Discovery of Subnanomolar and Highly Selective Carbonic Anhydrase II Inhibitors Featuring L-Shaped Scaffolds via Active-Site-Matched Fragment Growing.

Glaucoma is a leading cause of irreversible blindness worldwide, and lowering intraocular pressure (IOP) remains the only proven method to delay progression. While topical carbonic anhydrase (CA) inhibitors are established treatments, more potent and selective agents are needed. We report a series of subnanomolar and highly selective human carbonic anhydrase (hCA) II inhibitors developed through a multistage fragment growth strategy. By incrementally expanding a weakly bound sulfanilamide anchor to precisely complement the enzyme's topology, we achieved a remarkable 15,000-fold increase in pot

High-Throughput Binding Kinetic Measurements of DNA-Encoded Library-Derived Hits By Focal Molography.

DNA-encoded libraries (DELs) enable rapid discovery of large pools of small organic ligands against target proteins. Currently available hit validation methodologies are limited in their ability to characterize binding kinetics for a large number of molecules. Only a subset of hits identified by DEL screening are followed up on, while many potentially relevant binders remain uncharacterized. Here, we propose focal molography as a high-throughput, label-free optical methodology for parallel kinetic measurements of DEL-derived hits. The methodology was applied to measure binding kinetics (kon an

Research progress on potential high-risk drugs for Stevens-Johnson syndrome and toxic epidermal necrolysis with insights from adverse drug reaction database mining.

Stevens-Johnson Syndrome (SJS) and Toxic Epidermal Necrolysis (TEN) are rare yet life-threatening severe cutaneous adverse drug reactions (SCARs), characterized by high mortality and substantial morbidity risks. Identifying potential high-risk drugs associated with SJS/TEN is crucial for guiding clinical preventive interventions, enabling early detection, and enhancing risk management. With the rapid advancement of data science, adverse drug reaction (ADR) database mining has emerged as a powerful tool for systematically investigating the drug-SJS/TEN association, effectively overcoming the li

Furagin Derivatives-Nitrofuran-Based Hydrazones as Potential Dual Inhibitors of Thioredoxin Reductase and Carbonic Anhydrases.

Cancer remains one of the leading causes of mortality, accounting for millions of deaths worldwide each year and posing a persistent challenge to global health despite advances in diagnosis and treatment. Dual inhibition of thioredoxin reductase (TrxR) and cancer-associated carbonic anhydrase (CA) isoforms IX and XII represents a promising strategy for selective anticancer therapy by simultaneously disrupting cellular redox homeostasis and pH regulation. The antibacterial agent furagin has been previously explored as an inhibitor of cancer-associated carbonic anhydrases; however, its potential

Effect of carbonic anhydrase inhibitor on H+- and Cl--transients and epileptiform activity in brain slices of ClopHensor-expressing transgenic mice.

Alterations in H⁺ and Cl- concentrations are implicated in various neurological diseases, including epilepsy. Carbonic anhydrases (CAs) are crucial regulators of pH homeostasis, and certain CA inhibitors possess anticonvulsant properties. In this study, we investigated the effect of the CA inhibitor ethoxzolamide (EZA) on Cl- and H⁺ dynamics in hippocampal slices under control conditions and in an in vitro epilepsy model. We used transgenic mice expressing ClopHensor, a neuron-targeted sensor for simultaneous monitoring of Cl- and H⁺. Local field potential recordings and fluorescence imaging w

Sultams as Dual Inhibitors of Human Carbonic Anhydrase and Thioredoxin Reductase.

Cancer remains a leading cause of mortality, often complicated by drug resistance arising from single-target therapies. Dual inhibitors that simultaneously target multiple cancer-related defense mechanisms offer a promising strategy to overcome these limitations. Herein, we report the design, synthesis, and biological evaluation of a series of sultam-based N-acyl derivatives (8a-k and 9a-k) as potential dual inhibitors of thioredoxin reductase (TrxR1) and tumor-associated carbonic anhydrases (CA IX and XII). TrxR1 screening identified a few active compounds, with inhibitory potency strongly in

Carbonic anhydrase IX and XII.

The membrane-associated carbonic anhydrases (CAs) IX and XII have emerged as critical players in cancer biology due to their significant expression in various malignancies. The two enzymes differ in catalytic efficiency, with CA IX acting as a highly efficient catalyst while CA XII possesses moderate catalytic activity, and although both form a dimer, their resulting structural arrangement is diverse. Regarding their regulatory mechanisms, CA IX expression is primarily driven by the hypoxia-inducible factor 1 pathway in hypoxic microenvironments, whereas CA XII exhibits a more complex, context

Carbonic anhydrase XIV.

The carbonic anhydrase (CA) family comprises several isozymes with distinct cellular localizations and physiological functions. In mammals, sixteen different CA isozymes have been characterized to date, while additional isoforms have been identified in non-mammalian vertebrate species. Carbonic Anhydrase XIV (CA XIV) belongs to the membrane-associated subgroup of CAs. This chapter provides an overview of the biochemical and structural features of this enzyme, highlighting its functional properties and physiological significance. Particular attention is devoted to the mechanisms regulating its

Phenylsulfonylethyl-Substituted Benzimidazolium Salts: Synthesis, Characterization, Crystal Structure, Molecular Docking, and Inhibitory Properties Against Various Enzymes.

N-heterocyclic carbenes (NHCs) are chemical structures called heterocyclic structures that contain one or more nitrogen atoms. N-heterocyclic compounds are the basic building blocks of many drugs and biologically active molecules. One of these compounds is benzimidazolium salts. Herein, we present the synthesis of a series of phenylsulfonylethyl-substituted benzimidazolium salts. These compounds were characterized using NMR (1H- and 13C-) and FTIR spectroscopic techniques. Compound 1b exhibits a U-shaped benzimidazolium cation paired with a chloride anion. A hierarchical supramolecular archite

Drug-drug interaction signals between carbonic anhydrase inhibitors and vitamin D preparations in urinary tract stones: disproportionality analysis evaluation from Japanese spontaneous reports of adverse events.

BACKGROUND: Carbonic anhydrase inhibitors used as antiepileptic drugs promote calcium phosphate stones. Patients with epilepsy often receive vitamin D concurrently, which may increase urinary calcium and stone formation. We analyzed drug-drug interactions to determine if vitamin D contribute to renal and urinary tract stone disease associated with carbonic anhydrase inhibitors. METHODS: The Japanese Adverse Drug Event Report (JADER) was used to detect signals of the additive effects of vitamin D on carbonic anhydrase inhibitor-associated renal and urinary tract stone disease.
