The Diagnostic Elimination Diet and Challenge Protocol for Management of Patients with Food Intolerance-Purpose and Development.

Background: In the 1960s and 1970s, several authors reported that manipulation of diet by excluding certain additives and natural chemical substances led to apparent clinical benefit for patients with adverse food reactions involving the skin, gastrointestinal tract, respiratory tract, and central nervous system. Methods: Over the past 48 years, patients of all ages presenting to the Royal Prince Alfred Hospital Allergy Unit with adverse food reactions were offered an elimination diet low in natural salicylates, biogenic amines, and glutamate, and free of additives such as preservatives and co

Effects of salicylate on organ-of-Corti vibrations in the second turn of the gerbil cochlea.

Sound-evoked responses of the organ of Corti (OoC) in the living cochlea are enhanced by active processes driven by outer hair cells (OHCs). To assess how suppression of OHC function affects intracochlear mechanics, we applied salicylate-a blocker of OHC electromotility-to the round window of anesthetized Mongolian gerbils and measured both distortion product otoacoustic emissions (DPOAEs) and sound-evoked vibrations of the OoC in the second cochlear turn using optical coherence tomography (OCT). Salicylate reduced DPOAE amplitudes in a time- and frequency-dependent manner and decreased intrac

Targeted analysis of salicylate binding partners in renal epithelial cells reveals major AMPK dependency.

Autosomal dominant polycystic kidney disease (ADPKD) is the most prevalent genetic kidney disease and is characterized by the formation and growth of fluid-filled renal cysts. With the current treatment tolvaptan not suited for the majority of patients, an urgent need remains for novel therapeutics. We have shown before that activation of adenosine monophosphate activated protein kinase (AMPK) with salsalate ameliorates disease progression in vivo to a similar extent as tolvaptan. Salsalate achieves this by affecting multiple disease mechanisms known to be involved in ADPKD pathogenesis, such

A Hybrid Modeling Framework for Predictive Digital Twins of CHO Cell Culture.

Digital twins of mammalian cell cultures hold great potential for predictive bioprocess modeling, yet their development is challenged by the nonlinear dynamics and metabolic complexity of these systems. We present a hybrid computational framework that integrates mechanistic and data-driven modeling to construct predictive digital twins for Chinese hamster ovary (CHO) cell cultures producing monoclonal antibodies. The framework couples ordinary differential equation (ODE) models with constraint-based metabolic modeling and machine learning components trained on Bayesian-estimated metabolic rate

Should the Traditional Algorithm for Syphilis Screening be Abandoned? Neurosyphilis Presenting as Limbic Encephalitis: A Prozone False-Negative RPR Missed by an RPR-Only Initial Screen.

INTRODUCTION: Neurosyphilis is a treatable mimic of viral and autoimmune limbic encephalitis. The prozone phenomenon-antibody excess preventing the antigen-antibody lattice-can render an undiluted non-treponemal test such as the rapid plasma reagin (RPR) test falsely negative, with reactivity restored on dilution. CASE DESCRIPTION: A 37-year-old man presented with seizures and rapidly progressive limbic encephalitis; Brain magnetic resonance imaging (MRI) with fluid-attenuated inversion recovery sequences showed hyperintensities in the amygdalae, periventricular regions, left insula and left

Therapeutic Effect of Rosolic Acid against Endothelial Dysfunction in Diabetic Wistar Rats.

INTRODUCTION: Endothelial dysfunction (ED) results from impaired vascular endothelial cell function, disrupting key processes such as hemostasis, vascular tone regulation, vasculogenesis, angiogenesis, and inflammation. These processes are mediated by a complex signaling network involving hormones, cytokines, and chemokines. ED is recognized as a major contributor to the onset and progression of several micro- and macrovascular diseases, including diabetes. Our previous study demonstrated that the polyphenol Rosolic acid (RA) protects against endoplasmic reticulum (ER) stress-induced ED in vit

N-salicyloyl tryptamine derivative exerts antiatherosclerotic effects by ameliorating endothelial inflammation and apoptosis.

OBJECTIVES: Atherosclerosis (AS) is a chronic inflammatory disease driven by lipids that poses a serious threat to human health. Therefore, the development of drugs with both anti-inflammatory and lipid-lowering properties can provide new hope for the treatment of AS. METHODS: Previous studies have shown that the N-salicyloyl tryptamine derivative B2, formed by combining salicylic acid derivatives and the melatonin skeleton, exhibits strong anti-inflammatory activity and high biosafety. In this study, we used lipopolysaccharide (LPS)/ adenosine triphosphate (ATP) or LPS-stimulated macrophage-

Arboviruses manipulate rice's volatile emissions, protecting insect vectors from natural enemies in the field.

Vector-borne plant viruses depend on insect vectors for transmission and often suppress host defenses that limit vector survival and spread. However, their impact on volatile-mediated indirect defenses remains unclear. Here, we show that rice viruses inhibit methyl salicylate (MeSA) emission, impairing parasitoid recruitment and promoting vector persistence. Field experiments demonstrate that MeSA, a key herbivore-induced volatile, suppresses vector populations by attracting egg parasitoids. Viruses counter this by targeting basic-helix-loop-helix transcription factor OsMYC2, a jasmonic acid s

Targeted alleviation of local inflammation by a hydroxypropyl-cyclodextrin-encapsulated prodrug of methyl salicylate and menthol though Nrf2/NF-κB pathway.

Antibiotics remain the conventional mainstay for acute local inflammation, such as pharyngitis, but concerns over bacterial resistance, superinfections, and hepatorenal toxicity have driven research toward natural alternatives. Methyl salicylate (SAM) and L-menthol (MENT), derived from medicinal plants, exhibit anti-inflammatory, analgesic, and antimicrobial properties. However, their clinical translation is hampered by inadequate synergy, rapid metabolism, and low bioavailability in existing formulations. To overcome these issues, we synthesized an ROS-responsive prodrug, SSM, by conjugating

Occurrence of twelve UV filters and evidence of cis-octinoxate formation in Adriatic coastal waters: Environmental implications.

UV filters used in personal care products are increasingly recognised as emerging contaminants due to their persistence and ecological risks. Detected in aquatic systems at concentrations from ng L-1 to μg L-1, their monitoring is essential for environmental risk assessment but challenging because of their diverse physicochemical properties, requiring sensitive analytical methods. This study optimised a solid-phase extraction gas chromatography-tandem mass spectrometry (SPE-GC-MS/MS) method for the simultaneous determination of twelve UV filters in marine waters. The method showed suitable lin

Towards an attract-and-reward strategy: evaluating nectar resources and HIPVs under laboratory conditions to enhance Aphelinus mali parasitism activity, a key parasitoid of Eriosoma lanigerum.

BACKGROUND: The attract-and-reward strategy involves attracting natural enemies using volatile compounds or insectary plants and rewarding them with refuge and food sources. Plants with accessible nectar and early bloom together with herbivore-induced plant volatiles, such as phenylacetaldehyde (Pal) and methyl salicylate (MeSA), could enhance populations of Aphelinus mali Haldeman (Hymenoptera: Aphelinidae), a main parasitoid of Eriosoma lanigerum (Hausmann) (Hemiptera: Aphididae). RESULTS: The extrafloral nectary plant species Euphorbia segetalis was attractive to A. mali, whereas the flora

Liver enzyme alterations during pregnancy in IBD are not attributable to disease activity or medication.

BACKGROUND: Inflammatory bowel disease (IBD) is often diagnosed during reproductive age, requiring adequate management during pregnancy. Physiological changes during pregnancy affect liver enzymes, complicating interpretation in a population prone to liver test abnormalities. This study aims to investigate liver enzyme trends throughout pregnancy in women with IBD. METHODS: A retrospective cohort study was conducted in three Dutch university hospitals. Pregnant women with IBD were included; liver enzyme levels were analyzed throughout pregnancy using Bayesian modeling. The association between

Fundamental kinetic and selectivity properties of the anti-aging, antioxidant-active ingredient EUK-134.

EUK-134 is a manganese-salen complex, derived from the parent molecule EUK-8, and is widely used in anti-aging skincare formulations because of its potent antioxidant activity, resulting from the catalytic decomposition of reactive oxygen species (ROS). Despite this popularity, the fundamental kinetic properties (k) that govern its efficacy and overall recyclability are not well understood, limiting the understanding of its behavior and further optimization in skincare products and therapeutic applications. Importantly, this knowledge gap makes comparisons to newly developed molecules a challe

Turn-On Terbium(III) Photoluminescence Enables Sensing of Methyl Salicylate by a Biomimetic Airborne Messenger Mechanism.

A low cost, portable optical sensing platform enables rapid and sensitive detection of methyl salicylate (MeSal) in unprocessed samples. The sensing assay is based on a strong photoluminogenic response when MeSal forms a complex with a terbium cation (Tb3+) in aqueous buffer. The assay employs an airtight three-well sensing chamber that allows the volatile MeSal to spontaneously diffuse out of a sample well and partition into a neighboring test well where the intense green Tb-photoluminescence is detected by the naked eye or a cell phone camera. This MeSal vapor diffusion process mimics the bi

Role of methyl salicylate and spontaneous field margin vegetation on plant-arthropod interactions to enhance biological control.

BACKGROUND: Herbivore-induced plant volatiles (HIPVs) play multiple roles in mediating plant-arthropod species interactions. Methyl salicylate (MeSA) is a well-known HIPV given its effectiveness in attracting parasitoids and predators and its use in biological control strategies. In addition, non-crop vegetation of field margins are important habitats that harbor natural enemies but may fail to increase biological control. Volatiles like MeSA could synergistically interact with field margin vegetation and increase biological control within crops. The present study examines through a field expe

From field to landscape: scaling-up the response of natural enemies and predation to methyl salicylate.

In response to herbivory, plants emit herbivore-induced plant volatiles (HIPVs), many of which attract natural enemies of herbivores. One such HIPV, methyl salicylate (MeSA), has been deployed in agroecosystems to enhance biological control. However, the broader agroecosystem context-including field-level management and landscape complexity-can strongly influence natural enemy community dynamics. In this 2-yr study, we evaluated the interactive effects of MeSA, field-level practices (eg pesticide use and cultivar selection), and landscape complexity on natural enemy communities and their ecosy
