Development of a quality control test for diphtheria vaccine based on an electrochemical aptamer-based biosensor.

At every stage of the vaccine's development, including research and development together with the routine production (in-process control of batch release), quality control testing is a crucial operation for ensuring the efficacy, safety and consistency of final products. Conventional quality control tests rely on a combination of biochemical, physicochemical, and cell-based assays. In this study, we report a quality control test based on an electrochemical aptamer-based sensor with a specific application to Diphtheria vaccine. First, specific aptamers for Diphtheria are successfully selected a

Measuring the long-term impact of the COVID-19 pandemic on HPV vaccination in Quebec, Canada: A cohort study from the Canadian immunization research network.

OBJECTIVES: School-based vaccine programs in Quebec experienced significant disruptions due to the COVID-19 pandemic. Some vaccines offered in schools, such as the human papillomavirus (HPV) vaccine, continue to fall short of the 90% vaccine coverage (VC) target for the routine program. This study aims to assess temporal changes in VC for HPV, compare trends with other vaccines delivered in schools, and examine socioeconomic disparities in HPV VC. METHODS: We conducted a retrospective cohort study using data from the Quebec Immunization Registry linked with the Quebec Ministry of Education da

From School-Based Immunization to Family Health Centers: the Impact of a 2020 Policy Change on 13-Year-Old Tetanus-Diphtheria Vaccine Coverage in Turkey.

This study evaluated the impact of transferring the thirteen‑year tetanus-diphtheria booster from schools to Family Health Centers on vaccination coverage after 1 July 2020. In a two‑component descriptive cross‑sectional design conducted in the central districts of Şanlıurfa Province, annual non‑vaccination from 2018 to 2022 was tracked using administrative records; in addition, reasons for non‑vaccination were classified through face‑to‑face interviews with family physicians and on‑screen verification in nineteen Family Medicine Units selected by two‑stage stratified sampling. The primary out

Enhancing Adolescent Vaccine Acceptance: A Multi-Component Intervention Study on 13-Year-Old Tetanus-Diphtheria Vaccination.

OBJECTIVE: This study aimed to evaluate the impact of Turkey's 2020 policy shift from school-based to family physician-based individual vaccination for the adolescent Tetanus-Diphtheria (Td) vaccine and to assess the effectiveness of a multi-component intervention (education, reminders, and on-site vaccination) in improving vaccine uptake among initially unvaccinated adolescents. METHODS: The study consisted of two phases: first, a cross-sectional analysis comparing Td vaccination coverage rates from 2018 to 2024 before and after the policy change; second, a quasi-experimental intervention in

Severe rhabdomyolysis and refractory dysautonomia complicating generalized tetanus in an unvaccinated child: A case report.

Tetanus has become rare in settings with established immunization programs; however, severe cases continue to pose complex management challenges, particularly in children. We report a 12-year-old unvaccinated boy from an Indigenous community in southwestern Colombia who developed generalized tetanus seven days after a puncture wound, complicated by early severe rhabdomyolysis (creatine phosphokinase peak >32,000 U/L), acute kidney injury, and refractory autonomic instability. While rhabdomyolysis may occur in severe tetanus, its coexistence with marked dysautonomia created a therapeutic dilemm

Two decades of global research on tetanus vaccines and tetanus immunoglobulins: a comprehensive bibliometric analysis and integrative review (2000-2025).

BACKGROUND: Tetanus is a severe but vaccine-preventable disease controlled by tetanus toxoid (TT) immunization and tetanus immunoglobulin (TIG) therapy. Although maternal-neonatal tetanus has been eliminated in many regions, coverage gaps and sporadic cases persist in low-resource settings. As a longstanding component of global immunization and a key conjugate carrier platform, tetanus research remains essential despite advances in novel vaccine technologies. However, the global research landscape on tetanus vaccines and immunoglobulins remains unmapped. OBJECTIVE: This study aimed to conduct

Hepatitis B Virus, Pneumococcal, Diphtheria, and Tetanus Vaccination Responses in Compensated Cirrhosis.

BACKGROUND AND AIMS: Cirrhosis favours infections that can lead to liver decompensation and death. Some of these infections can be prevented by vaccination. This study aimed to evaluate the immune response after HBV, pneumococcal, diphtheria, and tetanus vaccination in cirrhotic patients. METHODS: Patients with cirrhosis were prescribed 3 doses of hepatitis B vaccine, a tetanus-diphtheria booster and/or a 13-valent pneumococcal conjugate vaccine followed by a 23-valent polysaccharide vaccine. HBs seroconversion was assessed 6 months after the last booster dose. Antibody concentrations for sev

Heterogeneous returns to antenatal care: Evidence from childhood immunization in Senegal.

Antenatal care (ANC) serves both as a medical input and as a source of health information in the child health production function. We use seven waves of the Senegal Demographic and Health Surveys, matched to regional health-facility data, to estimate the effect of ANC adherence on childhood vaccination outcomes. We address endogeneity arising from unobserved heterogeneity in ANC utilization using a two-stage copula-based control-function approach. We find that ANC adherence has no effect on the composite measure of full immunization, but significantly increases uptake of two injectable vaccine

Rare Survival of a 9-Year-Old Boy Managed for Post-Neonatal Tetanus Without Intensive Care at Soroti Regional Referral Hospital, Uganda: A Case Report.

BACKGROUND: Tetanus is a vaccine-preventable disease caused by neurotoxin from Clostridium tetani bacteria. Although neonatal tetanus has declined in many settings, post-neonatal tetanus continues to occur in low-resource countries where vaccination coverage and booster implementation remain suboptimal. Mortality in post-neonatal tetanus remains high, particularly where intensive care services are limited. CASE PRESENTATION: We report a 9-year-old African boy who presented three days after a minor toe injury with generalized painful muscular spasms, trismus, and hypertonia consistent with gen

Automated differentiation of parkinsonian disorders: an ROI-based analysis of subcortical shape and cortical surface features.

UNLABELLED: The clinical manifestations of early-stage parkinsonian syndromes overlap, making accurate differential diagnosis crucial yet challenging. This study aimed to develop a system for automated differentiation of idiopathic Parkinson's disease (IPD) from progressive supranuclear palsy (PSP) and corticobasal syndrome (CBS). Our sample included clinical data and T1-weighted magnetic resonance imaging from 50 IPD, 47 PSP, and 38 CBS patients. We introduced an atlas-based approach to extract shape features from subcortical regions in each subject's native coordinate image space. The surfac

Exploring the ethics of tuberculosis human challenge models.

We extend recent conversation about the ethics of human challenge trials to tuberculosis (TB). TB challenge studies could accelerate vaccine development, but ethical concerns regarding risks to trial participants and third parties have been a limiting factor. We analyse the expected social value and risks of different challenge models, concluding that if a TB challenge trial has between a 10% and a 50% chance of leading to the authorisation and near-universal delivery of a more effective vaccine 3-5 years earlier, then the trial would save between 26 400 and 1 100 000 lives over the next 10 ye

Enhanced immunogenicity of Staphylococcus aureus IsdE vaccine with SeNPs and BCG adjuvants in mice.

Staphylococcus aureus is a major cause of nosocomial infections and antimicrobial resistance, emphasizing the urgent need for an effective vaccine. The IsdE protein, a heme-iron transporter, has been identified as a promising antigen target. In this study, we evaluated the immunogenicity and protective efficacy of recombinant IsdE (r-IsdE) formulated with the MF59 adjuvant and co-adjuvanted with Bacillus Calmette–Guérin (BCG) and synthetic SeNPs (SeNPs) in BALB/c mice. Mice were immunized three times at two-week intervals, and immune responses were analyzed through cytokine assays, antibody is

Integrative profiling of transcriptome and H3K27ac modification reveals changes associated with BCG-induced trained immunity in bovine immune cells.

We previously demonstrated that Bacillus Calmette Guérin (BCG) vaccination induces trained immunity in bovine monocytes and gamma delta (γδ) T cells, enhancing IL-1β and IL-6 production. To define the underlying molecular mechanisms, we applied a multi-omic approach to circulating monocytes and alveolar macrophages (amac). Twenty Holstein-Angus heifers were randomly separated into two groups and immunized subcutaneously with 6 million CFUs of BCG Danish strain (n = 10), or saline (n = 10), and boosted after 2 weeks. Monocytes (4 weeks post-boost) and amac (6 weeks post-boost) were analyzed for

Inflammatory models of depression in rodents and humans.

The precise pathophysiological mechanisms underlying major depressive disorder (MDD) remain poorly understood. Substantial evidence implicates immune-mediated mechanisms in the pathogenesis of this clinically heterogeneous and multifactorial disease. This review provides a comprehensive synthesis of current knowledge regarding the association between inflammation and depression, critically evaluates established approaches for modeling inflammation-induced depressive states in both rodents and humans, and assesses these models against standard validity criteria. The empirical link between depre

Malathion-induced neurological alterations associated with dysregulation of RORγt/STAT3/IL-17 and FOXP3/STAT5/IL-10 pathways: potential neuroprotective effects of BCG.

Despite their effectiveness in agriculture to control a variety of pests, organophosphorus compounds (OPC) such as malathion were linked with neurological dysfunctions and possibly death. The present study aimed to investigate the impact of OPC exposure on neuroinflammation via disrupting the equilibrium between pro-inflammatory (RORγt/STAT3/IL-17/IL-22) and anti-inflammatory (FOXP3/STAT5/IL-10) pathways. The study extended to evaluate the potential of BCG vaccination in alleviating neuroinflammation. Rats were distributed into four groups: control, malathion-intoxicated group, BCG-treated gro

A randomized prospective study of neonatal hepatitis B vaccine immunogenicity in The Gambia and Papua New Guinea.

BACKGROUND: Protection of newborns from infection can be achieved through maternal or vaccine-induced antibodies, but the factors influencing vaccine protection (correlate of protection) and subsequent infant immunity remain insufficiently understood. Further investigation is essential to optimize early-life vaccination strategies. OBJECTIVE: We sought to evaluate the impact of timing and sequence of hepatitis B vaccine (HBV) and Bacille Calmette-Guérin (BCG) vaccine within the first week of life on infant HBV immunogenicity and its relationship with maternal antibodies. METHODS: This was an

Engineered BCG selectively triggers trained immunity in tumor-associated macrophages and sensitizes glioblastoma to radiotherapy in mice.

Radiotherapy plays a crucial role in antitumor immunity in glioblastoma, yet its efficacy is often limited, resulting in tumor recurrence. Here, we engineer a macrophage membrane-camouflaged Bacillus Calmette-Guérin (BCG) via bioorthogonal chemistry to enhance radiotherapy against glioblastoma. This engineered BCG penetrates the blood-brain barrier, targets tumors, and alleviates hypoxia through intrinsic catalase activity, exerting antitumor effects in both murine orthotopic glioblastoma and humanized mouse models. Notably, it also initiates trained immunity in tumor-associated macrophages. D
