The Delay in the Licensing of Protozoal Vaccines: A Comparative History.

Although viruses and bacteria have been known as agents of diseases since 1546, 250 years went by until the first vaccines against these pathogens were developed (1796 and 1800s). In contrast, Malaria, which is a protozoan-neglected disease, has been known since the 5th century BCE and, despite 2,500 years having passed since then, no human vaccine has yet been licensed for Malaria. Additionally, no modern human vaccine is currently licensed against Visceral or Cutaneous leishmaniasis. Vaccination against Malaria evolved from the inoculation of irradiated sporozoites through the bite of Anophe

Successful vaccines for naturally occurring protozoal diseases of animals should guide human vaccine research. A review of protozoal vaccines and their designs.

Effective vaccines are available for many protozoal diseases of animals, including vaccines for zoonotic pathogens and for several species of vector-transmitted apicomplexan haemoparasites. In comparison with human diseases, vaccine development for animals has practical advantages such as the ability to perform experiments in the natural host, the option to manufacture some vaccines in vivo, and lower safety requirements. Although it is proper for human vaccines to be held to higher standards, the enduring lack of vaccines for human protozoal diseases is difficult to reconcile with the compara

Delayed type hypersensitivity theilerin test in cattle vaccinated against Theileria annulata infection.

Theilerin was prepared and its total nitrogen was determined at level of 1 mg/ml. Seventy-two Holstein Friesian cattle recovered from acute theileriosis or vaccinated received 0.1 ml of theilerin intradermally at normal concentration of total nitrogen or other dilutions (1 / 10 and 1 / 100). Some 60(% of calves and 66.67% of milking cows receiving normal theilerin showed more than 1 mm increase in skin thickness at the site of inoculation (mean value of 1.86 mm for calves and 3.46 mm for milking cows). The pathological examination of the inoculation site showed infiltration of lymphocytes. No

Recent development in control of Theileria annulata in Iran.

During 1973-1990 cattle immunization, in Iran, was induced by two strains within an interval of one month: first the milder and then the mild strain. Although this method of vaccination rendered satisfactory results in the field, yet production, maintenance in deep-freezers and transportation in liquid nitrogen particularly to remote areas of the country proved to be uneconomical and time consuming. Therefore, in order to reduce cost and save time, a new method involving only one local and live attenuated vaccine strain was sought. Reports received from different ecological areas of the countr

Seroprevalence of Toxoplasma gondii in cattle, sheep and goats in Iran.

Serum samples from 2000 cows, 3311 sheep and 638 goats from Iran were examined for antibodies to Toxoplasma gondii by use of the latex agglutination (LAT) and indirect hemagglutination tests (IHAT). Antibodies to T. gondii were found in 24.50% sheep and 19.25% of goats. Antibodies to T. gondii were not detected in cow sera by LAT and IHAT in 1:8 and 1:64 dilutions of bovine sera, respectively. Toxoplasma gondii was not found in tissues of 300 aborted fetuses from cows by direct microscopy and bioassay in mice.

Studies on the cellular site of action of the adjuvant activity of saponin for sheep erythrocytes.

Saponin, which complexes with cholesterol in cell membranes, is a powerful adjuvant for antigens such as whole cell protozoal vaccines and SRBC. Saponin attached to SRBC ghosts potentiated the humoral response; but injection of saponin 1 or 2 days before or after SRBC at the same subcutaneous site was also effective. After the subcutaneous injection of saponin and SRBC the cellularity of the draining lymph node increases sixfold, and indirect PFC appear in it and the spleen. Splenectomy increases immunopotentiation by saponin, but T cell depletion abolishes it. The data show that attachment of

A comparison of saponin with other adjuvants for the potentiation of protective immunity by a killed Plasmodium yoelii vaccine in the mouse.

The protective immunity conferred by subcutaneous injection of outbred CD-1 mice with a killed Plasmodium yoelii (YM strain) vaccine was strongly potentiated by saponin. By adjusting the dose of antigen, the number of immunizations and the number of living parasites in the challenge infection, conditions were defined where antigen alone was non-protective but 100% protection was obtained by the addition of saponin. Inbred BALB/c, CBA/CA and C57 B1 mice were much less responsive than the CD-1 mice. The following adjuvants were compared with saponin: mineral oil emulsions (Freund's incomplete an
