A Modular Biomimetic Nanovaccine for TBEV: Orchestrating Cascade Immunoprotection via Programmed Dendritic Cell Targeting and Antigen-Adjuvant Co-delivery.

Tick-borne encephalitis (TBE) is an escalating global health threat, increasingly exacerbated by climate change, and underscores the need for innovative vaccine strategies. Conventional subunit vaccines are limited by suboptimal spatiotemporal dissociation between antigens and adjuvants, as well as insufficient specific targeting, resulting in compromised immunogenicity. Herein, we report a biomimetic nanovaccine platform engineered to overcome these limitations through the synergistic co-delivery of antigen and adjuvant. This platform is constructed by co-displaying the TBE virus envelope pro

Enhancing foot-and-mouth disease virus thermostability with a VP1 substitution selected by host HSP60 inhibition.

Foot-and-mouth disease virus (FMDV) exhibits greater temperature sensitivity compared to many other picornaviruses, which poses a substantial challenge for the stability of inactivated vaccines. Therefore, enhancing the thermal stability of the FMDV capsid is crucial. FMDV exists as a quasispecies with high mutation rates, providing a basis for selection under pressure. Our group previously discovered that heat shock protein HSP60 was recruited to support FMDV replication by promoting the assembly of the viral replication complex (RC). By applying the HSP60 inhibitor mizoribine, we selected FM

Efficacy and economic evaluation of a novel genotype VII marker vaccine for Newcastle disease in layers.

Newcastle disease (ND) poses a serious threat to poultry, with vaccination remaining the most effective control strategy. However, the widely used ND vaccine strain exhibits significant antigenic mismatch with prevalent genotype VII Newcastle disease virus (NDV), necessitating increased vaccination dosages and frequencies. This study evaluated optimization of immunization schedules for the ND marker vaccine based on genotype VII MG7 strain. Three groups were tested: Group A (4 vaccinations with inactivated ND marker vaccine), Group B (5 vaccinations with combined inactivated and live ND marker

Immunoinformatics and reverse vaccinology design of self-amplifying RNA vaccine against marburg virus with structural dynamics and immune simulation analysis.

Marburg virus disease (MVD), a deadly zoonotic illness in humans, is caused by the Marburg virus (MARV), a single-stranded RNA virus belonging to the Orthomarburgvirus genus of the Filoviridae family. There are currently no approved vaccines or efficient antiviral therapies for MARV, despite high morbidity and fatality rates of the virus, particularly in Africa. Recognizing its epidemic potential and its inclusion on the WHO list of blueprint priority pathogens, an immunoinformatics approach was used to design novel multi-epitope saRNA vaccine candidates (MB-saVaxI and MB-saVaxII) targeting th

Hantaviruses as Emerging Zoonotic Pathogens: Molecular Biology, Epidemiology, Therapeutics, vaccine, and Pathogenesis.

Hantaviruses are globally distributed zoonotic pathogens belonging to the family Hantaviridae and are responsible for two major human diseases, hemorrhagic fever with renal syndrome (HFRS) and hantavirus cardiopulmonary syndrome (HCPS). Despite substantial progress in hantavirus research, important challenges remain in understanding the interactions among viral evolution, reservoir ecology, environmental change, transmission dynamics, and host immunopathology. This review provides an integrated One Health perspective that connects molecular virology and host-virus interactions with transmissio

A subunit vaccine protects A129 mice from death and joint swelling against lethal Chikungunya virus challenge.

INTRODUCTION: Chikungunya virus (CHIKV) is primarily transmitted to humans through bites of infected Aedes mosquitoes, causing febrile illness as well as acute or chronic arthritis. With increasing reports in previously non-endemic regions, it poses an emerging threat to public health and a growing risk to travelers. Despite the recent approval of live-attenuated and virus-like particle-based vaccines, a safe, efficacious, and cost-effective vaccine remains urgently needed. The envelope glycoproteins E1 and E2, which play crucial roles in viral attachment and entry, are the major immunodominan

Integrated immunoreactivity analysis of MHC-I epitopes derived from the Marburg virus glycoprotein.

INTRODUCTION: Marburg virus (MARV) is a hemorrhagic fever virus, and its glycoprotein (GP) represents an important target for vaccine research. MHC class I (MHC-I)-restricted T cell epitopes are critical for inducing cytotoxic CD8+ T cell responses. In this study, we aimed to identify MHC-I-restricted T cell epitopes derived from the MARV Angola strain GP through an integrated bioinformatics and experimental approach. METHODS: We retrieved the MARV Angola strain GP sequence and divided it into 673 overlapping 9-mer peptides. By integrating five computational prediction algorithms, immunogenic

Transcription Regulatory Sequences in Coronaviruses: Mechanistic Foundations for Antivirals and Vaccines.

Coronaviruses employ a distinctive discontinuous transcription mechanism to generate a nested set of subgenomic mRNAs (sgRNAs) from a single large positive-strand RNA genome. Transcription regulatory sequences (TRSs)-comprising a leader TRS (TRS-L) near the 5' end and multiple body TRSs (TRS-Bs) upstream of downstream open reading frames-serve as the primary cis-acting signals that direct polymerase pausing and template switching during negative-strand synthesis. The COVID-19 pandemic spurred intensive investigation of SARS-CoV-2 gene expression, yielding high-resolution transcriptome maps, ge

Discovery of KC-1: a novel porcine dendritic cell-targeting peptide with potential applications in swine vaccine design.

Dendritic cells (DCs) are key antigen-presenting cells essential for initiating and regulating immune responses. While DC targeting has proven to be an effective strategy for vaccine enhancement, and targeting peptides have been extensively utilized as efficient delivery tools in DC-targeted drug and vaccine development, there remains a notable scarcity of peptides specifically selected through porcine dendritic cell screening platforms. In this study, phage display biopanning was employed to identify a novel DC-targeting peptide, designated KC (KCCYPNQMAAFA). Systematic alanine-scanning mutag

Recombinant Antigen for Newcastle Disease Virus Vaccine Focusing on Currently Circulating Virus Variants.

Newcastle disease virus (NDV) causes a highly contagious avian infection and is characterised by its high genetic diversity and mutation rate. Currently applied vaccines are based on NDV genotypes I and II, which are considerably distinct from the circulating pathogenic genotypes, of which genotype VII is predominant, and therefore, they may not provide complete protection for birds or stop virus transmission. This work is a proof-of-concept study aiming to develop a genotype VII-focused recombinant antigen for a vaccine targeting relevant NDV strains. The research primarily focuses on novel a

Viral Respiratory Infections: Upper and Lower Respiratory Tract Infections.

Respiratory infections are a major health concern and are divided into upper and lower respiratory tract infections. Upper respiratory tract infections are mostly viral and self-limiting; billions of cases occur globally each year. Lower respiratory tract infections such as pneumonia and bronchitis can cause significant illness, especially in children, older adults, and individuals with comorbidities or immunocompromise. In 2021, lower respiratory tract infections caused more than 2 million deaths globally, mainly in low- and middle-income countries. Preventive measures include hand hygiene, m

Designing a multi-epitope vaccine against La Crosse virus using immunoinformatics approach.

La Crosse virus (LACV) is a mosquito-borne Orthobunyavirus responsible for pediatric encephalitis, with no currently approved vaccine or targeted antiviral therapy. This study aimed to design a multi-epitope subunit vaccine targeting the conserved nucleoprotein of LACV using an integrated immunoinformatics approach. The nucleoprotein exhibited strong antigenicity and favorable physicochemical properties, including stability and hydrophilicity. Highly conserved CTL, HTL, and B-cell epitopes were initially predicted, from which high-affinity, non-allergenic, and non-toxic epitopes were selected.

Fusion-loop-engineered Zika mRNA vaccine elicits convergent B cell immunodominance to uncouple protection from ADE in macaques.

Antibody-dependent enhancement (ADE) remains a major hurdle for flavivirus vaccine development. Engineering the fusion loop (FL) represents a promising solution, yet validations restricted to murine models limit human translatability due to profound species divergence. Here, we provide a primate-level demonstration that FL-engineered Zika virus (ZIKV)-M/E mRNA vaccination protects against ZIKV challenge with negligible dengue virus (DENV) ADE. The vaccine-induced B cells are overwhelmingly ZIKV specific (>97%), dominated by a convergent population using a germline ortholog of human IGHV5-10-1,

Early classical swine fever virus persistence is associated with cytotoxic and immunoregulatory transcriptional signatures distinct from protective C-strain vaccination.

Persistent classical swine fever virus (CSFV) infection represents a major challenge for disease control because persistently infected pigs remain clinically healthy while continuously shedding virus. However, the early immunological events associated with the establishment of viral persistence remain incompletely understood and require further investigation. In this study, we compared the peripheral blood mononuclear cell (PBMC) transcriptomes of neonatal piglets infected with the moderately virulent Catalonia01 CSFV strain, previously shown to induce postnatal persistent infection, with thos

An immunoinformatics-driven multiepitope subunit vaccine targeting Oropouche virus: Molecular docking, immune and dynamics simulation approaches.

The Oropouche virus (OROV) is an emerging zoonotic orthobunyavirus responsible for recurring outbreaks of febrile illness and neurological disorders in South and Central America. Despite its growing public health issues, no FDA-approved vaccines or antiviral therapies currently exist. To address this gap, a multiepitope subunit vaccine was designed to target OROV glycoproteins G1 (Gn), G2 (Gc), and the nucleocapsid (N) protein using a reverse-vaccinology (RV) approach. For vaccine construction, suitable epitopes (MHC-I, MHC-II, and linear B-cell) were selected based on percentile rank, antigen

Concurrent administration of a combined clostridial vaccine or ivermectin suppresses the antibody response to foot-and-mouth disease vaccine in sheep.

Foot-and-mouth disease (FMD) is a highly contagious viral disease affecting cloven-hoofed animals and poses severe economic losses to the sheep industry. In intensive sheep farming, simultaneous administration of the FMD vaccine with other vaccines or anthelmintics is frequently practiced to reduce handling stress and labor costs. However, whether such combined administration can influence the immunological efficacy of FMD vaccine still lacks sufficient research evidence. In this study, 96 healthy sheep were randomly allocated into three groups (n=32 each). Group A received FMD vaccine alone (
