[Parasympathetic control of secretion of pancreatic juice in the rabbit (author's transl)].

The secretion of pancreatic juice in response to stimulation of the vagus nerves in the neck, and to injection of parasympathomimetic drugs, was studied in anesthetized rabbits. Vagal stimulation caused a marked increase of protein content in pancreatic juice, and also a small increase in flow of juice, highly variable from one amimal to another, preceded by a short period of inhibition. Atropine supressed vagal effect on protein concentration, but failed to abolish the flow response. Injection of parasympathomimetic drugs closely imitated the results of vagal stimulation, with the difference

Sialic acid in rabbit lacrimal gland fluid.

To date, there has been no attempt to determine which of the orbital glands contribute the sialic acid, which has been found in tears and tear mucoids. In the present study, sialic acid was found in the fluid collected directly from the lacrimal gland excretory duct, uncontaminated by the secretions of the other orbital glands (Nictitans, Harderian, conjunctival) as well as in the fluid secreted into the conjunctival sac by the other orbital glands, uncontaminated by lacrimal gland fluid. At all flow rates, the rate of secretion of sialic acid increased as flow rate increased in both fluids an

Centrally mediated cardiovascular effects of intracisternal application of carbachol in anesthetized rats.

The pressor response to the intracisternal (i.c.) injection of carbachol (1 mug) in anesthetized rats was analyzed. This response was significantly reduced by the intravenous (i.v.) injection of guanethidine (5 mg), hexamethonium (10 mg) or phentolamine (5 mg), and conversely, potentiated by i.v. desmethylimipramine (0.3 mg), while propranolol (0.5 mg) i.v. selectively inhibited the enlargement of pulse pressure and the tachycardia following i.c. carbachol (1 mug). On the other hand, the pressor response to i.c. carbachol (1 mug) was almost completely blocked by i.c. atropine (3 mug) or hexame

Effects of phospholine iodide on the metabolites of the glycolytic, pentose phosphate and sorbitol pathways in the rabbit lens.

Steady-state concentrations of the key intermediates from the glycolytic, pentose phosphate, and sorbitol pathways as well as the pyridine nucleotides were measured from the lens after 0.25% phospholine iodide had been instilled into rabbits' eyes twice a day for 18 weeks. In the lenses of those rabbits which had received treatment in both eyes fructose-1,6-diphosphate and pyruvate levels were increased, whereas 6-phosphogluconate, sorbitol and alpha-glycer0phosphate concentrations were decreased. alpha-Ketoglutarate and concentrations and ratios of NAD+ and NADH did not show any changes. In c

Histamine hypersensitivity in mice induced by Bordetella pertussis or pharmacologic beta adrenergic blockade. Effects of adrenergic, cholinergic, and other drugs.

The effects of prostaglandin E1, E2, F2alpha (PGE2 PGF2alpha), isoproterenol, epinephrine, norepinephrine, salbutamol, practolol, atropine, aminophylline, and corticosterone on the hypersensitivity to anaphylaxis, histamine, and serotonin in Bordetella pertussis-treated mice and propranolol-treated mice were investigated. Female HLA-SW (ICR) mice, 27-29 gm, were injected with pertussis vaccine intravenously 4 days before challenge with antigen, histamine, or serotonin. Alternatively, instead of pertussis vaccine, propranolol was injected intraperitoneally 45 min before histamine challenge. Tes

Rapid eye movements in myasthenia gravis. II. Electro-oculographic analysis.

Voluntary saccades were studied by electro-oculography in ten patients with myasthenia gravis (MG) and in eight patients with other types of ophthalmoplegia. Despite limited range of eye movements, maximum velocities of 20 degree and 40 degree saccades in patients with MG were not significantly different from those in normal individuals, whereas maximum velocities in patients with other types of ophthalmoplegia were significantly decreased. In some myasthenic patients, small amplitude saccades were hypermetric and had high velocities, appearing clinically as "quiver" movements characteristic o

The effect of miotics on the intraocular pressure of conscious owl monkeys.

The intraocular pressure of conscious, unsedated owl monkeys (Aotus trivirgatus) was measured with an applanation tonometer. Untreated eyes of the conscious animals were found to have higher values than those reported for owl monkeys anesthetized with pentobarbitone. Locally applied pilocarpine, carbachol, and oxotremorine gave concentration-related reduction in pressure, oxotremorine being the most potent and having longer duration of effect than the other compounds. Slight reductions were also observed with aceclidine and R. S. 86. These results are discussed in relation to the effects of mi

Effect of humoral modulators of morphine-induced increase in locomotor activity of mice.

The effect of humoral modulators on the morphine-induced increase in locomotor activity of mice was studied. The subcutaneous administration of 10 mg/kg of morphine-HC1 produced a marked increase in locomotor activity in mice. The morphine-induced hyperactivity was potentiated by scopolamine and attenuated by physostigmine. In contrast, both methscopolamine and neostigmine, which do not penetrate the blood-brain barrier, had no effect on the hyperactivity produced by morphine. Pretreatment of mice with alpha-methyltyrosine (20 mg/kg i.p., one hour), an inhibitor of tyrosine hydroxylase, signif

Cholinergic stimulation of the rat hypothalamus: effects of liver glycogen synthesis.

Cholinergic stimulation of the lateral hypothalamic neurons with intrahypothalamic microinjections of acetylcholine or carbachol caused a marked increase in the content of the active form of glycogen (starch) synthase in the liver. Total activity of the enzyme (active plus inactive forms) was not increased significantly. The lowest effective dose of acetylcholine was 5 X 10(-10) mole, and the optimum dose was 5 X 10(-9) mole. Similar applications of other neurotransmitters, such as norepinephrine, dopamine, serotonin, and gamma-aminobutyric acid, did not affect the enzyme's activity.

[Clinical and experimental observations on idiopathic urticaria due to the contact with heat].

This study describes the probably eleventh case, mentioned in literature, of acquired heat contact uticaria in an otherwise healthy young woman. With regard to true contact induction heat contact urticaria clearly differs from the more common cholinergic uticaria. On the other hand, heat contact urticaria is completely analogous to cold uticaria because of the exposure area, reversible blocking by unphysiological prolonged heating of the skin, sensitivity to antihistamines and resistance to corticosteroids. In this case, whealing of the skin occurred on exposure to heating of 39 degrees C for

Experimental tests of hypotheses about neurochemical mechanisms underlying behavioral tolerance to the anticholinesterase, diisopropyl fluorophosphate.

Neurochemical and psychopharmacological studies of rats were designed to examine four hypotheses which have been proposed to account for the development of behavioral tolerance to the anticholinesterase, diisopropyl fluorophosphate (DFP). The fact that the activity of the enzymes, adenosine triphosphatase, alkaline phosphatase and cytochrome oxidase, did not change concomitantly with behavioral measures during chronic treatment with DFP suggests that nonspecific metabolic changes are unlikely mechanisms of behavioral tolerance. Similarly, a lack of change in choline acetylase activity coupled

Factors affecting the time course of decay of end-plate currents: a possible cooperative action of acetylcholine on receptors at the frog neuromuscular junction.

1. End-plate currents have been studied in gylcerol-treated frog sartorius nerve-muscle preparations with the voltage-clamp technique. 2. Adding the anticholinesterase prostigmine (3 muM) to the solution bathing the muscle caused a 2-7 (mean 3-3) times increase in the time constant of decay of end-plate currents. The anticholinesterase edrophonium (15 muM) also prolonged the time course of end-plate currents. 3. Pre-treatment of the preparation with collagenase, which leads to the removal of acetylcholinesterase in the synaptic cleft, prolongs the time course of end-plate currents. 4. Curare (

Pharmacology of the esophageal motor funciton.

In the striated muscle of the upper esophageal sphincter, tonic maintenance of closure is probably mediated via tonic central excitation of the extrinsic motor innervation; relaxation represents central inhibition of this mechanism. The motor nerves are probably cholinergic and act through nicotinic receptors like those of somatic striated muscle. In the striated muscle of the esophageal body, swallowing-induced contraction is also probably a cholinergic and nicotinic response. In the smooth muscle of the esophageal body, the control of contractions is cholinergic and muscarinic in part, but t

Central action of drugs acting on the cholinergic muscarinic receptor I. Influence of cholinomimetic drugs administered into the lateral cerebral ventricle on behavior in rats.

Carbachol (CCh), physostigmine (Pht) and pilocarpine (Pil) injected into the lateral cerebral ventricle (ivtr), diminished spontaneous motility and open-field performance, lowered body temperature, depressed amphetamine-induced hypermotility, and inhibited amphetamine stereotypy. On the other hand, they had no effect on duration of hexobarbital anesthesia, except Pil, which slightly prolonged it, or on the action of threshold dose of pentetrazole and maximal electroshock. The compounds raised levels of "free" and "total" ACh, but not those of "bound" ACh. Level of NA in the brain was decreased

[Treatment of severe forms of myasthenia with prednisolone and triamcinolone].

On the basis of a long-term observation of 1000 patients with myasthenia the authors stress the necessity of a manifold treatment of this disease with the use of anticholinesterase preparations, by correcting the mineral metabolism and by influencing the thymus gland. In specially severe cases an intensive hormone therapy is recommended with large doses of triamcinolon and prednisolon. In 39 cases of a severe myasthenia the used treatment was successful. The paper deals with theoretical prerequisites and methods of treatment by corticosteroid preparations, their combination with other drugs an

The modulation of spontaneous and anti-Ig-stimulated motility of lymphocytes by cyclic nucleotides and adrenergic and cholinergic agents.

Translational movement in B lymphocytes was stimulated by anti-Ig antibody. Drugs that are presumed to elevate cyclic AMP stopped this stimulated motility. Such was the case with dibutyryl cAMP and theophylline, cholera enterotoxin, and isoproterenol, a beta adrenergic agonist. Conversely, in the absence of anti-immunoglobulin antibody, cyclic GMP and the cholinergic drugs acetylcholine and carbamylcholine increased spontaneous motility of lymphocytes, with the B class of lymphocytes demonstrating greater responsiveness. The increase in motility brought about by cholinergic drugs was totally s

Drug effects in a novel biphasic writing syndrome induced by acetylcholine in mice.

The writing induced in mice by i.p. administration of 3 mg/kg acetylcholine chloride showed a biphasic response with time. The primary phase of 1-15 sec after acetylcholine appeared to be the result of nicotinic receptor stimulation. The secondary phase from 30 sec following acetylcholine was selectively suppressed by anti-inflammatory agents and by atropine, while centrally acting analgesicw, other strong central nervous system drugs and ganglion blockers suppressed both phases of writhing to an equal extent. The narcotic partial agonists significantly suppressed primary writhing more readily

In-vitro investigation of a new neuromuscular relaxant, AH8165.

AH8165 was compared with other neuromuscular relaxants in an in-vitro rat phrenic nerve-diaphragm preparation. Concentrations of 6-10 mug/ml AH8165 produced progressive decreases in strength of concentration. AH8165 was 0.1 times as potent as d-tubocurarine, and its effects were more rapidly reversed by washing. The times to recovery from 90 per cent blockade were the same for succinylcholine and AH8165, but the time to recovery from 50 per cent blockade was shorter for succinylcholine than for AH8165. Neostigmine reversed blockade induced by d-tubocurarine to 80 per cent of control, while it

Clinical neuromuscular pharmacology of AH 8165 D, an azobis-arylimidazo-pyridinium-compound.

The neuromuscular blocking properties of AH 8165 D (1,1'azobis-(3-methyl-2-phenyl-1H-imidazo-1,2a-pyridinium)dibromide) were quantitatively studied in 50 patients, undergoing extra-abdominal surgery and anaesthetized with barbiturates, nitrous oxide, fentanyl and droperidol, by means of mechanograms of the hand muscles obtained by ulnar nerve stimulation. AH 8165 D is a rapid-acting nondepolarizing agent (full effect within 1--2 min) providing good intubation conditions similar to those obtained with suxamethonium. AH 8165 D may be considered to be indicated for emergency intubation in patient

Sequestration of neostigmine and metabolites by perfused rat liver.

Isolated rat livers were perfused with 0.9 to 18 mumol of 14C-neostigmine (NEO) or 14C-(3-hydroxyphenyl)trimethylammonium (HPTMA). Within 30 min. 30-90% of the administered doses were sequestered by the liver. The initial extraction rate was equivalent to about 15% of the perfusion rate. There was no evidence for saturation of the rate or the extent of sequestration of NEO or HPTMA over the dosage range examined. The amount of radioactivity in the liver remained relatively constant for several hours while NEO was converted to HPTMA and its glucuronide (G-HPTMA), plus small amounts of other met
