Therapeutic advances in pruritus as a model of personalized medicine.

Pruritus or itch is a sensation that evolved to protect humans and other vertebrates against environmental irritants, parasitic infestations and disease-borne insects. However, in the setting of chronic skin inflammation and other medical conditions, pruritus can become pathologic and chronic in nature; indeed, chronic pruritus is defined as itch that persists for greater than 6 weeks and significantly impairs the quality of life of patients. It also poses a major therapeutic challenge. However, recent advances in itch biology have led to the development of novel therapies. This review highlig

Efficacy of Chi-Huang cream as an anti-inflammatory and anti-pruritic agent in treating chronic pruritus.

Pruritus is the main clinical symptom of skin diseases in animals and humans. Treatment outcomes from current conventional medicine are often unsatisfactory, since the same pruritus mediator may play multiple roles in different pruritus-associated diseases. Chi-Huang formula is the experiential traditional Chinese medicine formula for canine atopic dermatitis (CAD), and Chi-Huang solution for topical use has been shown to exhibit significant anti-inflammatory and anti-pruritic effects. This study aims to assess the anti-inflammatory and antipruritic effects of Chi-Huang cream in a squaric acid

Topical menthol mitigates pruritus and inhibits spinal microglial activation in mice.

ETHNOPHARMACOLOGICAL RELEVANCE: Mentha piperita L. (Lamiaceae), commonly known as peppermint, has a long history in traditional medicine. Its diverse essential oil composition, with menthol as a key bioactive component, supports a range of medicinal applications, including pruritus relief. This suggests a potential role in modulating itch via neuropharmacological mechanisms. AIM OF THE STUDY: This study aimed to investigate the anti-pruritic mechanisms of menthol by assessing its effects on scratching behavior and spinal microglial activation in a mouse model of pruritus. Research on the antipruritic active ingredients of Mikania micrantha.

Mikania micrantha is a perennial liana of the genus Mikania of the Asteraceae family. It is a commonly used medicine in South America for treating fever, malaria, dysentery, snake bites, etc. Because of its strong adaptability and ability to inhibit the growth of its associated plants, Mikania micrantha is considered an invasive species in China and is known as a plant killer. Preliminary studies have shown that Mikania micrantha has an antipruritic effect, but the antipruritic active substance is not yet clear. In this study, a 4-aminopyridine-induced itching model in mice was used to determi

Histamine- and pruritogen-induced itch is inhibited by a TRPM8 agonist: a randomized vehicle-controlled human trial.

BACKGROUND: Allergies often present challenges in managing itch and the effects of histamine. Cooling agents that act via transient receptor potential melastatin 8 (TRPM8) agonism have shown potential in itch management. However, animal studies on itch have limitations, as animals cannot communicate subjective events and their fur-coated skin differs from that of humans. Human studies offer more direct and reliable information. OBJECTIVES: To investigate the effects of a specific TRPM8 agonist gel (cryosim-1) on itch induced by various pruritogens in human skin. METHODS: Calcium imaging expe

Discovery of a Small Molecule Activator of Slack (Kcnt1) Potassium Channels That Significantly Reduces Scratching in Mouse Models of Histamine-Independent and Chronic Itch.

Various disorders are accompanied by histamine-independent itching, which is often resistant to the currently available therapies. Here, it is reported that the pharmacological activation of Slack (Kcnt1, KNa1.1), a potassium channel highly expressed in itch-sensitive sensory neurons, has therapeutic potential for the treatment of itching. Based on the Slack-activating antipsychotic drug, loxapine, a series of new derivatives with improved pharmacodynamic and pharmacokinetic profiles is designed that enables to validate Slack as a pharmacological target in vivo. One of these new Slack activato

Atopic Dermatitis: Managing the Itch.

Atopic dermatitis has a substantial impact on sleep, appearance, psychological well-being, and other qualities of life. The visual appearance of lichenification, cheilitis, hyperpigmentation, ichthyosis, and erythema can be socially stigmatizing, and treatment of these symptoms is challenging. In managing pruritus in patients, practitioners should assess and document pruritus through questionnaires at each routine visit. Initially, practitioners should advise patients to employ nonpharmaceutical treatments such as emollients with wet wraps, elimination of triggers, changing scratching habits,

Model of Chronic Itch in Aged Mice: Beneficial Effects of Drugs Affecting Descending Modulatory Systems.

Pruritus in the elderly, particularly those cases without skin dryness or other identifiable causes, makes treatment challenging due to the lack of evidence regarding the therapeutic effects of antipruritics. This study proposes an age-related alloknesis mouse model for an evaluation system for such cases, and aimed to investigate the effectiveness and mechanisms of action of several drugs commonly used as antipruritics in Japan, utilizing this model. Mice 69-80 weeks old were used as aged mice, and the level of mechanical alloknesis was counted as the number of scratching behaviours in respon

A Diterpenoid of the Medicinal Plant Andrographis paniculata Targets Cutaneous TRPV3 Channel and Relieves Itch.

Transient receptor potential vanilloid subtype 3 (TRPV3) is an ion channel implicated in skin physiology and itch. TRPV3 inhibitors can present a novel strategy for combating debilitating itch conditions, and medicinal plants are a natural pool of such compounds. Here, we report the isolation of a TRPV3-inhibiting compound from Andrographis paniculata, a medicinal plant with anti-inflammatory properties whose bioactive components are poorly characterized in terms of molecular targets. Using 1H and 13C NMR and high-resolution mass spectrometry, the compound was identified as a labdane-type dite

Lasmiditan ameliorates serotonergic itch in mice: Possible involvement of 5-HT1F receptors.

Previously, some allergic conditions involving pruritus have been linked to migraine, raising the possibility that migraine and itching may be governed by similar underlying mechanisms. We aimed to investigate the efficacy of Lasmiditan, a highly selective agonist of the 5-hydroxytryptamine 1F (5-HT1F) receptor and a recently approved medication for the treatment of migraine headaches, in ameliorating serotonergic itching. Forty animals were employed in the present study (n = 40). Eight animals were randomly assigned to each of the following study groups (n = 8, in each group): (1) "Normal Sal
