Connecting the dots between sleep deprivation and obesity: the role of melatonin in appetite regulation and insulin sensitivity from a mechanistic review perspective.

This review aims to clarify the mechanistic links between sleep deprivation, appetite regulation, and insulin sensitivity, and to explore the role of melatonin in these pathways. A systematic search identified 82 relevant studies, which were critically and thematically analysed. Evidence indicates that sleep deprivation lowers melatonin levels, which impairs insulin sensitivity via disruptions in the IRS/PI3K/AKT and cAMP/PKA/CREB pathways. Melatonin interventions appear to influence these pathways and affect leptin and appetite-regulating neuropeptides such as POMC, AgRP, and NPY, although fi

Cagrilintide-semaglutide (CagriSema) versus semaglutide or cagrilintide in people with type 2 diabetes (REIMAGINE 2): a double-blind, randomised, controlled, phase 3 study.

BACKGROUND: The amylin receptor agonist cagrilintide and the GLP-1 receptor agonist semaglutide have complementary effects on glycaemic control and bodyweight. We aimed to investigate the efficacy and safety of a fixed-dose combination of cagrilintide and semaglutide (cagrilintide-semaglutide; known as CagriSema) versus semaglutide or cagrilintide for glycaemic control in people with type 2 diabetes and overweight or obesity. METHODS: REIMAGINE 2 was a randomised, double-blind, placebo-controlled and active-controlled, parallel-group study conducted in 30 countries (trial sites included unive

IAPP surface-induced aggregation and corilagin's inhibitory effect.

Proteins and peptides spontaneously assemble into various structured supramolecular entities that perform vital physiological functions. In pathology and biopharmaceutics, protein assemblies can form cytotoxic oligomers or protofibrils. Most studies focus on aggregation and inhibition processes in bulk solutions. However, local surface interactions are largely responsible for protein structural changes leading to aggregation. To date, research on surface-induced protein aggregation remains insufficient to comprehend underlying mechanisms. This work reports a systematic process to produce islet

Safer solutions for sea: Montipora capricornis coral responses to naturally-derived versus copper-based antifouling agents.

While conventional synthetic antifouling agents (AFs) threaten coral reef ecosystems, the environmental safety profiles of naturally-derived AFs remain inadequately studied. To this end, a synthetic AF Cu2O, and 7 naturally-derived ones (7-Amino-4-methylcoumarin (AMC), butenolide, ε-polylysine (EPL), camptothecin (CPT), eugenol, gramine, and BAF-1) were chosen for a comparative study to reveal the physiological responses of coral Montipora capricornis. The selected AF concentration was 50 μg/L, referenced from Cu2O bleaching-inducing concentration. After 15 days, Cu2O demonstrated the most sub

Not only gut feelings: pancreatic hormone, amylin, controls emotionality and sociability, in a sex divergent manner.

Amylin, a pancreatic peptide hormone, has gained attention for its role in appetite regulation. Its analogues are approved for use in diabetic patients, and it is now under investigation as a potential anti-obesity therapeutic. However, despite its clinical use, its effects on emotionality remain poorly understood. Given that many other food intake-modulating substances influence psychiatric functions, here we determined the role of systemic amylin in modulating key aspects of emotionality and sociability in male and female rats. We further evaluated whether the central amygdala (CeA) is suffi

Enantioselective synthesis of antibiotic inducing gamma-butenolides from Streptomyces rochei and derivatives.

Given the challenges of discovering new natural products, further investigations into the regulatory systems that control their production are needed. Herein, we describe an enantioselective, divergent synthesis of two known autoregulators from Streptmyces rochei, the Streptomyces rochei butenolides (SRBs), which regulate production of the antibiotics lankacidin and lankamycin. The synthesis hinges on two key reactions: enantioselective alkynylation and regioselective photo-oxidation. The synthetic route also enabled access to nine derivatives, allowing for structure activity relationship stud

Amycretin in obesity: Mechanisms, clinical efficacy, and future perspectives.

The global escalation of obesity necessitates therapeutic interventions that transcend the efficacy ceilings of current mono-target pharmacotherapies. Amycretin, a novel unimolecular co-agonist targeting glucagon-like peptide-1 (GLP-1) and amylin receptors, has emerged as a promising candidate for weight management. In this review, we examine the developmental rationale of amycretin, elucidating how its dual-agonist mechanism synergistically engages hindbrain-mediated satiety pathways and delays gastric emptying to overcome metabolic plateaus. We summarize pivotal findings from recent clinical

Sibutramine high-doses effects in cafeteria diet-induced obese rats.

Obesity, defined as excessive fat accumulation that impairs health, is a global challenge with high mortality rates. Sibutramine, a centrally acting anorectic, was widely used in obesity treatment due to its effects on appetite control and weight loss. It represented a key step in anti-obesity pharmacotherapy before being withdrawn in many countries. This study evaluates how a high sibutramine dose (10 mg.kg.day-1 followed by 13 mg.kg.day-1) influences body weight, biochemical profiles, liver and intestinal health, and gut microbiota composition in rats subjected to a cafeteria diet for 16 wee

A fuel partitioning perspective on appetite suppression by GLP-1 receptor agonists.

Appetite suppression by glucagon-like peptide-1 receptor agonists (GLP-1RAs) is thought to result from direct actions on neural circuits controlling food intake. In this article, we (1) propose that a shift in fuel partitioning toward fat oxidation caused by GLP-1RAs contributes to appetite suppression and (2) explore potential lines of research to test this hypothesis.

Digital SERS-lateral flow immunoassay platform for ultrasensitive and dosage-sensitive analysis of sibutramine.

Rapid detection techniques play a critical role in the field of food hazard factor analysis, as food safety constitutes a fundamental cornerstone of public health. However, the application of Surface-Enhanced Raman Scattering (SERS)-lateral flow immunoassay (LFIA) as a rapid detection method in this area still faces significant challenges. Consequently, traditional quantification methods that rely on average Raman signal intensity often prove inadequate for accurate measurement under such conditions. In response to these limitations, we developed a digital SERS-LFIA approach, which employs dig

The duration of phentermine therapy in the private healthcare sector of South Africa: A retrospective analysis.

BACKGROUND: Phentermine is an amphetamine derivative and is globally the most used oral appetite suppressant for obesity. Its long-term (LT) use (a period >12 consecutive weeks) may be associated with amphetamine-like adverse effects. The US Food and Drug Administration has restricted its use to short term (ST) only (not longer than 12 consecutive weeks), specifically stated in the patient information leaflet. This serves as a prescribing guideline that is provided to assist both general and specialist practitioners in drug prescribing that is rational and safe, and to maximise patient outcome
