Trogocytosis: Emerging insights into cellular communication, immunity, and pathogenesis.

Trogocytosis is a unique intercellular process that can occur when two cells become in contact to each other and one cell "nibbles" portions of another cell's membrane and surface molecules. This phenomenon is widely observed among immune cells such as B cells, T cells, NK cells, and antigen-presenting cells (APCs). Through trogocytosis, molecules including major histocompatibility complexes (MHC), costimulatory receptors, and antigens are transferred, influencing cellular communication and immune regulation. While trogocytosis supports processes like antigen presentation, immune activation, a

Targeting ROCK Isoforms by Natural Phytocompounds: An In-Silico Approach to Modulate Mitochondrial Fission and Mitophagy in Neurological Diseases.

One of the major key organelles for neuronal survival is the mitochondria. To maintain synaptic functions and neurotransmission, these cells rely upon high ATP levels and the regulated process of mitochondrial dynamics (fission and fusion) and mitophagy. Dysregulation in any of the mitochondrial processes, such as excessive fission and/or impaired removal of damaged mitochondria, can lead to damage to nerve cells and cause neuroinflammation, a major cause of various neurodegenerative diseases. Rho-associated coiled-coil kinase isoforms (ROCK1 and ROCK2) have emerged as critical upstream regula

Lactoferrin-modified MSC-derived extracellular vesicles facilitate BBB penetration and enhance gastrodin-induced neuroprotection in Parkinson's disease: involvement of mitophagy.

BACKGROUND: The blood-brain barrier (BBB) severely impedes neuroprotective agent delivery to the brain in Parkinson's disease (PD). Although gastrodin (Gas) possesses neuroprotective properties, its efficacy is limited by poor BBB permeability. This study aimed to develop a brain-targeted extracellular vesicle (EV) delivery system for Gas and elucidate its neuroprotective mechanisms. METHODS: A brain-targeted EV delivery system was developed using mesenchymal stem cell-derived EVs modified with lactoferrin (Lf). Gas was loaded into Lf-EVs via sonication to construct Lf-EVs-Gas nanocomplexes.

Nanoformulated Curcumin Ameliorates Ulcerative Colitis via Gut Microbiota Modulation and Barrier Function Restoration.

The gut microbiota is increasingly recognized as a key mediator of the anticolitis effects of curcumin (CUR). However, the clinical translation of this promising activity is severely hampered by CUR's low solubility and poor bioavailability. Although nanoencapsulation represents a promising strategy to address these limitations, the preventive effect of nanocurcumin (nano-CUR) against colitis remains largely unexplored. The present study aimed to elucidate the preventive effects of nano-CUR on intestinal barrier function, gut microbiota composition, and metabolic profiles in a murine model of

Functional Exhaustion Limits CAR-NK Therapy in Glioblastoma but Can Be Mitigated by Early Repeated Dosing.

With the commercial success of Chimeric Antigen Receptor T (CAR-T) cell therapies in the blood cancer setting, there are increasing numbers of clinical trials evaluating this treatment modality in patients with solid tumors, including brain tumors. Allogeneic CAR-natural killer (NK) cell therapy offers an attractive "off-the-shelf" option using the CAR platform. However, this approach remains comparatively untested clinically, with only one published clinical trial to date in patients with central nervous system (CNS) tumors. Building on our prior in vitro characterization of allogeneic B7H3-C

Natural Products as Anti-Angiogenic Agents: Molecular Targets, Signaling Pathways, and Mechanistic Insights.

Angiogenesis, the process of forming new blood vessels from pre-existing vasculature, is a tightly regulated physiological process that is essential for embryonic development, wound healing, tissue regeneration, and the maintenance of normal tissue homeostasis. However, its pathological activation plays a pivotal role in tumor progression, chronic inflammation, and other angiogenesis-dependent diseases. Current antiangiogenic therapies, though clinically effective, are often limited by toxicity, resistance, and high costs. In this context, natural products have emerged as promising sources of

Advances in CAR-T and CAR-NK cell therapy in follicular lymphoma: a review.

Follicular lymphoma (FL) is the most common indolent B-cell non-Hodgkin lymphoma and is characterized by prolonged survival but recurrent relapses. The unmet need is greatest among patients with high-risk disease, including progression of disease within 24 months of first-line immunochemotherapy (POD24), double-refractory disease, histologic transformation, and exposure to multiple previous lines of therapy. In pivotal single-arm studies enrolling heavily pretreated patients, CD19-directed chimeric antigen receptor T-cell (CAR-T) therapies produced overall response rates of 86-97% and complete

IGF2BP1 in 17q-gain cooperates with MYCN to regulate purine biosynthesis and immunotherapy efficacy in neuroblastoma.

Concurrent chromosomal 17q-gain and MYCN amplification define a subset of neuroblastoma associated with particularly adverse clinical outcomes. However, how these genetic alterations synergize to promote tumor progression and whether their cooperation confers therapeutic vulnerabilities remain elusive. We herein performed a CRISPR-based functional screen targeting 17q-gain genes and identified the RNA m6A reader IGF2BP1 as a critical MYCN collaborator to promote aggressive neuroblastomas via activation of phosphoribosylaminoimidazole succinocarboxamide synthetase (PAICS), a key enzyme involved

The H3K27me3-NR1D1-IFN-γ axis in trained macrophages induced by early respiratory virus infection drives non-T2 asthma in children.

Unlike that of T2 asthma, the pathophysiology of non-T2 asthma remains largely unexplored. However, increasing evidence supports a role for trained macrophages in asthma. In this study, we utilized a non-T2 asthma mouse model associated with early respiratory viral infection to identify the H3K27me3-NR1D1-IFN-γ axis in lung-trained macrophages, which drives non-T2 asthma by promoting Th1 differentiation. Early infection combined with allergen sensitization induced the formation of trained macrophages, which displayed hyperactive methionine cycling and elevated levels of S-adenosylmethionine (S

Effects of natural killer cell depletion on innate and adaptive immune responses to COVID mRNA vaccination.

This study assessed the role that natural killer (NK) cells play in a murine model of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Spike mRNA vaccination. Flow cytometric analyses revealed that mRNA vaccination results in recruitment and maturation of NK cells in the spleen. Primed (CD27+ CD11b+) NK cells increased from 0.35% of all CD45+ cells pre-vaccination to 2.44% at 4 d post-vaccination, while terminally mature (CD27- CD11b+) NK cells increased from 0.85% of all CD45+ cells pre-vaccination to 3.91% at 7 d post-vaccination. We next evaluated Spike mRNA and protein expressi

FDX1 Depletion Activates CD8+ T Cell Antitumor Immunity by Promoting DMBT1 Secretion in Cuproptosis of Colorectal Cancer.

Cuproptosis, a copper-dependent form of regulated cell death, has been documented in colorectal cancer (CRC), yet its impact on the tumor immune microenvironment remains incompletely understood. Here, we demonstrate that elesclomol-induced cuproptosis potently activates innate and adaptive antitumor immunity, markedly enhancing CD8+ T cell cytotoxicity while attenuating exhaustion. Mechanistically, FDX1, which correlates inversely with CD8+ T cell infiltration and predicts poor prognosis, functions as a core regulator of CD8+ T cell functionality during cuproptosis. FDX1 promotes CRC prolifera

Induced pluripotent stem cell-derived natural killer cell therapy induces durable responses in glioblastoma and overcomes resistance via a B7-H3-targeted tri-specific killer engager.

BACKGROUND: Glioblastoma is the most aggressive primary brain tumor, with a poor prognosis and limited treatment options. Natural killer (NK) cell therapy is a promising immunotherapeutic strategy; yet, its efficacy remains limited. We evaluated FT538, a clinical-grade NK product derived from induced pluripotent stem cells (iPSCs), in glioblastoma models. METHODS: FT538, engineered with a high-affinity noncleavable CD16 Fc receptor, a membrane-bound IL-15/IL-15Rα fusion protein, and CD38 knockout, was tested against 13 patient-derived glioblastoma stem-like cells (GSCs) in vitro and in orthot

A strategy for relative quantification of chiral amines in nontargeted metabolomics via formation of fixed adduct ions.

Chemical derivatization technology significantly expands the detection scope and enhances the sensitivity of untargeted metabolomics. However, the complex adduct ion formations and diverse ionization patterns of derivatized products present substantial challenges for compound screening and data interpretation. Additionally, understanding the correlations between chiral metabolites and biological systems is essential for uncovering molecular mechanisms underlying physiological processes. To address these challenges, this study developed the (S), (R)-(5-(2-(((1-((N,4-dimethylphenyl) sulfonamido)

Lysozyme-Associated Quercetin-Manganese Nanocomposites for Kidney-Targeted Antiferroptotic Therapy of Acute Kidney Injury.

Acute kidney injury (AKI) is a life-threatening syndrome in which oxidative stress, ferroptosis, and inflammatory amplification converge to damage renal proximal tubular epithelial cells. Although antioxidant and antiferroptotic therapies are conceptually attractive, their efficacy is restricted by rapid clearance and insufficient accumulation in injured renal tubules. Here, we report lysozyme-associated quercetin-manganese metal-phenolic nanocomposites (QMLNCs) as a kidney-targeted therapeutic platform for AKI. In this design, quercetin (QCT) functions simultaneously as an antioxidant drug an

Profiling peripheral immune cells in Parkinson's disease: A scoping review.

Parkinson's disease (PD) is increasingly recognized as a multi-system disorder with immune dysregulation extending beyond the central nervous system. Although numerous studies have examined peripheral immune alterations in people with PD, findings remain heterogeneous and difficult to reconcile. To clarify the current landscape, we conducted a comprehensive scoping review of human studies profiling peripheral blood immune cells in PD. Following PRISMA-ScR guidelines, we systematically screened the literature and curated studies reporting in vivo and ex vivo immune characterizations from PD pat

Sorafenib enhances graft-versus-leukemia activity through IL-18-mediated macrophage and NK cell crosstalk in FLT3-ITD acute myeloid leukemia.

Sorafenib maintenance post-transplantation reduces relapse via inducing graft-versus-leukemia (GVL) effect in FMS-like tyrosine kinase 3 (FLT3)-internal tandem duplication (ITD) + acute myeloid leukemia (AML) undergoing allogenic hematopoietic stem cell transplantation (allo-HSCT), but the mechanism remains unclear. In this study, we investigated the effect of sorafenib in bone marrow immune microenvironment using single-cell RNA sequencing (scRNA-seq) and explored the mechanisms. We found that sorafenib promoted the macrophage-natural killer (NK) cell crosstalk through interleukin (IL)-18, el

Reviving low-affinity glycan-targeting CAR with combinatorial design.

CD19CAR T cells demonstrate remarkable therapeutic efficacy in the treatment of B cell malignancies. A global limitation of conventional CD19CAR therapy is its mono-specificity. Thus, defining complementary targets to CD19 is critical to avoid target-dependent limitations. Alternatives to protein antigens include secondary modifications, such as glycosylation. One example of aberrant glycosylation in lymphoma is the increase in N-glycans capped with α2,6-linked sialic acid, named CD75s. Here, we generate a CAR that recognizes CD75s based on an IgM monoclonal antibody called HH2. HH2CAR shows c

Acaudiolide attenuates MASH-related liver fibrosis through covalent engagement of SMAD4 and blockade of p-SMAD2/3 nuclear translocation.

Hepatic stellate cell (HSC) activation is a central driver of fibrosis progression in metabolic dysfunction-associated steatohepatitis (MASH), yet effective antifibrotic pharmacotherapies remain limited. Here, we identify acaudiolide, a sulfur-bridged symmetric dimeric macrodiolide isolated from the fungus Acaulium sp. H-JQSF, as a natural-product lead that suppresses HSC activation and experimental liver fibrosis. In a Western diet (WD)/CCl4 murine model of MASH, acaudiolide reduced collagen deposition, alpha-smooth muscle actin (α-SMA) expression and fibrogenic gene expression. Transcriptomi

Kaolin integrated gellan gum composite sponge for rapid hemostasis.

Uncontrolled bleeding can result in severe complications, including hemorrhagic shock, hyperthermia, acidosis, coagulopathy, multiple organ failure, and infection. The development of hemostatic agents or devices capable of achieving rapid hemostasis is therefore critical to minimize excessive blood loss during surgical procedures. In this study, a novel kaolin-integrated, gellan gum based composite hemostat sponges were developed via ionic crosslinking mechanism. The established safety and efficacy of kaolin as a hemostatic agent were leveraged in the design, while a composite strategy was emp
