Structural mimics of SARS-CoV-2.

Since its first detection in 2019, Severe Acute Respiratory Syndrome Coronavirus 2 (SARS-CoV-2) has infected approximately 778 million people and claimed 7.1 million lives globally. A deeper understanding of the biology of SARS-CoV-2 was instrumental in facilitating the development of protective vaccines and new therapeutics, as well as evaluating the impact of drug re-purposing to limit the pandemic. To date, approximately 13.64 billion vaccine doses have been administered; with approximately 67% of the global population having completed their primary series of COVID-19 vaccinations. The FDA

An oral multivalent fusion vaccine based on antigenic fragment VP56310-500 and FlaC adjuvant confers effective protection against grass carp reovirus.

Grass carp reovirus (GCRV) poses a serious economic threat to grass carp aquaculture and oral vaccination has become the strategy of choice for its control. In this study, multiple sequence alignment was conducted on the sequences of the VP56 of seven GCRV-II strains. The most frequent amino acid at each site was assigned to the fusion sequence to obtain the sequences of GCRV-FUSION (VP561-512). Through antigenic index analysis, the full-length sequence (VP561-512) was truncated, thereby obtaining five truncated antigen fragments: VP561-210, VP56100-209, VP56210-310, VP56310-500, and VP56410-5

Multiple directed mutagenesis reduces enzymatic activity and antibody recognition of the African Swine Fever Virus E2 ubiquitin-conjugating protein (ASFV-pI215L).

African Swine Fever virus (ASFV) causes a contagious and fatal disease in domestic pigs and Eurasian wild boars, representing a serious threat to the global pig industry, since no antivirals are available and vaccine use is currently restricted to Vietnam. Notably, ASFV encodes for an E2 ubiquitin-conjugating enzyme (ASFV-pI215L) which is essential for viral replication and evasion from immune interferon type I responses, suggesting that its functional impairment could lead to a live attenuated vaccine. In this study, we showed that ASFV-pI215L is highly conserved among 222 ASFV isolates, incl

Development of duck hepatitis A virus type 1 attenuated vaccine E23-SP80 and its protective efficacy evaluation against DHAV-1 infection in ducks.

Duck Hepatitis A Virus type 1 (DHAV-1) is a highly pathogenic virus that causes severe mortality in ducklings and results in substantial economic losses to the global duck industry. Live-attenuated DHAV vaccine remains one of the most effective strategies for controlling this disease. We developed a safe and effective live attenuated vaccine candidate E23-SP80 adapted to specific-pathogen-free (SPF) chicken embryos by serial passage of a field isolate. The E23-SP80 exhibited an adaptive growth capacity in SPF chicken embryos with a viral titer of 107.25 ELD50/0.2 mL and lost its pathogenicity

Evaluated the immune efficacy of a bivalent inactivated vaccine against post-weaning multisystemic wasting syndrome and mycoplasmal pneumonia of swine.

PMWS and MPS are severe respiratory diseases in piglets, causing developmental disorders and significant economic losses. The conventional approach to control these diseases relies on separate immunization with individual vaccines, which is time-consuming, labor-intensive, and may induce stress reactions. The aim of this study was to evaluate the immune efficacy of a bivalent inactivated vaccine containing the clinically isolated Mhp Q strain and purified PCV2 Cap VLPs. Thirty-five Changbai piglets were randomly divided into seven groups (n = 5, per group): A (bivalent inactivated vaccine with

Combination immunization with mRNAs encoding PRRSV antigens enhances immune responses and confers protective immunity against highly pathogenic PRRSV in piglets.

Porcine reproductive and respiratory syndrome virus (PRRSV) has caused significant economic losses in the livestock industry. Due to high genetic variability and limited cellular immunity induction by traditional vaccines, current options offer insufficient protection. In contrast, mRNA vaccines offer flexibility in antigen design and enhanced cellular immune response, presenting a promising solution for PRRSV vaccination. In this study, multiple mRNAs encoding PRRSV structural proteins GP3, GP4, GP5, M, and N, as well as the fusion proteins GP345 and GP5MN, were constructed, encapsulated in l

A novel ORF127/ORF130 gene deletion Orf virus vaccine candidate: construction and evaluation.

BACKGROUND: Orf is a zoonotic infectious disease caused by the Orf virus (ORFV), posing a serious threat to the health of ruminants and humans. Vaccination remains the most effective strategy for disease control. However, the limitations of conventional vaccines necessitate the development of safer and more efficient alternatives. METHODS: Based on the previously attenuated Orf vaccine strain (HB-TS09F65) as the parental virus, two gene-deletion strains, rHB09-ΔORF127 and rHB09-ΔORF130, were successfully constructed by deleting the ORF130 and ORF127 genes via homologous recombination. Development of a High-Hydrostatic-Pressure-Treated Recombinant Vaccine Targeting the Major Capsid Protein of Red Sea Bream Iridovirus.

Red sea bream (Pagrus major) aquaculture represents one of the most economically important marine aquaculture industries in Japan and East Asia. However, viral diseases, particularly those caused by red sea bream iridovirus (RSIV), pose a serious threat to aquaculture production in this region. In this study, we applied high-hydrostatic-pressure (HHP) refolding technology to develop a recombinant vaccine targeting the RSIV major capsid protein (MCP). The recombinant MCP (RSIV-rMCP) expressed in Escherichia coli was insoluble; however, HHP treatment under alkaline (pH 10) conditions in the pres

Development of a Cellular Membrane Nanovesicle-Based Vaccine Against Porcine Epidemic Diarrhea Virus.

Porcine epidemic diarrhea virus (PEDV) has emerged as a major pathogen responsible for porcine diarrheal diseases, causing outbreaks of severe diarrhea and high mortality in neonatal piglets, thereby inflicting severe economic losses on the global swine industry. Current commercial PED vaccines, comprising conventional inactivated and live attenuated formulations, have exhibited progressively diminished efficacy in the face of emerging PEDV variants. The development of high-efficiency vaccine platforms is therefore critical for PED control. This study engineered a cellular membrane nanovesicle

Dynamics of Tilapia Lake Virus in Recirculating Aquaculture Systems and the Impact of Vaccination on Outbreak Control.

Tilapia lake virus (TiLV) is a highly virulent pathogen that has caused substantial mortality in tilapia farms, particularly those with open-water systems. However, TiLV can also emerge and persist in closed environments, such as recirculating aquaculture systems (RAS), where environmental accumulation and repeated exposure may intensify infection and sustain outbreaks. In this case study, we conducted three field experiments to better understand TiLV dynamics among Nile tilapia in RAS. In experiment I, we quantified the TiLV levels in the fish, water, and sediment to compare outbreak and no-o

Sex Differences in Response to Viral Vector Vaccines-Implications for Future Vaccine Design.

Vaccination represents one of the most impactful public health achievements, preventing 3.5 to 5 million deaths annually according to estimates of the World Health Organization. Yet, recent outbreaks of emerging and reemerging infectious diseases highlight the need for rapid and strategic vaccine development using vaccine platforms technologies. Sexual dimorphism in vaccine-induced immune responses has received significant attention in recent years. To ensure vaccine safety and efficacy across sexes, sex-based differences should be considered in vaccine design, dosing, and regimen. Evidence on

Role of γδ T cells in turkey herpesvirus vaccine protection against Marek's disease virus.

γδ T cells are a highly abundant lymphocyte subset in chickens and play key roles in early immune responses to infection. It has been recently shown that γδ T cells restrict Marek's disease virus (MDV) pathogenesis; however, it remained elusive if they play a role in vaccine protection. In this study, we vaccinated γδ T-cell-knockout chickens with the commercial turkey herpesvirus (HVT) vaccine and challenged them with very virulent MDV. The disease incidence was significantly increased in vaccinated chickens in the absence of γδ T cells. This increase was comparable to a previous study in unv

Chitosan nanoparticle based ΔgE IBRV Vaccine: A Novel DIVA compatible strategy for enhanced immunity in guinea pigs.

Infectious Bovine Rhinotracheitis (IBR), caused by Bovine Herpesvirus-1, is a significant disease in cattle that results in substantial economic losses worldwide. Vaccination remains the primary control strategy, with inactivated vaccines offering a safer alternative to live-attenuated counterparts. However, their reduced immunogenicity necessitates the use of adjuvants to enhance protective immune responses. Conventional adjuvants, such as aluminium salts and oil emulsions, are constrained by their Th2-biased immune stimulation, limited ability to elicit strong cellular or mucosal immunity, a

Effects of porcine epidemic diarrhoea virus vaccination timing and route of administration on colostrum antibody levels and prevention of diarrhoea in piglets on a pig farm with porcine epidemic diarrhoea.

Piglets rely on passive immunity acquired from maternal-derived antibodies obtained by ingesting colostrum and milk from immunised sows. This study evaluated the differences in colostrum antibody profiles (IgA, IgG, and neutralising antibodies) according to the timing, frequency, and administration route of the porcine epidemic diarrhoea virus (PEDV) vaccination in pigs with confirmed PEDV infection, and to assess the corresponding clinical protective effects of PEDV vaccination in neonatal piglets. Piglets of sows that received two or more vaccinations, including an oral administration of a l

Comparative efficacy of three mismatched enterotropic Newcastle disease vaccines against genotype VII challenge in chickens.

Newcastle disease (ND) is a highly contagious disease with the potential to cause massive losses in the global poultry industry. This study evaluated the efficacy of three commercial live ND vaccines based on genotypes I and II (enterotropic) on mortality and viral shedding in specific pathogen-free (SPF) chickens challenged with circulating virulent Newcastle disease virus (vNDV) of genotype VII. Forty one-day-old SPF chickens were randomly divided into four experimental groups (n = 10) and vaccinated (one dose ≥ 106 EID50) with one of three enterotropic ND vaccines (E1, E2, or E3) or receive

Molecular characterization and vaccine matching of foot-and-mouth disease virus in Khyber Pakhtunkhwa, Pakistan.

BACKGROUND: Foot-and-mouth disease (FMD) remains a major constraint to livestock productivity in Pakistan, particularly in Khyber Pakhtunkhwa (KP), where recurrent outbreaks cause substantial economic losses. Despite routine biannual vaccination, the continued circulation of FMD virus (FMDV) serotype A predominantly the A-Iran-05 lineage raises concerns regarding antigenic mismatch between field strains and vaccine formulations. METHODS: Results A total of 244 epithelial tissue samples were collected from clinically infected cattle and buffaloes across four districts of KP. Of these, 226 samp

Synergistic effects of oral inoculation with recombinant Lactobacillus plantarum NC8 Co-expressing interleukin-2, interleukin-17b and interleukin-26 on infectious bronchitis vaccination in chickens.

Vaccination against Infectious Bronchitis Virus (IBV) stands as a proven, effective approach for significantly curtailing chicken mortality rates in the poultry sector. Recent extensive research has thoroughly explored this area. Our study validates that a specifically engineered Lactobacillus plantarum NC8 strain, capable of co-producing a fusion protein encompassing chicken interleukin-2 (IL-2), IL-17B, and IL-26 (termed NC8-ChIL2/17B/26), amplifies the comprehensive immune response elicited by the IBV vaccine. We assessed its immune-enhancing potential by orally co-administering it with the

Predicting genetic evolution of viruses to identify suitable vaccines using artificial intelligence.

The evolution of the viruses is rapidly becoming a global challenge to the creation of vaccines since the new variants are often capable of escaping the immune system and decreasing the vaccine efficacy. The traditional methods of genomic epidemiology rely on the retrospective phylogenetic analysis, which can elucidate the previous mutations, but cannot predict the evolutionary trends in the future. In order to address these disadvantages, a new Refined Deep Evolutionary Learning Framework (R-DELF) is proposed that combines the genomic, structural, and temporal intelligence in predicting proac
