With the success of viral-vectored vaccines against Ebola and SARS-CoV-2, these platforms are now established as promising tools for biodefence against outbreak pathogens. Here, we report the development of adenoviral-vectored vaccines against plague, caused by Yersinia pestis. Replication-deficient adenoviral-vectored constructs expressing the protective Y. pestis antigens F1 and LcrV, alone or in combi and combinations thereof were created. Expression of the antigens was confirmed in vitro, and immunogenicity assessed in mice. Protective efficacy was evaluated using a virulent Y. pestis aero

We evaluated the association of alleles of human leukocyte antigen (HLA) class II genes and the levels of secretion of IL-4 and IFNγ in the blood of individuals vaccinated/revaccinated with a live plague vaccine and living in the Gorno-Altai high-mountain and Tuvinian mountain natural plague foci. Significant differences were identified in the blood content of IL-4 and IFNγ in volunteers with different variants of alleles in the HLA-DRB1, HLA-DQA1, and HLA-DQB1 loci, which indicates a substantial influence of their polymorphism on the immunological reactivity during specific plague prevention.

BACKGROUND: The COVID-19 pandemic led to unprecedented interest and participation in vaccine trials globally, and a concurrent increase in vaccine hesitancy. Whether this impacted recruitment of healthy volunteers to subsequent non-COVID vaccine trials is not well studied. We explored the impact of the COVID-19 pandemic on motivations for participating in two clinical trials of the same novel anti-plague vaccine, conducted in the United Kingdom (UK) and Uganda in 2021 and 2022.
METHODS: Participants enrolled in PlaVac (UK) and PlaVac Uganda, Phase I trials of ChAdOx1 Plague vaccine, were invi

BACKGROUND: Plague is a severe zoonotic disease caused by Yersinia pestis, a pathogen characterized by high infectivity and mortality rates. Historically, three global pandemics have inflicted heavy disasters on human society. Despite improvements in control measures and the application of antibiotics, plague has been somewhat controlled; however, since the beginning of the 21st century, plague outbreaks have continued to occur in regions with a high burden of neglected tropical diseases (NTDs) in Africa, the Americas, and Asia. In recent years, the rapid development of technologies such as mo

Pneumonic plague remains a high-consequence respiratory disease, highlighting the need for vaccines that induce durable lung-localized immunity. We show that intramuscular prime-boost vaccination with Alum-absorbed OMV46-LcrV (OMV46-LcrV/Alum) is safe but provides limited long-term protection against respiratory Yersinia pestis challenge in mice. To enhance lung immunity, we deliver an adjuvant-free intranasal LcrV "spike" following IM OMV46-LcrV/Alum priming. This "prime-spike" regimen is well tolerated and confers complete short- and long-term protection against high-dose pulmonary Y. pestis

BACKGROUND: Yersinia pestis, a category A infectious organism, is known to cause bubonic, septicemic, and pneumonic plague. With the emergence of streptomycin-resistant strains, there is an urgent need for new therapeutic strategies that can protect populations from Y. pestis infection. The main strategy for developing vaccines and therapeutic antibodies involves F1 antigen. Previous research has demonstrated that both human and murine monoclonal antibodies(mAbs) confer protective effects against Y. pestis infection. While, no relevant studies were identified on mAbs against drug-resistance Y.

Two live-attenuated vaccines, LMA and LMP, were evaluated alone or in combination with a trivalent adenoviral vector-based vaccine (Ad5-YFV) for their protective efficacy against pneumonic plague in wild-type (WT) and interferon-γ (IFN-γ) knockout (KO) mice. LMA and LMP comprise triple deletion mutants of Yersinia pestis CO92, which causes pneumonic plague, and Ad5-YFV incorporates three protective plague immunogens. Protection of 80 to 100% was observed in vaccinated mice when challenged with highly lethal intranasal doses of parental Y. pestis CO92. All vaccinated mice generated robust humor

The Journal of Genetics, started by William Bateson in 1910, played a distinguished role in the early years of genetics. However, it stopped publishing in 1978. The Indian Academy of Sciences revived it in 1985, and has published it regularly since then. To commemorate this landmark, I highlight one of the 17 articles published that year. 'The isolation and genetic analysis of a Caenorhabditis elegans… X-chromosome balancer' by András Fodor and Péter Deak, of the Institute of Genetics, Biological Research Centre, Hungarian Academy of Sciences, Szeged, Hungary. During the last 100 years, vaccine development has evolved from an empirical approach to one of the more rational vaccine designs where the careful selection of antigens and adjuvants is key to the desired efficacy for challenging pathogens and/or challenging populations. To improve immunogenicity while maintaining a favorable reactogenicity and safety profile, modern vaccine design must consider factors beyond the choice of target antigen alone. With new vaccine technologies currently emerging, it will be possible to custom-design vaccines for optimal efficacy in groups of people with differe

Yersinia pestis, the causative agent of plague, remains a threat to public health worldwide. From the perspective of developing safe and effective vaccines, we present a derived version of our Y. pseudotuberculosis VTnF1 live attenuated vaccine candidate that lacks the pYV virulence plasmid coding for the Type 3 Secretion system (T3SS) and carries no antibiotic resistance cassettes (VTnF1-S). This strain, named VpYV-, fails to cause disease in immunocompromised mice when given orally, and can be considered as avirulent in such conditions. It retains a tropism for Peyer's patches and mesenteric

The importance of rapidly developing and distributing safe and effective vaccines was a major lesson from the COVID-19 pandemic, which drove new vaccine development technologies. In May 2024, the World Health Organization (WHO) updated the identification of bacterial pathogens of importance to public health as guidance for research and development of strategies to prevent and overcome global antimicrobial resistance (AMR). In June 2024, the WHO updated the 2017-18 list of priority pathogens to provide a scientific framework for epidemic and pandemic preparedness. The 2024 updated WHO list of p

The aim of this article is to complicate and rethink histories of non-Western responses to vaccination, as well as to see how a historical perspective may contribute to contemporary discussions of vaccines within the medical humanities. This is done through a case study approach. The focus of this article is a collection of essays written by a Bengali prophet-astrologer and published during the plague pandemic in Calcutta, British India, at the turn of the twentieth century (1899). While one essay in the collection was critical of the plague vaccine, another essay, in a later section of the sa

OBJECTIVE: Plague (Yersinia pestis infection) is a flea-borne zoonotic disease mainly affecting African countries, with most human cases reported from Madagascar and the Democratic Republic of Congo. Although curable, plague can be fatal without prompt treatment, highlighting the importance of reliable diagnostics. Current tests include bacteriological culture, PCR, and anti-F1 ELISA, however the anti-F1 serology has limitations due to the existence of F1-negative virulent strains. To address this, we developed a serological test detecting IgG antibodies against LcrV or V antigen, the main vir

Yersinia pestis is the causative agent of plague - the archetypal bacterial pandemic disease. Plague remains endemic in several countries in Africa, South America, and Asia, posing high risks of zoonotic spill-over and epidemic spread or threat of deliberate release. Plague vaccine development remains a priority for pandemic preparedness initiatives but generating sufficient field data for vaccine licensure is challenging. Controlled human infection studies have been deployed to test candidate vaccines against diseases with low and sporadic incidences of outbreaks where field trials are diffic

Duck plague virus (DPV) is the causative agent of duck plague (DP), which poses a significant threat to production safety in the duck industry. Little is known about the UL24 protein (pUL24) encoded by the DPV UL24 gene. In this study, pUL24, when transfected into duck embryo fibroblasts, localized to the nucleus, as demonstrated by immunofluorescence techniques. We discovered that UL24 contains two nuclear localization signals (NLS), with the first also serving as a nucleolar localization signal (NoLS). To further analyse the function of pUL24, two viral strains were constructed: a UL24 delet

Yersinia pestis caused the three plague pandemics that claimed more than two hundred million human lives. There is still no vaccine that meets all WHO requirements, and many researchers continue to develop plague vaccines using various technological platforms. For example, researchers led by Roy Curtiss 3rd have developed a new approach to achieve controlled, delayed attenuation of bacterial pathogens. Mutants generated using this method were superior in protecting Y. pestis-infected mice immunized with strains generated using traditional gene knockout. However, further studies are needed to d

Yersinia pestis, the causative agent of plague, remains a significant global health hazard and a potential top-tier biothreat despite modern medical advances. Here, two mRNA constructs encoding different versions of the low-calcium response virulence (LcrV) protective antigen, an essential virulence factor of Y. pestis, are designed and evaluated. Next, the immunogenicity and protective efficacy both independently and in combination is assessed with the previously reported F1-encoding mRNA construct in the well-established mouse model of pneumonic plague. The findings reveal that human Fc-conj

Yersinia pestis, the causative agent of plague, is classified as a Tier I Select Agent of bioterrorism and is among a few pathogens of high concern as a potential cause of a future pandemic. Currently, there is no licensed vaccine against plague. Previously, we developed a live attenuated vaccine candidate, rLVS ΔcapB/Yp F1-V, that utilizes a highly attenuated capB mutant of Francisella tularensis Live Vaccine Strain as a vector to express a fusion protein of Y. pestis F1 and LcrV antigens. We showed that homologous prime-boost vaccination with this vaccine provided potent protection in mice a

Background/Objectives:Yersinia pestis is an important zoonotic pathogen responsible for the rare but deadly disease of people with bubonic, septic, or pneumonic forms of plague. The emergence of multidrug-resistant Y. pestis strains has attracted more and more researchers' attention to the search for molecular targets for antivirulence therapy, including anti-nutritional-virulence therapy. The glnALG operon plays a crucial role in regulating the nitrogen content within a bacterial cell. This operon codes for three genes: the structural gene glnA and the two regulatory genes glnL and glnG. In t

INTRODUCTION: Yersinia pestis is the etiological agent of plague, a disease that remains a concern as demonstrated by recent outbreaks in Madagascar. Infection with Y. pestis results in a rapidly progressing illness that can only be successfully treated with antibiotics given shortly after symptom onset. Live attenuated or whole cell inactivated vaccines confer protection against bubonic plague, but pneumonic plague has been more difficult to prevent. Novel effective subunit vaccine formulations may circumvent some of these shortfalls. Here, we compare the immunogenicity generated by an advanc
