Subcellular Energetics and Metabolism: Potential Therapeutic Applications.

Part I of this review discussed the similarities between embryogenesis, mammalian adaptions to hypoxia (primarily driven by hypoxia-inducible factor-1 [HIF-1]), ischemia-reperfusion injury (and its relationship with reactive oxygen species), hibernation, diving animals, cancer, and sepsis, and it focused on the common characteristics that allow cells and organisms to survive in these states. Part II of this review describes techniques by which researchers gain insight into subcellular energetics and identify potential future tools for clinicians. In particular, P nuclear magnetic resonance to

Unique pumping-out fracturing mechanism of a polymer-shelled contrast agent: an acoustic characterization and optical visualization.

This work describes the fracturing mechanism of air-filled microbubbles (MBs) encapsulated by a cross-linked poly(vinyl alcohol) (PVA) shell. The radial oscillation and fracturing events following the ultrasound exposure were visualized with an ultrahigh-speed camera, and backscattered timedomain signals were acquired with the acoustic setup specific for harmonic detection. No evidence of gas emerging from defects in the shell with the arrival of the first insonation burst was found. In optical recordings, more than one shell defect was noted, and the gas core was drained without any sign of a

Update on ischemia-reperfusion injury in lung transplantation.

PURPOSE OF REVIEW: Primary graft failure, which represents one of the most frequent causes of early mortality, is mostly caused by ischemia-reperfusion injury (IRI). IRI may also induce rejection, which is the principal cause of mortality after transplantation. It is essential to understand the mechanism of pulmonary IRI for improving the outcomes of lung transplantation, and therefore we reviewed the state of the art concerning pulmonary IRI in lung transplantation. RECENT FINDINGS: Numerous strategies have been conducted to reduce IRI after lung transplantation both from the experimental an

Controlled therapeutic gas delivery systems for quality-improved transplants.

Therapeutic gases enriched into perfusion solutions have been effectively used for the improvement of organ transplant quality. At present, the enrichment of perfusion solutions with gases requires complex machinery/containers and handling precautions. Alternatively, the gas is generated within the perfusion solution by supplemented carbonylated transition metal complexes with associated toxicological concerns when these metals contact the transplant. Therefore, we developed therapeutic gas releasing systems (TGRSs) allowing for the controlled generation and release of therapeutic gases (carbo

Oral drug delivery of therapeutic gases - carbon monoxide release for gastrointestinal diseases.

Deploying the therapeutic potential of carbon monoxide (CO) in various gastrointestinal diseases is challenged by inappropriate oral delivery modes. It is for this challenge, that we developed an easy to use tablet referred to as oral carbon monoxide release system (OCORS) providing precise, controlled, tunable and targeted CO delivery for the treatment of sequelae of gastrointestinal diseases. OCORS is an oral tablet based on sulfite induced CO release from the CO releasing molecule 2 (CORM-2). OCORS performance was detailed as a function of the presence of buffer within the tablet core and t

Inhalation gases or gaseous mediators as neuroprotectants for cerebral ischaemia.

Ischaemic stroke is one of the leading causes of morbidity and mortality worldwide. While recombinant tissue plasminogen activator can be administered to produce thrombolysis and restore blood flow to the ischaemic brain, therapeutic benefit is only achieved in a fraction of the subset of patients eligible for fibrinolytic intervention. Neuroprotective therapies attempting to restrict the extent of brain injury following cerebral ischaemia have not been successfully translated into the clinic despite overwhelming pre-clinical evidence of neuroprotection. Therefore, an adequate treatment for th

Effect of heliox on end-tidal CO2 measurement in healthy adults.

BACKGROUND: Therapeutic gases and other modalities delivered by inhalation may affect the accuracy of capnographic measurements in 2 ways. First is the specificity of the measurement of CO2 within the device, and second is the dilution effect of supplemental gases in the ambient air during CO2 sampling by the device. Our goal was to determine if variables such as inhaled gas composition, gas flows delivered via non-rebreather mask, and mouth open or closed affect measurements of end-tidal CO2 pressure (PETCO2) measured with the Capnostream 20 capnograph. METHODS: We measured PETCO2 and breath

A hypothesis on biological protection from space radiation through the use of new therapeutic gases as medical counter measures.

Radiation exposure to astronauts could be a significant obstacle for long duration manned space exploration because of current uncertainties regarding the extent of biological effects. Furthermore, concepts for protective shielding also pose a technically challenging issue due to the nature of cosmic radiation and current mass and power constraints with modern exploration technology. The concern regarding exposure to cosmic radiation is biological damage that is associated with increased oxidative stress. It is therefore important and would be enabling to mitigate and/or prevent oxidative stre

Soybean oil: a potentially new intravascular perfusate.

BACKGROUND: Given that micelles of lipids are colloids, the hypothesis was generated that the rapid administration of large volumes of soybean oil micelles would be an effective perfusion fluid. We also hypothesized that oxygen loading would be enhanced due to the greater solubility of oxygen in lipids compared to water. METHODS: A 100% lethal mouse model of blood loss was used to compare the ability of soybean oil micelles to that of Ringer's lactate, blood and other fluids, with respect to raising and maintaining the blood pressure for one hour. Oxygen on- and off-loading of various concent

Nitric oxide counteracts the hyperoxia-induced proliferation and proinflammatory responses of mouse astrocytes.

Preclinical studies in the premature baboon evaluating the efficacy and potential toxicity of inhaled nitric oxide indicated a significant effect on astrocyte area density, suggesting phenotypic and functional changes in astrocytes upon exposure to nitric oxide. However, the effects of nitric oxide and oxygen, the two major therapeutic gases utilized in neonatal intensive care, on astrocyte morphology and function remain vastly unknown. Herein, we report that exposure of mouse neonatal cortical astrocytes to hyperoxia results in a proinflammatory phenotype and increase in proliferation without

[Therapeutic gases for newborns: recent developments and future perspectives].

In respiratory care some gases are generally used to support the ventilation. Though oxygen is the most frequently dispensed, other specialty gases has become common practice in the last years. This report reviews the literature concerning the four gases mainly utilized in Neonatal Intensive Care Unit (NICU). Inhaled Nitric oxyde is a selective pulmonary vasodilator largely employed in NICU, while the helium-oxygen mixture, also knows as heliox, is less common, although it is widely note both the capability of decrease the pressure and work of breathing necessary to ventilate the lung, and the

Therapeutic gases for neonatal and pediatric respiratory care.

Though oxygen is the most frequently administered gas in respiratory care, the use of other specialty gases has become common practice in neonatal and pediatric intensive care and emergency departments across the United States. This report reviews the literature and evidence regarding 4 such specialty gases: heliox (helium-oxygen mixture), nitric oxide, hypoxic gas (ie, < 21% oxygen), and carbon dioxide. Because heliox is less dense than air or nitrogen, it offers less resistance and turbulence as an inhaled gas and therefore decreases the pressure and work of breathing necessary to ventilate

Detection of DNA damage in stimulated human lymphocytes after enflurane exposure in vitro.

DNA damage was detected by nucleoid sedimentation in human lymphocytes stimulated with pokeweed mitogen after exposure to enflurane. Enflurane induces DNA damage at an exposure concentration of 0.2 vol%. Higher enflurane concentrations increase the rate of DNA damage. The DNA damage seen after exposure to enflurane concentrations of 0.2 and 3.0% vol is comparable to damage after X-radiation of 0.1 and 0.7 Gy. DNA single-strand breaks can be demonstrated by nucleoid sedimentation and can indicate damage before DNA repair begins. Therefore, detected DNA single-strand breaks may be reversible. Ho
