Procedures of data merging in precision cancer medicine: the PRIME-ROSE project.

BACKGROUND AND PURPOSE: As more interventional clinical trials in Precision Cancer Medicine (PCM) are introduced, molecular descriptions of tumours have led to multiple subtypes, even within common tumour types. Therefore, the main limitation of these trials is the small number of eligible patients to assess the clinical benefit. The PRIME-ROSE project addresses this limitation by pooling data from multiple European Drug Rediscovery Protocol (DRUP)-like clinical trials, such that slowly accruing cohorts are accelerated. To achieve this task, a well-documented commonly approved procedure for da

Targeting cancer stem cells: Dual-drug delivery systems and their role in cancer therapy.

Cancer stem cells (CSCs) represent a distinct subpopulation within tumors, intricately linked to tumorigenesis, progression, drug resistance, recurrence, and metastasis. Therefore, targeting CSCs is essential for achieving a definitive cure for cancer. The concomitant administration of two drugs, aimed at both tumor cells and CSCs, presents a promising strategy for the comprehensive eradication of tumors. However, ensuring the synergistic efficacy and targeted delivery of these dual drugs at the tumor site remains a significant challenge. This review begins by elucidating the biological charac

Discovery of benzimidazo-2-amino-1,3,4-thiadiazole carboxylate small-molecule STAT3 inhibitors for colorectal carcinoma therapy.

To identify potent candidate drugs for colorectal cancer (CRC), a series of N-benzimidazole-1,3,4-thiadiazole-2-amine derivatives featuring diverse ester groups were designed and synthesized, based on our preceding research. Systematic pharmacological evaluation demonstrated their inhibitory effects against the IL-6/JAK/STAT3 signaling pathway and CRC cell lines with constitutively activated STAT3. Among these derivatives, compound L20 demonstrated the most potent anti-proliferative activity against HCT116 (IC50 = 0.45 ± 0.05 μM) and other CRC-relevant cell lines. Mechanistic investigations co

Inhalable stealth liposomes improve peptide delivery for pulmonary fibrosis treatment.

Pulmonary fibrosis is a life-threatening lung disease with limited therapeutic options and suboptimal clinical outcomes. Peptide-based agents such as CSP7, a heptapeptide derived from caveolin-1, have shown promising antifibrotic activity with low systemic toxicity. However, effective inhaled administration of CSP7 is impeded by physiological barriers in fibrotic lungs, including alveolar edema fluid and clearance by inflammatory macrophages. Here, we report a high-loading stealth liposomal system designed to enhance pulmonary delivery of CSP7 by inhalation. CSP7 was covalently conjugated to a

Design and Synthesis of Marine Sarocladione Derivatives with Potential Anticancer Activity.

The discovery of structurally novel anti-tumor agents remains a crucial objective in cancer drug research. In this study, we systematically explored the bioactivity potential of sarocladione (5), a structurally unique marine-derived 14-membered ring diketone steroid. Guided by a function-oriented strategy, seven new derivatives (6-13) were synthesized based on an efficient biomimetic synthesis of sarocladione. Evaluation of their antiproliferative activities against human cancer cell lines demonstrated that the intact macrocyclic scaffold is indispensable for activity. Extension of the conjuga

Multidimensional chromatographic fingerprint fusion with machine learning: Entropy-based feature evaluation for TCM quality marker discovery.

Chromatography is a cornerstone methodology employed for quality evaluation of traditional Chinese medicine (TCM). However, the complexity of chromatographic data, where signals from multiple compounds overlap and interfere, often impedes the accurate identification of chemically significant features. This study proposed an integrated approach that combines multidimensional chromatographic fingerprinting with machine learning to trace the molecular origins of characteristic compounds in a representative TCM formula, Fufang E'jiao Jiang (FEJ). Following comprehensive chemical profiling, we cons

A Reproducible Workflow for Macrophage Membrane Isolation and Nanocore Selection Toward Bioinspired Nanoparticles Targeting Immunologically Cold Tumors.

Macrophage membrane-coated nanoparticles (M2M-NPs) offer a promising strategy for targeting immunologically "cold" tumors resistant to conventional therapies. However, workflows for membrane isolation and NP coating remain limited and often lack reproducibility. Herein, a multi-step process was established for isolating plasma membranes from human M2-like macrophages. This optimized workflow combines hypotonic lysis, Dounce homogenization, and differential centrifugation, yielding M2M fractions with consistent protein, lipid, and DNA profiles. Building on this process, a detailed and reproduci

Balancing immune evasion and hepatocyte delivery: a hybrid liposomal platform for enhanced nanotherapy in non-alcoholic fatty liver disease.

The clinical translation of nanomedicine for non-alcoholic fatty liver disease (NAFLD) is frequently hindered by rapid hepatic clearance mediated by Kupffer cells, which are the predominant phagocytic macrophages in the liver. Conventional stealth strategies, such as PEGylation, protect host cells from phagocytosis; however, their exploitation in drug delivery is limited by the prevailing assumption that immune evasion impairs cellular uptake. Our previous study demonstrated that liposomes functionalized with a CD47-derived self-peptide (SLip) effectively evaded macrophage clearance while reta

Construction of biomimetic nanomedicine delivery system based on biomedical materials for treating brain diseases: A review.

Brain diseases are often characterized by a high mortality rate and high treatment difficulty, posing significant challenges to human healthcare. However, due to the presence of the blood-brain barrier (BBB), which separates brain blood vessels from brain tissue, the delivery efficacy of traditional delivery systems is limited. This makes it difficult to deliver drugs effectively to the affected areas, thereby severely limiting their therapeutic efficacy in treating these brain diseases. Recently, biomimetic drug delivery systems, particularly those based on biomaterials, have revolutionized t

Microwave plasma torch mass spectrometry for rapid prediction of photo-thermal stability and degradation pathways of pharmaceuticals.

Photo-thermal stability is a key factor in pharmaceutical development, as light and heat can compromise drug safety and efficacy. Conventional methods such as HPLC, DSC, and GC-MS are slow, require complex pretreatment, and often treat photolysis and thermolysis separately. Here, we demonstrate microwave plasma torch mass spectrometry (MPT-MS) as a rapid, pretreatment-free approach for simultaneous analysis of photo-thermal stability and degradation pathways. Using direct desorption/ionization of intact tablets, six pharmaceuticals: chlorpheniramine, ornidazole, metronidazole, tinidazole, acet
