Background/Objectives: Thrombosis, a critical condition that can have severe consequences, such as myocardial infarction and cerebral infarction, can be induced by steroid drugs. Although the mechanisms for inducing thrombosis are known for some types of steroid drugs, much remains unknown about the differences in the tendency and mechanisms for thrombosis. Methods: To address this knowledge gap, we analyzed the relationship between thrombosis and steroid use by utilizing the U.S. Food and Drug Administration Adverse Event Reporting System database. From the database, we extracted demographic

Measurement of adrenal hormones in amphibians is important in studies on stress, development, osmoregulation, endocrine disruption, and conservation to help reveal mechanisms within amphibians and evolution among vertebrates. Corticosteroids measured in frogs are typically corticosterone and/or aldosterone, but steroid intermediates that may activate hormone receptors are thus far not quantified. Also, steroidogenesis in frogs has been examined in vitro but little work has been done using tadpoles with mutations affecting steroidogenesis. To advance such studies, we developed a comprehensive l

OBJECTIVE: Accurate assessment of steroid hormones is essential for understanding sexual development and metabolic health. Blood-based steroid assays are informative but invasive and reflect only short-term fluctuations. We compared serum and hair steroid hormone profiles in adolescents and examined their associations with body size and composition, with attention to sex-specific patterns.
DESIGN: Cross-sectional analyses in a population-based sample of adolescents attending the 8-year follow-up examinations of the PANIC study in the city of Kuopio, Finland.
METHODS: The analyses included 24

Adrenocortical carcinoma (ACC) is an endocrine malignancy with a poor prognosis due to its advanced stage at diagnosis and early metastatic potential. Misclassification is common when tumors arise near the renal-adrenal interface, particularly in cases with vascular invasion. We describe the case of a patient with a large retroperitoneal mass extending into the inferior vena cava whose initial imaging suggested a renal origin, resulting in deferred hormonal evaluation. Postoperative pathology confirmed ACC, and subsequent endocrine testing was consistent with a nonfunctional tumor. This case r

Polycystic ovary syndrome (PCOS) is one of the most frequent endocrinopathology affecting women in their reproductive ages. However, PCOS is also related to metabolic abnormalities such as metabolic syndrome (MS), insulin resistance (IR), and type 2 diabetes, among others. Consequently, an inflammatory and pro-oxidative status is also present in these patients, aggravating the syndrome's symptoms. This work aims to discuss some late treatments that focus on oxidative stress (OS) as a central feature related to primary PCOS abnormalities. Therefore, this review focuses on the evidence of anti-o

Steroids, including mineralocorticoids, glucocorticoids, and sex hormones play a critical role in pregnancy. Liquid chromatography tandem mass spectrometry (LC-MS/MS) analysis offers an opportunity to conduct simultaneous multiplex steroid analysis within a given sample. This paper describes the LC-MS/MS steroid analysis method developed for assessing plasma specific reference ranges of 18 steroids from plasma samples (200 µL) of pregnant women participating in the PREDO study. Samples were prepared using supported liquid extraction and analyzed on an Acquity I-Class UPLC and a QTrap6500 + mas

Despite advances in treatment and the expansion of standard care, pneumonia remains a major cause of mortality. It frequently leads to complications such as septic shock and acute respiratory distress syndrome (ARDS), both of which carry high fatality rates. Although antimicrobial therapy is the cornerstone of treatment, additional supportive care and adjunctive therapies, such as corticosteroids, are often required, especially in severe community-acquired pneumonia (CAP).Recent updates to major guidelines on CAP, sepsis, ARDS, and critical illness-related corticosteroid insufficiency generall

Adrenal insufficiency is a disorder of the adrenal glands, where the glands fail to produce adequate mineralocorticoids, glucocorticoids, and adrenal androgens. The most common cause of primary adrenal insufficiency in developed countries is autoimmune; however, medications, infectious diseases, and trauma can also induce adrenal insufficiency. Exogenous steroid use is also a common cause of secondary adrenal insufficiency. Symptoms are related to alterations in the salt and water balance in the body and develop slowly over several months. Patients usually develop fatigue, nausea, vomiting, ab

The mineralocorticoid receptor plays a central role in homeostasis, mediating the regulation by aldosterone of epithelial sodium transport. In addition, it regulates a range of responses in other tissues where it is likely responding to both mineralocorticoids and glucocorticoids. Structural, functional and evolutionary studies have provided insights into the mechanisms of receptor activation by agonist ligands and how interactions within the domains of the mineralocorticoid receptor may modulate the response to individual ligands including the mechanisms of antagonism. This review will discus

INTRODUCTION: Patients with heart failure with reduced ejection fraction (HFrEF) experience higher rates of in-hospital and all-cause mortality. On the basis of the VERINA trial, vericiguat is indicated for patients who have had a worsening heart failure (WHF) event despite optimal heart failure (HF) therapy, or for those with tolerability concerns.
METHODS: This multicenter, prospective, single-arm, non-randomized, real-world, descriptive study assessed the efficacy and safety of vericiguat in Indian patients aged ≥18 years with chronic HFrEF who were vericiguat-naïve and started treatment a

It is well-established that the majority of steroid hormones in the bloodstream are in a bound state, complexed with carrier proteins. Transcortin (corticosteroid-binding globulin, SERPINA6) serves as the principal transport protein for steroid hormones and is predominantly synthesized in the liver. Its primary function is to regulate the systemic bioavailability of glucocorticoids and mineralocorticoids. This review article presents an analysis of the existing literature on the impact of various factors and conditions on Transcortin, encompassing its synthesis, secretion, and affinity, as wel

Hypoadrenocorticism is a serious condition in dogs that results from autoimmune adrenalitis and depletion of mineralocorticoids and glucocorticoids. Affected dogs respond well to glucocorticoid supplementation and treatment with the synthetic mineralocorticoid desoxycorticosterone pivalate (DOCP). DOCP injected once monthly resolves serum Na/K abnormalities and normalizes water balance, but therapy is expensive. Cost abatement involves prolongation of the 30-day dosage interval or decreasing the 2.2 mg/kg dosage. These approaches are not based on DOCP pharmacokinetics. A full assessment of the

Orchidectomy and estrogenization of the male represent a procedure that is applicable in sex reassignment or in prostate cancer therapy. This approach has an influence on the hypothalamic-pituitary-adrenal axis and thus affects cardiovascular function and metabolism. We utilized orchidectomized rats to evaluate the effects of estradiol on the structure and hormonal output of the adrenal gland. Adult Wistar rats were divided into sham-operated (SO; n=7), orchidectomized (Orx; n=7), and estradiol-treated orchidectomized (Orx+E; n=7) groups. Estradiol-dipropionate (0.625 mg/kg b.m.) was administe

The adrenal glands are paired endocrine organs that play a vital role in maintaining homeostasis by regulating blood pressure, metabolism, electrolyte balance, and the body's response to stress. Each gland comprises the adrenal cortex, which produces steroid hormones, and the adrenal medulla, responsible for catecholamine synthesis. The cortex is further divided into three functional zones, the zona glomerulosa (mineralocorticoids), zona fasciculata (glucocorticoids), and zona reticularis (androgens), while the medulla secretes adrenaline, noradrenaline, and dopamine under sympathetic control.
