82 herds comprising 4,022 small ruminants in three provinces of central Algeria (Blida, Médéa, Djelfa), representing contrasting bioclimatic zones (humid, semi-arid, and arid). Blood samples were collected from 1,052 animals older than six months to assess the apparent seroprevalence of T. gondii and a

Administration of injectable platelet-rich fibrin improved repair of thin endometrium via mitochondria in a rat model.

BACKGROUND: Thin endometrium(TE) has been widely recognized as a critical factor contributing to reduced success rates of assisted reproductive technology (ART). In recent years, platelet-rich plasma (PRP) has been extensively employed in the treatment of thin endometrium; however, its underlying mechanisms remain poorly understood, and its clinical efficacy remains controversial. Given the superior therapeutic outcomes demonstrated by platelet concentrates (PCs) in regenerative medicine, the present study aimed to investigate the therapeutic potential and mechanistic basis of injectable plate

Ifi27l2a Facilitates the Recruitment of Pathogenic T Cells by Retinal Microglia in the Initiation Phase of Experimental Autoimmune Uveoretinitis.

The mechanisms by which pathogenic T cells enter the immune-privileged retina at the onset of uveitis remain unclear. To identify regulatory targets that promote retinal inflammation, we established an experimental autoimmune uveoretinitis (EAU) model by immunizing C57BL/6J mice with IRBP651-670 and dynamically evaluated retinal inflammation after induction; fundus lesions were prominent, with the first 14 days representing the initiation phase, Days 14-28 the amplification phase, Days 28-35 the peak phase, and Days 35-42 the regression phase. We characterized retinal cellular alterations from

Osteoarthritis as a marker and potential contributor to cardiovascular risk: a glycocalyx-cartilage axis hypothesis.

Osteoarthritis (OA) may be more than simply a reflection of aging; it could represent a symptom of underlying vascular disease processes with serious consequences for health and quality of life. The association of OA with other conditions like cardiovascular disease (CVD) suggests OA may be partially systemic and metabolic in nature. As such, this connection underscores the value of exploring the biochemical parallels between articular joints and the endothelial glycocalyx, the protective lining of blood vessels. N-acetylglucosamine may be central to this link because both articular cartilage

Female hormones enhance skin T cell expansion and suppress IFN-γ production.

INTRODUCTION: Estrogen receptors (ERs) regulate immune function, but their expression across immune cell subsets and hormone responsiveness in human skin are not fully understood. METHODS: We used 10-color flow cytometry to quantify ER (ERα, ERβ, and GPER1) expression in peripheral blood mononuclear cells (PBMCs) from healthy donors. We analyzed sex hormone gene expression using public single-cell RNA datasets. Skin immune responses to sodium lauryl sulfate (SLS) were assessed in pre- and post-menopausal women, with or without hormone replacement therapy (HRT). Furthermore, we measured the pr

eVLP-mediated CREM editing enhances cytotoxicity of bicistronic IL-15-armored CD19/BCMA dual-target CAR-NK cells.

Chimeric antigen receptor (CAR)-engineered natural killer (NK) cells represent a promising platform for cancer immunotherapy; however, optimization of CAR architecture and additional engineering strategies remain necessary to improve their functional activity. In this study, we generated CD19/BCMA dual-target CAR constructs based on tandem and bicistronic configurations with or without membrane-bound interleukin-15 (mbIL-15), and further evaluated engineered virus-like particle (eVLP)-mediated CREM editing as an additional strategy for CAR-NK cell engineering. Four CAR configurations were eval

Lipid-based mRNA chimeric antigen receptor T cells for enhanced in vitro antitumor activity.

Chimeric antigen receptor (CAR) T cell therapy provides substantial clinical benefits in hematologic malignancies. However, cell engineering using conventional viral vectors is costly, labor intensive, and prone to safety risks, while electroporation-induced cellular damage limits manufacturing scalability. This study reports a safe, high-efficiency, nonviral platform for generating mRNA-engineered CAR T cells. Herein, eight ionizable lipids are synthesized and formulated into lipid nanoparticles. Systematic screening and structural optimization identified lipid 6 as the lead candidate. Transf

Extracellular vesicles and immune-glial remodeling in lung cancer brain metastasis: potential intersections with exercise-related myokines.

Lung cancer brain metastasis develops through reciprocal interactions between metastatic tumor cells and a brain-specific vascular, glial, and immune microenvironment. Tumor-derived extracellular vesicles (EVs) can participate in this process by altering blood-brain barrier (BBB) properties, influencing brain-directed distribution, and transferring regulatory cargo to endothelial and myeloid recipient cells. This review integrates direct evidence from non-small cell lung cancer (NSCLC) brain or central nervous system metastasis with related lung cancer EV studies, while distinguishing these fi
