A dual-gene-deleted ASFV Lv17/WB/Rie1-ΔCD candidate administered orally to wild boar confers DIVA-compatible protection against virulent challenge.

African swine fever (ASF) continues to expand worldwide. Recent detection in wild boar in Spain highlights the urgent need for effective control tools, with oral vaccination as a key priority. Following previous evaluation of the attenuated Lv17/WB/Rie1 strain, we assess an improved derivative, Lv17/WB/Rie1-ΔCD, lacking EP402R (CD2v) and EP153R, replaced by GFP to abrogate haemadsorption and enable Differentiating-Infected-from-Vaccinated-Animals (DIVA) diagnostics. Vaccinated animals received either a single high dose (104 TCID₅₀) or a prime and re-exposure regimen (102 TCID₅₀ plus a 104 TCID

Canine parvovirus type 2 infection in vaccinated puppies: role of vaccination practices and viral antigenic variation.

BACKGROUND: Canine parvovirus type 2 (CPV-2) infection is reported in vaccinated puppies in Egypt, yet contributing factors remain poorly investigated. This study evaluated the role of vaccination practices and CPV-2 antigenic variation in disease occurrence in puppies whose primary vaccination series was recorded as finished by attending veterinarians. METHODS: Puppies with clinical signs of parvoviral enteritis at three veterinary clinics in Giza, Egypt (June–October 2020) were enrolled if vaccination history was documented. All non-vaccinated puppies were included, whereas among vaccinated

Development of the European Veterinary Medicines Gaps and Needs Compass for Sheep and Goats Based on Online Survey and Expert Knowledge Elicitation.

Limited availability of veterinary medicinal products (VMPs) for small ruminants is a long-standing challenge. This mixed-methods study provides the first systematic definition and assessment of (i) shortages, (ii) lack of availability and (iii) unmet needs for sheep and goats across Europe. Survey data from 96 European veterinarians in 13 European countries (the majority of whom were from Spain, Germany, France, and Greece), a market analysis of authorized and available VMPs via the EMA Union Products Database (UPD) and expert knowledge elicitation (EKE) by 23 specialists were combined. An inactivated vaccine confers potent and durable protection against a screened virulent rhabdovirus in hybrid snakehead.

Hybrid snakehead rhabdovirus (HSHRV) poses a serious threat to hybrid snakehead (Channa maculata ♀ × C. argus ♂) aquaculture, causing severe outbreaks characterized by high mortality. To develop an effective vaccine, five rhabdovirus strains were isolated from diseased fish across Guangdong Province. Comparative analysis of virulence and host specificity identified a highly pathogenic strain, designated HSHRV-JM0909, which replicated to high titers consistently exceeding 108.0 TCID50/mL in mandarin fish brain (MBC) cells. Phylogenetic analysis classified the virus within the genus Siniperhavir

The α-Gal Epitope (Galα1-3Galβ1-4GlcNAc) as Therapeutic Agent in Cancer Immunotherapy, Vaccine Effectiveness Amplification and Injured Tissue Regeneration.

The α-gal epitope is synthesized in non-primate mammals and New-World monkeys by the glycosylation enzyme α1,3galactosyltransferase (α1,3GT), encoded by the GGTA1 gene. Ancestral Old-World monkeys and apes synthesizing α-gal epitopes underwent extinction 20-30 million years ago. Their mutated offspring, with the inactivated GGTA1 gene, survived and produced the natural anti-Gal antibody, specifically binding α-gal epitopes. Anti-Gal protected the surviving offspring from lethal viruses presenting α-gal epitopes, which killed α-gal-synthesizing parental primates. Anti-Gal constitutes ~1% of hum

Isolation of a Variant of the Felid Herpesvirus 1-Live Attenuated Vaccine Strain F2 from Unvaccinated Cats.

INTRODUCTION: Felid herpesvirus 1 (FeHV-1) is widespread worldwide and is sometimes associated with chronic diseases in cats; therefore, it is clinically important. Vaccination can be used to control FeHV-1 infection. We previously isolated a strain identical to the live attenuated vaccine (LAV) strain F2 from a feline eye with a dendritic ulcer, indicating that the F2 strain may be pathogenic. The origin of the F2 strain that we previously isolated is unknown, and we surmised that the F2 strain may exist in the field. To determine whether the LAV strain F2 exists in the field, we, therefore,

Recombination with a vaccine strain drives the evolution of a more virulent phenotype in avian infectious bronchitis virus.

Avian infectious bronchitis virus (IBV), a major pathogen in the poultry industry, evolves rapidly via genetic mutations and recombination. While recombination between field and live vaccine strains is well-documented, its direct role in viral virulence remains unclear. In this study, four novel IBV field strains (SX/2407, SX/2408, SX/2409, SX/2410) were isolated from a single layer farm in Shanxi Province, China, and genetically characterized. Comprehensive genomic analysis revealed that SX/2409 is a natural recombinant, with SX/2408 (co-isolated) as the major parent and a 4/91-like vaccine l

Synthetic progestin treatment alters cytokine expression and impairs vaccine response in horses.

Altrenogest is commonly used in equine veterinary medicine for the control of estrus, behavioral modifications, and maintenance of pregnancy. Recent studies have found that altrenogest may impact aspects of the immune system, including altering cytokine production in circulation and within the reproductive tract. Therefore, we hypothesize that this alteration may lead to a deviated immune response to vaccination. Sixteen mares were divided into two groups, with ten mares (n = 10) receiving long-acting injectable altrenogest (100 mg/mL; IM) administered every 10 days, and six mares (n = 6) rece

Structure-guided discovery of neutralizing epitopes enables rational design of a multi-epitope nanoparticle vaccine against SVCV.

Spring viremia of carp virus (SVCV) imposes a significant economic burden on carp aquaculture, whereas the availability of precisely designed vaccines against this pathogen remains limited. Here, we propose a structure-guided strategy for epitope discovery based on the interaction interface between a neutralizing antibody and the SVCV glycoprotein (G). Three conserved antibody-targeted epitopes on the G protein were identified through protein docking and alanine-scanning energy analysis, and their immunological relevance was further validated by epitope-specific antibody depletion assays and i

Assessment of humoral immune responses elicited by an inactivated lumpy skin disease vaccine in cattle: a preliminary study.

The control of lumpy skin disease (LSD) is often hampered by adverse effects associated with currently available live attenuated vaccines (LAVs). This preliminary study was therefore designed to determine the immune responses in dairy cattle immunized with either an inactivated LSD vaccine (derived from a local strain) or a commercial LAV. Specifically, five cattle received an inactivated vaccine, and five received a commercial LAV. Humoral immunity was assessed: ELISA titers persisted for over six months in both groups. Furthermore, the virus neutralization test showed no statistical differen

Targeting TLR2 agonists as immunomodulators with broad antiviral activity.

Toll-like receptor-2 (TLR2) serves as an innate immune sensor that recognizes specific viral proteins, thereby initiating signaling pathways that can either enhance antiviral host defenses or induce pathological inflammatory responses. In this forum, we discuss the role of TLR2 and its agonists in enhancing antiviral and vaccine-induced immune responses.

In silico design of a novel multi-epitope mRNA vaccine candidate for BtHKU5-CoV-2 using immunoinformatics.

Bat HKU5-CoV-2 (BtHKU5-CoV-2), a recently discovered bat-infecting merbecovirus, was found to infect human cell lines by utilizing the human angiotensin-converting enzyme 2 (ACE2) receptor, similar to SARS-CoV-2, which caused millions of deaths. Moreover, its broad host tropism has raised significant concerns about potential human spillover risk. Therefore, there is an urgent need to develop vaccines to combat the potential outbreak of BtHKU5-CoV-2. However, research focusing on BtHKU5-CoV-2 remains limited. In this study, we designed a novel multi-epitope vaccine for BtHKU5-CoV-2 using an imm

Oral immunization with recombinant Lactococcus lactis expressing VP3 and VP3-IFNα fusion protein protects ducklings against novel goose parvovirus infection.

The novel goose parvovirus (NGPV), an emerging variant of goose parvovirus, is the causative agent of short beak and dwarfism syndrome (SBDS) in ducks, posing a substantial threat to the global waterfowl industry. There is an urgent need for safe and convenient vaccines that can effectively induce mucosal immunity. This study aimed to develop and evaluate the immunogenicity and protective efficacy of an oral vaccine based on recombinant Lactococcus lactis (L. lactis) expressing the major capsid protein VP3 of NGPV, with or without fusion to duck interferon-alpha (IFNα). Diagnostic approach, vaccination and treatment priorities for Lumpy Skin Disease in cattle: lessons from Balkan epidemics to Western Europe re-emergence (2025).

Lumpy skin disease (LSD) remains a high-impact, transboundary infection of cattle driven by capripoxvirus transmission via mechanical vectors, causing morbidity, production losses, and trade disruption. Following the 2015–2017 Balkan epidemics, coordinated high-coverage vaccination campaigns and movement restrictions substantially reduced disease transmission across southeastern Europe. However, the recent re-emergence of LSD in Western Europe in 2025, including outbreaks reported in Italy, France, and Spain, demonstrates that regional elimination remains fragile and requires sustained prepara

T cell responses to nonstructural proteins promote cross-serotype immunity to foot-and-mouth disease virus.

UNLABELLED: T-cell responses play a pivotal role in protection against foot-and-mouth disease virus (FMDV), by enhancing neutralizing antibody (NAb) titers and shortening the duration of infection. To further elucidate the protective immune mechanisms mediated by T cells and develop novel candidate vaccines with cross-protective potential, multiple CD4+ and CD8+ T-cell epitopes were identified targeting FMDV nonstructural proteins 2B and 2C in the infected pigs. Given its critical role in dendritic cells targeting and enhancing antigen presentation, CD40L was fused with T-/B-cell epitopes to c

Foot-and-mouth disease virus-like particle mRNA vaccine induces strong humoral and cellular immunity.

Foot-and-mouth disease (FMD) is a major animal infectious disease that has garnered significant international attention. Currently, conventional inactivated vaccines and virus-like particle (VLP) vaccines primarily induce protective immune responses through the generation of neutralizing antibodies. However, these vaccines exhibit limited activation of cellular immunity and offer short-term immune persistence. Furthermore, inactivated vaccines raise certain biosafety concerns. In this study, an FMD mRNA vaccine capable of self-assembling into non-infectious VLP was developed. Immunization of m

Development and validation of a crystal digital PCR assay for duck Tembusu virus detection and vaccine monitoring.

Duck Tembusu virus (DTMUV) threatens the duck industry and public health due to its broad host range. This study established and validated a crystal digital PCR (cdPCR) assay for molecular detection of DTMUV, comparing it with quantitative real-time PCR (qPCR). A recombinant plasmid standard (pDTMUV, targeting the DTMUV E gene) served as a reference. Optimized at 58°C annealing temperature with optimal primer/probe concentrations, cdPCR showed high specificity (only DTMUV positive) and precision (low intra-/inter-assay CVs), with a limit of detection (LOD) of 0.19 copies/μL-100-fold more sensi

Immune protection in chickens via lipid nanoparticle-encapsulated VP2 DNA vaccine against very virulent infectious bursal disease virus.

Infectious bursal disease (IBD), caused by infectious bursal disease virus (IBDV), is an acute and highly contagious avian infection that primarily causes severe immunosuppression in chickens. Since the 1980s, very virulent IBDV (vvIBDV) has triggered a global crisis in the poultry industry due to its high mortality. Although IBD has become largely sporadic in recent years following widespread vaccination, the persistent threat of vvIBDV underscores the critical need for novel vaccines that are both safer and more effective. In this study, a lipid nanoparticle (LNP)-encapsulated DNA vaccine en
