Morphological Description and Physiological Changes in the Hindgut of Female Asiophrida xanthospilota (Chrysomelidae, Coleoptera) Across Reproductive Stages.

Fecal retention is a distinctive reproductive strategy in certain leaf beetles, which enables females to use accumulated fecal material to protect their eggs and enhance offspring survival. The adult flea beetle Asiophrida xanthospilota (Baly, 1881) is a specialist herbivore that feeds on the leaves of Cotinus coggygria Scop. (Anacardiaceae). Using light microscopy, scanning electron microscopy, and micro-computed tomography, we described and illustrated the hindgut anatomy of adult female A. xanthospilota during the pre-mated and post-mated reproductive phases. We further examined the physiol

Long-term outcomes of Kono-S anastomosis for ileocecal resections in Crohn's disease: a comparative analysis.

OBJECTIVE: This long-term follow-up study evaluates clinical and functional outcomes after ileocecal resection with either Kono-S or conventional anastomosis techniques in patients with Crohn's disease. The goal was to determine whether the Kono-S approach confers a long-term advantage in preventing disease recurrence and improving quality of life. While the Kono-S anastomosis has shown promise in reducing recurrence rates in Crohn's disease following surgery, most existing evidence stems from short- to medium-term follow-up. High-quality long-term data remain scarce, particularly in real-worl

Cotton farming affects ileal virome in a sedentary wild passerine.

Although a few studies have focused on avian gut virome variation in response to environmental stressors, none have assessed virome in relation to the production of chemically intensive crop-based agriculture that alters food resources and detrimentally affects various aspects of avian health and fitness. In this study, we used shotgun metatranscriptomics to assess whether exposure to cotton (Gossypium spp.) production had a deleterious effect on the ileal virome of sedentary northern mockingbirds (Mimus polyglottos) sampled from two cotton-producing areas (16 birds in total) and one uncultiva

Rumen-protected betaine supplementation improves ileal function by affecting microbe and metabolite in Tibetan sheep.

The research explored the effect of dietary rumen-protected betaine (RPB) supplementation on development and function of ileum in Tibetan sheep. Sixty male lambs (17.72 ± 0.19 kg, aged 2 months) were randomly divided into control group (Ctrl) and RPB group (0.08% Bet). The adaptation feeding period was 10 days, and the experimental period was 90 days. Results suggested that dietary RPB significantly increased villus height, villus height-to-crypt depth (VH/CD) ratio, total antioxidant capacity (T-AOC) (P < 0.05), and decreased malondialdehyde (MDA), lipopolysaccharide (LPS), tumor necrosis fac

Ileal microbial and luminal biomarker dysbiosis precede acute cellular rejection following intestinal transplantation.

The role of gut microbiota in acute cellular rejection (ACR) after intestinal transplantation (ITx) remains poorly defined. This study investigated whether ileal microbial dysbiosis precedes ACR and is associated with barrier injury. We conducted a longitudinal and cross-sectional analysis of 16 ITx recipients. Ileal effluents were profiled via 16S ribosomal RNA gene sequencing, short-chain fatty acid quantification, complemented by graft histopathology and assessment of systemic biomarkers of barrier dysfunction. Eight recipients (50.0%) developed biopsy-proven ACR (median 25.5 days post-ITx)

Crohn's lymphoid aggregates with endothelial clusters colocalise with submucosal fibrosis in fibrostenosing Crohn's disease.

Crohn's disease (CD) involves chronic transmural inflammation of the intestines, leading to progressive wall fibrosis with stenosis and luminal obstruction, predominantly in the terminal ileum. Fibrosis is a significant therapeutic challenge, thus improved understanding of localisation, cellular composition, and cell-cell interactions in CD fibrostenosing lesions (FSLs) may identify potential targetable pathways. Using CD FSL patient resection samples, we identify and quantify novel pathological changes in structure, collagen, and cell numbers for each ileal layer (mucosa, muscularis mucosae,

Candidate Biomarkers for Crohn's Disease: Hub Genes and Regulatory miRNAs Identified by Bioinformatics Analysis.

BACKGROUND: Crohn's disease (CD) is a chronic, complex inflammatory condition that can affect the entire digestive tract, most commonly the terminal ileum. The exact cause of CD remains unknown. Bioinformatics was used in this study to identify the differentially expressed genes (DEGs) and microRNAs (miRNAs) that show potential as diagnostic and therapeutic agents in treating CD. MATERIALS AND METHODS: Datasets were downloaded from the Gene Expression Omnibus database and filtered. DEGs between CD samples and healthy control samples were identified using the GEO2R tool (including GEOquery and

Targeting donor XCR1+ and CD11b+ dendritic cells prevents Th1- and Th17-dependent GVHD within the gastrointestinal tract.

Acute graft-versus-host disease (GVHD) of the gastrointestinal tract is a primary cause of early transplant mortality after bone marrow transplantation (BMT), driven by local antigen presentation and T-cell expansion. We sought to clarify the role of various subsets of donor dendritic cells (DC) in this process to define therapeutic strategies to prevent gut GVHD. Donor plasmacytoid DC (pDC) were prominent in the ileum of BMT recipients without GVHD but were largely absent in gut during GVHD. In contrast, donor XCR1+ conventional DC (cDC) were dramatically increased in the mesenteric lymph nod

Stricture dynamics and prognosis during biologic therapy in ileal Crohn's disease: a real-world cohort.

BACKGROUND AND AIMS: Clinicians need data on the responsiveness of ileal strictures of Crohn's disease (CD) to biologic therapy, yet most trials prioritize mucosal healing. We quantified short‑term changes in ileal strictures under biologic treatment and evaluated their prognostic relevance. PATIENTS AND METHODS: In a post hoc analysis of a prospective cohort, consecutive patients with ileal or ileocolonic CD initiating or switching a biologic (anti‑tumor necrosis factor [TNF], ustekinumab [UST], or vedolizumab [VDZ]) underwent segment‑specific endoscopic assessment at baseline and 6 months.

Tissue-layer-resolved proteome landscape of Crohn's disease strictures highlights potential drivers of fibrosis progression.

The chronic inflammation of Crohn's disease frequently leads to fibrosis and muscular hypertrophy of the intestinal wall. This often culminates in strictures, a serious condition lacking directed therapy. Severe pathological changes occur in the submucosa and muscularis propria intestinal wall layers of strictures, yet stricture-associated proteome changes in these layers is unexplored. We perform unbiased proteomics on submucosa and muscularis propria microdissected from transmural sections of strictured and nonstrictured ileum. Proteome changes in strictured submucosa reflected a transition

Host genotypes interact with microbial communities to modulate gene expression in the human intestine.

BACKGROUND: Inflammatory Bowel Disease (IBD) is characterized by chronic intestinal inflammation and is associated with both altered gut microbiome composition and host genetic risk. Both host genetic variants and the gut microbiome can affect host gene expression in the colon; however, it remains unclear whether interactions between the two (genotype × microbiome, GxM) shape intestinal gene regulation in humans and their contribution to IBD risk. METHODS: We analyzed publicly available data for 86 individuals (64 patients with IBD and 22 controls) in the Inflammatory Bowel Disease Multi'omic

Myosin 5b deficiency alters liver proliferation, zonation, and bile acid composition.

BACKGROUND: Myosin 5b (Myo5b) is a motor protein critical for trafficking proteins to the apical surface of intestinal epithelial cells. Inactivating mutations in MYO5B cause microvillus inclusion disease (MVID), a congenital diarrhea disorder that often leads to liver cholestasis. While Myo5b's role in the intestine is well characterized, its function in the liver remains unclear. METHODS AND RESULTS: To define the hepatic consequences of Myo5b loss, we analyzed germline Myo5b knockout (KO) mice. Bulk RNAseq of KO livers revealed significant transcriptomic alterations, notably downregulation

From Agro-Waste to Therapeutic Potential: Spasmolytic Mechanisms of Vaccinium myrtillus L. Leaf Extract on Isolated Rat Ileum.

Bilberry (Vacciniummyrtillus L., Ericaceae) is chiefly valued as an edible plant for its berries, widely consumed as a functional food, whereas the leaves, as agro-waste, remain an underutilized natural source of bioactives. The traditional use of V. myrtillus leaves is well documented, particularly for managing diabetes and gastrointestinal disorders. However, their potential spasmolytic activity, which could support such uses, remains unexplored. The main objective of this study was to evaluate the spasmolytic potential of V. myrtillus leaf extract on the gastrointestinal tract and to elucid

Ethanolic extract of Morinda citrifolia improves gut microbiota, intestinal morphology, and performance without adverse effects on hematological profiles in broiler chickens.

Over the last six decades, the extensive use of antibiotics as growth promoters in animal nutrition has contributed to antimicrobial resistance and ultimately poses a risk to public and environmental health. Therefore, there is a growing interest in alternative strategies, such as plant extracts and essential oils, to address this issue. The aim of this study was to evaluate the effect of Morinda citrifolia leaf ethanolic extract (MCEE) on the intestinal health and performance of broiler chickens. A total of 360 Cobb 500 broilers were randomly distributed into groups C1, C2, S1, S2, and S3, wi

Quantification of total endogenous nitrogen losses in broilers using a minimally invasive 15N-isotope dilution method.

Total endogenous nitrogen (N) losses (ENLs) measurement in broilers is important for quantifying dietary true N digestibility. Basal ENLs are commonly quantified using enzymatically hydrolysed casein (EHC) or EHC-ultrafiltration (EHC-UF), whereas diet-specific ENLs require regression methods. Total (tENL; basal + diet-specific) ENL can be measured using the 15N-isotope dilution (ID) method. This study aimed to quantify tENL using a minimally invasive ID method based on urinary 15N-enrichment as a proxy for 15N-enrichment in the precursor pool for endogenous protein synthesis, and to compare ID
