2-(Ferrocenylmethyl)pentanoic acid (Fc-VPA), a ferrocenyl analog of valproic acid (VPA), was synthesized, fully characterized and its effect on glutamate decarboxylase (GAD) evaluated in mouse brain homogenates using an HPLC assay. Fc-VPA produced robust, concentration-dependent GAD inhibition (IC50 = 51 ± 4 μM), comparable to 3-mercaptopropanoic acid (3-MPA; IC50 = 38 ± 2 μM), whereas VPA showed weak/biphasic effects under identical conditions. Blind docking to human GAD (PDB 2OKK) supported the trend: Fc-VPA achieved a modestly higher XSCORE than VPA (5.287 vs 4.887) and engaged a broader in

Autism spectrum disorder (ASD) has recently been described as a synaptopathy where dysregulation at the level of the synapse is thought to evoke an excitation-inhibition (E/I) imbalance implicated in its pathogenesis. The mechanisms through which alterations in glutamatergic signaling bring about an E/I imbalance remain elusive. Vesicular glutamate transporter 1 (VGLUT1) and postsynaptic density protein-95 (PSD-95) are two major regulatory proteins of glutamatergic signaling. This study aimed to determine whether a valproic acid-induced (VPA) rat model of autism would be associated with change

Valproic acid (VPA) exposure is widely used to model autism spectrum disorder (ASD)-like phenotypes; however, the developmental timing and sex-specific molecular consequences of postnatal exposure remain incompletely understood. Disruptions in synaptogenesis and synaptic protein dynamics are considered central to the neurobiological underpinnings of ASD. This study investigated age- and sex-dependent behavioral and synaptic alterations following postnatal VPA administration during a critical developmental window (P30-P60). C57BL/6 female and male mice were exposed to a single intraperitoneal d

We present the case of a young gentleman admitted to an inpatient psychiatric ward with a diagnosis of Bipolar Affective Disorder who developed Valproate-induced Hyperammonaemic Encephalopathy (VHE) alongside infection with COVID-19. His condition had exhibited a relapsing and remitting course, relapses being associated with non-compliance with prescribed medication and substance misuse. He had previously been successfully treated with a combination of antipsychotic (haloperidol) and semi-sodium valproate which were re-prescribed following the index admission to hospital. On this occasion howe

Neurodegenerative diseases, like Alzheimer's disease (AD), are characterized by the accumulation of tau aggregates, leading to neuronal dysfunction and cognitive decline. This study explores the development of dual-acting compounds combining sigma-1 receptor (σ1R) agonists and histone deacetylase inhibitors (HDACi) to target these pathological mechanisms. Compounds 2d and 3a demonstrated high affinity for σ1R and significantly reduced tau aggregation and phosphorylation in vitro, notably at the AT8 epitope. These dual-acting compounds destabilized tau aggregates, increased tau solubility, and

Antiepileptic drugs (AEDs) are primarily indicated for controlling epileptic seizures. However, accumulating clinical evidence suggests that their benefits in patients with central nervous system (CNS) tumors extend beyond seizure management. Emerging evidence indicates that AEDs possess direct antitumor activity independent of their antiepileptic effects, highlighting a promising novel direction for CNS tumor therapy. This review elucidates the multifaceted antitumor mechanisms of classic (e.g., valproic acid and levetiracetam) and novel (e.g., cannabidiol) AEDs, including their impacts on me

Parkinson's disease (PD) is characterized by progressive degeneration of nigrostriatal dopaminergic neurons, leading to motor dysfunction and compensatory postsynaptic dopamine receptor alterations. Valproic acid (VPA), a histone deacetylase inhibitor, has shown neuroprotective properties; however, its dose-dependent effects on dopaminergic integrity and dopamine D2 receptor (D2R) regulation remain unclear. Adult male Wistar rats received VPA (200 or 400 mg/kg, p.o.) or vehicle for 20 days prior to unilateral 6-hydroxydopamine (6-OHDA) lesioning. Motor performance was evaluated using the beam

Owing to less efficiency of currently available antiepileptic medications, research is continuing toward the most effective solution. It was observed that the antidiabetic drug metformin (MET) is getting more attention in the modern world due to its good anti-inflammatory and neuroprotective characteristics, and this is the main purpose of this investigation for assessing the antiepileptic impact of MET. In this study, we sought to assess MET's antiepileptic effectiveness in a model of mice by examining its effects on the duration, frequency, and intensity of seizures. Furthermore, we aimed to

BACKGROUND: Schizophrenia (SZ) is a long-term psychiatric condition that affects cognition, thought processing, emotional regulation, and behavior. Epigenetic modifications during pregnancy or early postnatal development increase vulnerability to SZ by disrupting the balance of GABAergic-glutamatergic neurotransmission.
OBJECTIVE: The association between GABAergic interneurons and inflammation has been increasingly investigated. Systemic inflammation alters the expression of GABAergic interneuron markers, including somatostatin and parvalbumin, in various brain regions. These modifications co

Understanding epilepsy, one of the most frequent chronic neurological conditions, requires recognizing it as a spectrum of brain disorders characterized by a persistent tendency to generate seizures. In the 20th century, researchers like James Kiffin Penry advanced the development and clinical testing of antiseizure medications, improving treatment options and patient outcomes. Dr. Penry helped shape modern standards for epilepsy treatment, expanded therapeutic options, strengthened collaboration between researchers and advocacy organizations, and contributed to better outcomes and quality of

BACKGROUND: Neurodevelopmental disorders like autism are on the rise, with the role of genetics and epigenetics being increasingly cited as reasons or contributors. The environmental factors were found to be important as epigenetic modifiers, and pregnant mothers are incrementally exposed to many of these environmental factors, like aluminium (Al), known to damage neurons, even before delivery. However, its presence and persistence inside the central nervous system (CNS), leading to changes in the expression of behaviours, need further research. Hence, this animal study was conceived.

Dopamine (DA) signaling is implicated in neurodevelopmental disorders such as autism spectrum disorder (ASD). Cell-type-specific tools like DAT-IRES-Cre mice are essential for dissecting the underlying circuit mechanisms. However, as the transgenic Cre insertion may affect DA transmission, it is crucial to validate that these Cre mice recapitulate key physiological and behavioral phenotypes in established ASD models. Prenatal exposure to valproic acid (VPA) is a widely used ASD model that captures gene-environment interactions in etiology, and DA system dysfunction has been reported in this mo

Neurodevelopmental disorders, including autism spectrum disorder (ASD), are influenced by both genetic abnormalities and environmental toxicants. Among environmental risk factors, endocrine-disrupting chemicals such as bisphenol A (BPA) and pharmaceutical drugs such as valproic acid (VPA) have been associated with an increased risk of autism. In this study, human induced pluripotent stem cell (iPSC)-derived forebrain organoids were used to model early neurodevelopmental disruptions induced by BPA and VPA exposure. On day 62 of differentiation, forebrain organoids were treated with physiologica
