Dopamine receptor agonists, partial agonists and psychostimulant addiction.

Despite the epidemic growth of psychostimulant addiction over the past years, few pharmacological means of intervention are available to date for clinical treatment. This is of importance since the withdrawal syndrome that follows abstinence from drugs such as cocaine and the amphetamines is characterized, among other symptoms, by intense craving for the abused drug, and this is considered a critical factor leading into relapse of drug use. In this article, Luigi Pulvirenti and George Koob focus on the modulatory role shown by drugs acting at the dopamine receptor on the various phases of psyc

Evaluation of the stereoisomers of deprenyl for amphetamine-like discriminative stimulus effects in rats.

The antiparkinsonian agent l-deprenyl, a selective monoamine oxidase (MAO)-B inhibitor, is a phenylalkylamine derivative which is metabolized in part to l-methamphetamine and l-amphetamine. As the clinical use of amphetamine-like psychostimulants is limited by their potential for abuse, we evaluated l-deprenyl for amphetamine-like discriminative stimulus effects over a range of experimental conditions. Male Fisher rats were trained under a 5-response, fixed-ratio schedule of stimulus-shock termination or a 10-response, fixed-ratio schedule of food-presentation to discriminate between d-ampheta

The pharmacology of 1-phenyl-2-propylamino-pentane (PPAP), a deprenyl-derived new spectrum psychostimulant.

The peculiar tyramine uptake inhibitory effect of (-)deprenyl prompted structure-activity relationship studies aiming to develop new spectrum central nervous system stimulants which are devoid of MAO inhibitory potency and operate de facto as indirectly acting, nonreleasing sympathomimetics. Of the derivatives synthesized for this purpose, 1-phenyl-2-propylaminopentane (PPAP) was selected as the reference substance and its pharmacological spectrum is presented. PPAP is taken up by the catecholamine axon terminal membrane and the vesicular membrane but it is devoid of catecholamine-releasing pr

Sensitization of locus ceruleus neurons during withdrawal from chronic stimulants and antidepressants.

The effects of chronic exposure to cocaine, amphetamine and desipramine on noradrenergic neurons in the locus ceruleus were examined using intracellular recordings in rat brain slices. Animals were treated for 2 weeks with either cocaine, amphetamine or desipramine, and all recordings were made after a 1-week withdrawal period. Cells from all three drug-treated groups showed a significant increase in sensitivity to the acute effects of cocaine and amphetamine in prolonging the time course of the noradrenaline-mediated inhibitory postsynaptic potential. At the highest cocaine and amphetamine do

[The psychopharmacology of narcotics: a preclinical study].

Simple and economic tests for estimating addictive potential of drugs using conditioned place preference and intravenous self-administration paradigms have been developed. Various groups of substances were revealed among 12 opioids which differed by their potencies in analgesics and reinforcing tests. Index of "narcological safety" and "addictive index" were proposed for comparative evaluation of therapeutic and adverse (addictive) effects. Using the self-administration procedure a group of addictive psychostimulants were investigated and quantitative criteria for the reinforcing action of the

Monoamine reuptake inhibitors enhance the discriminative state induced by cocaine in the rat.

Cocaine inhibits the reuptake of dopamine (DA), norepinephrine (NE), and serotonin (5-HT). To investigate the relative role of such reuptake processes in the discriminative stimulus properties of cocaine, male rats (N = 16) were trained to discriminate cocaine (10 mg/kg) from saline in a two-lever, water-reinforced drug discrimination task and were administered neuroactive compounds during substitution or combination tests. The DA reuptake inhibitor GBR 12909 (2-16 mg/kg) completely mimicked cocaine. The reuptake inhibitors for NE (desipramine; 2-8 mg/kg) and 5-HT (fluoxetine; 0.625-5 mg/kg) d

Dopamine transporter: biochemistry, pharmacology and imaging.

In recent years, there has been substantial progress in studying the dopamine transporter, a unique component of the functioning dopaminergic nerve terminal. The transporter has been studied by direct binding techniques using a variety of ligands which function as inhibitors of transport. Analogues of these ligands have been used as photoaffinity labels to solubilize and further characterize the transporter. While a variety of drugs bind to the transporter, it is clear that the transporter may serve as an important drug receptor, particularly for the reinforcing properties of some psychostimul

Psychostimulants, analeptics, nootropics: an attempt to differentiate and assess drugs designed for the treatment of impaired brain functions.

The common characteristic properties of psychostimulants, analeptics, and nootropics are excitatory and disinhibitory effects on the central nervous system. The differences lie in the type of excitatory effect. Psychostimulants produce a general, yet nonphysiologic, activation with subsequent sedation. They generally act in a destabilising manner, disturbing the homoeostatic functions of centrally regulated reactions. Nootropics produce a physiological activation of disturbed or reduced adaptation functions. They have a stabilising effect, increasing the homoeostatic functions of the centrally

Intravenous self-administration of the indirect dopaminergic agonist amfonelic acid by rats.

The reinforcing properties of amfonelic acid, a nonamphetamine psychostimulant, were evaluated in an intravenous self-administration paradigm. Rats were trained to self-administer cocaine (0.5-3.0 mg/kg/infusion) on a fixed-ratio 10 schedule of reinforcement via surgically implanted intravenous catheters during daily 4-hr sessions. Substitution of amfonelic acid (0.0625-0.250 mg/kg/infusion) reliably maintained self-administration in a dose-dependent manner. These data indicate that amfonelic acid can act as a reinforcer in rats, and further suggest that amfonelic acid may have abuse potential

Medical management of the depressed alcoholic patient.

Since the advent of modern psychopharmacology in the 1950s, use of medications in the treatment of many psychiatric disorders has become commonplace. Alcohol use is also widespread. As alcohol can interact with a wide variety of medications to alter drug effects, understanding the interactions between it and psychiatric medicines is important to the efficacious treatment of the depressed alcoholic with these drugs. This article briefly outlines the effects of alcohol and the mechanisms of its interactions with psychiatric medications. The most commonly used classes of psychiatric medicines, in
