Vaccines and antimicrobial resistance: from science to policy-summary and outcomes.

In April 2024, the Royal Society convened a Science+ meeting in London on 'Vaccines and antimicrobial resistance: from science to policy'. The purpose was to review the science of how vaccines reduce antimicrobial resistance (AMR) and discuss policy in advancing development and equitable deployment of such vaccines. The meeting adopted a One Health approach with international speakers presenting from both human and veterinary perspectives. Presentations on Day 1 focused on scientific aspects of the AMR threat and the role of vaccines as counter measures. On Day 2, presentations covered associa

Molecular Detection and Phylogenetic Characterization of Avian Infectious Bronchitis Virus (IBV) in Vaccinated Commercial Chicken Flocks Experiencing Outbreaks in Faisalabad, Pakistan.

Avian infectious bronchitis virus (IBV) remains a persistent threat to poultry health and production, particularly in Pakistan, where frequent outbreaks occur despite routine vaccination. To investigate suspected vaccine breakthrough events, one hundred clinical samples (oropharyngeal/tracheal swabs and kidney, lung, and liver tissues) were collected between January and May 2025 from ten commercial chicken farms experiencing IBV-like outbreaks. Postmortem findings revealed classic IBV-associated lesions, including hemorrhagic tracheitis, severe airsacculitis, nephritis with urate deposits, and

Mucosal vaccination in mice provides protection from diverse respiratory threats.

Traditional vaccines target specific pathogens, limiting their scope against diverse respiratory threats. We describe an intranasal liposomal formulation combining toll-like receptor 4 and 7/8 ligands with a model antigen, ovalbumin, which provided broad, durable protection in mice for at least 3 months against infection with severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and Staphylococcus aureus. In addition, the vaccine protected mice from other viruses (SARS-CoV-2, SARS, SHC014 coronavirus), bacteria (Acinetobacter baumannii), and allergens. Protection was mediated by persist

A Self-amplifying mRNA vaccine for infectious laryngotracheitis virus (ILTV) induces efficient protective immunity.

Infectious laryngotracheitis (ILT) continues to pose a significant threat to intensive poultry farming regions worldwide. Current control strategies primarily rely on live attenuated and recombinant live ILT virus (ILTV) vaccines. However, concerns persist regarding the potential reversion to virulence, recombination of vaccine viruses, and interference from maternal antibodies. mRNA vaccine platforms offer a novel and promising approach for developing next-generation ILTV vaccines. In this study, we designed a self-amplifying RNA (saRNA) vaccine incorporating the ILTV gD gene and evaluated it

Comparison of pre-weaning bovine respiratory disease treatment rates between non-vaccinated control and variably primed and boosted beef calves receiving commercially available bovine coronavirus vaccines.

OBJECTIVE: The primary objective was to determine effectiveness of bovine coronavirus (BCoV) vaccination of neonatal calves in the face of natural respiratory infection in a commercial herd. ANIMALS: At a privately owned ranch in north-central Alberta with a history of bovine respiratory disease (BRD), beef calves of mixed sex and breed were randomized into a clinical vaccine trial. PROCEDURE: At birth, 447 calves were enrolled into the vaccine (VAC) group and administered an intranasal dose of BCoV vaccine, and 439 calves were enrolled as controls (CON). Most VAC calves (n = 389) also recei

Immunoinformatics based designing of a broad-spectrum multi-epitope vaccine against co-infection of human metapneumovirus, respiratory syncytial virus, and influenza A virus.

Co-infections involving human metapneumovirus (hMPV), respiratory syncytial virus (RSV), and influenza A virus (IAV) often exacerbate disease severity in vulnerable populations. Here, we employed a structure-based immunoinformatics approach to design a multi-epitope subunit vaccine targeting these pathogens. The construct incorporated two epitopes each for cytotoxic T lymphocytes (CTLs), helper T lymphocytes (HTLs), and B cells, derived from the fusion proteins of hMPV and RSV, as well as the neuraminidase protein of IAV. These epitopes were linked with an adjuvant and optimized spacers to enh

SFTSV: a priority pathogen in need of a cure-reviewing the therapeutic.

Severe fever with thrombocytopenia syndrome (SFTS), caused by the Bandavirus dabieense (Severe fever with thrombocytopenia syndrome virus: SFTSV), is an emerging tick-borne disease with a case-fatality rate of 5–30%. Endemic to East Asia with increasing global incidence, SFTS poses a significant public health threat, yet no specific antiviral or vaccine is approved. This review provides a comprehensive analysis of current therapeutic candidates for SFTS. We reviewed progress in drug repurposing (e.g., favipiravir), host-directed therapies, immunomodulators (including problematic steroid use),

Bluetongue in China: Current Status of Viruses, Vectors, Detection Methods, and Vaccines.

Bluetongue (BT) is a vector-borne viral disease caused by the bluetongue virus (BTV), which can affect a variety of wild and domestic ruminants. Due to its significant impact on ruminant health and national economies, BT is classified as a notifiable multispecies disease by the World Organization for Animal Health (WOAH). In China, BT is listed as a Class II multispecies animal disease. This article provides a comprehensive review of the distribution of BTV and its primary insect vector, Culicoides, in China. Since BTV was first reported in China in 1979, BTV antibody-positive samples have bee

MVA-Spike encoding the A subunit of dmLT safely enhances systemic and mucosal immune responses.

INTRODUCTION: Mucosal immunity provides frontline protection at respiratory, gastrointestinal, and urogenital surfaces, where secretory IgA and tissue-resident T cells such as TH17 limit colonization and early replication of pathogens. Conventional parenteral vaccines typically induce robust systemic immunity but fail to elicit strong mucosal responses. Therefore, the development of safe and effective strategies to enhance mucosal immunity remains a key priority in vaccine research. METHODS: We designed a modified vaccinia virus Ankara (MVA) construct expressing the double mutated heat-labile

Preparation of poloxamer-based CPV virus-like particle vaccine and its immunological evaluation in mice.

Poloxamers are thermosensitive triblock copolymers valued for their safety and sustained-release properties. Using China's prevalent CPV-2c strain (JSLYG-18) of canine parvovirus (CPV), we produced virus-like particles (VLPs) via prokaryotic expression and formulated a thermosensitive poloxamer gel vaccine. The results showed that the optimal formulation of the poloxamer thermosensitive gel consisted of 17% poloxamer 407 (P407), 3.2% poloxamer 188 (P188), and 0.3% hydroxypropyl methylcellulose (HPMC), with a gelation temperature (Tm) of 35.8 °C, demonstrating excellent stability and ease of pr

Vaccine-Associated Recombination and Virulence Heterogeneity of GI-19 (QX Type) Infectious Bronchitis Virus in Eastern and Southern China, 2024-2025.

Infectious bronchitis (IB) virus (IBV) remains a major pathogen threatening the poultry industry. Its rapid mutation and recombination continuously generate variants that disseminate worldwide. Between May 2024 and February 2025, 49 field strains were isolated from chickens vaccinated with live-attenuated IBV vaccines (H120, 4/91, or QXL87) in four Chinese provinces (Jiangsu, Anhui, Shandong, and Guangdong). Based on full-length S1 gene sequencing, all isolates were classified into genotype GI, including lineages GI-13 and GI-19. Phylogenetic analysis revealed that GI-19 (QX-type) comprised as

RBD-SD1 Nanoparticle Vaccines From DPP4-Using Merbecoviruses Elicit a Cross-Reactive Antibody Response but Limited Cross-Protective Immunity.

Coronaviruses within the Merbecovirus subgenus, including Middle East respiratory syndrome coronavirus (MERS-CoV) and its dipeptidyl peptidase 4 (DPP4)-using relatives, pose a persistent zoonotic threat. Efforts to prepare for future Merbecovirus spillover events require vaccines that protect beyond a single virus strain. To evaluate antigenic conservation and cross-protective potential, SpyCatcher-mi3 nanoparticles displaying the receptor-binding domain and subdomain 1 (RBD-SD1) from three DPP4-using merbecoviruses, MERS-CoV, NL140422, and HKU4, were generated. Female mice immunized with thes

Vertical transmission of Orf virus in goats and its prevention.

Contagious ecthyma dermatitis, caused by Orf virus (ORFV), is an important zoonotic disease of small ruminants, particularly goats and sheep, and can occasionally infect humans. The morbidity of the disease is high in newborn kids or lambs as compared with adult animals, even leading to death in severe cases due to emaciation or secondary infections. Despite its significance, the precise route of ORFV transmission to newborn kids or lambs has remained unclear. In this study, we hypothesized that ORFV may be transmitted transplacentally from dam to fetus and investigated the presence of the vir

A lyophilized stabilizer formulation enhances thermostability and preserves the immunogenicity of Senecavirus A post thermal challenge.

Senecavirus A (SVA) is a pathogenic virus that causes vesicular lesions, locomotor disorders, and neonatal mortality in pigs. Its increasing global prevalence poses a persistent threat to the swine industry. Vaccination remains the primary control strategy; however, SVA exhibits pronounced thermal instability, and exposure to unsuitable temperatures during storage or transport can impair vaccine immunogenicity. We developed a heat-stable lyoprotectant formulation for live SVA vaccines to overcome this limitation. The formulation, composed of 10% trehalose, 2% inulin, 2% dextran 40, 2% mannitol

Generation of a Bivalent Recombinant Vaccine Conferring Dual Protection Against Potyvirus and Orthotospovirus in Cucurbits.

Climate warming has facilitated the expansion of insect vectors and plant viral pathogens, leading to increased incidence of viral diseases in crops. Cucurbit crops, including cucumber (Cucumis sativus), melon (Cucumis melo), squash (Cucurbita pepo), and watermelon (Citrullus lanatus), are of major economic importance worldwide, but their production is severely threatened by viral infections. Among the most damaging viruses are zucchini yellow mosaic virus (ZYMV; genus Potyvirus), transmitted by aphids, and melon yellow spot virus (MYSV; genus Orthotospovirus), transmitted by thrips, both of w

Vaccination trends and operational challenges in Peste des Petits Ruminants eradication in Ethiopia.

Peste des Petits Ruminants (PPR) is a major transboundary disease in Ethiopia, where the national goal is eradication by 2027. However, evidence on vaccination performance and operational challenges remains limited. This study assessed vaccination trends over six years (September 2018–August 2024) and conducted surveys with animal health professionals (AHPs) (January–May 2024) to identify key enabling factors and constraints influencing eradication efforts in the North Shewa Zone. The findings revealed that a risk-based vaccination strategy (RBVS) has been implemented since 2018, focusing on h

Immunodominance is a poor predictor of vaccine-induced T follicular helper cell quality.

BACKGROUND: Rational engineering of vaccine immunogens to focus B cell responses on potently neutralising epitopes is a promising approach to improve the potency, breadth and durability of viral vaccines. Such strategies, however, can compromise vaccine immunogenicity through the unintended exclusion of CD4+ T cell epitopes, which are critical for the development of T follicular helper (TFH) cells and to support high affinity antibody production. METHODS: Using a prototypic influenza haemagglutinin (HA) stem immunogen lacking effective CD4+ T cell help in C57BL/6 mice, we interrogated the min
