ImmunoCAP-Based Detection of Alpha-Gal Syndrome: A Reproducible Diagnostic Protocol for Human Patients.

Alpha-GAL syndrome (AGS) is a type of allergy to mammalian meat and other foods or pharmaceuticals made from mammalian substrates. It is an IgE-mediated reaction caused by sensitization to the polysaccharide galactose-α-1,3-galactose (alpha-GAL) found in tick saliva after a previous tick bite. Alpha-GAL is present on proteins of mammals except primates. Sensitization to alpha-GAL in tick saliva predisposes to cross-reactivity following exposure (oral or non-oral) to glycated galactose-α-1,3-galactose proteins, regardless of their origin. Symptoms of varying severity (from dyspepsia to anaphyla

Protocols for Diagnosis and Management of Arthropod Bite Allergies in Humans.

Allergic diseases have increased significantly in recent decades, with a higher incidence of severe manifestations such as anaphylaxis. This increase is attributed mainly to environmental factors associated with the Western lifestyle that alter the epithelial barrier, promoting sensitization to allergens, such as those from arthropods. Hymenoptera (bees and wasps) are the most implicated arthropods. The sting introduces a venom composed of highly sensitizing proteins that are capable of triggering reactions ranging from local and mild to potentially fatal systemic reactions. The incidence is h

The Zebrafish Model for Alpha-Gal Syndrome.

Zebrafish (Danio rerio) has emerged as a robust vertebrate model for studying immune and allergic disorders, including food allergies. The alpha-Gal syndrome (AGS) is a tick-borne allergy characterized by an IgE-mediated delayed hypersensitivity to the oligosaccharide galactose-α-1,3-galactose (α-Gal), which is present in tick saliva and mammalian-derived foods and drugs. This chapter describes the experimental procedures used to establish and analyze the validated zebrafish model for AGS. Adult zebrafish were intramuscularly injected with Ixodes ricinus tick saliva and maintained on a mammali

Flow Cytometry in Human Cells Exposed to Tick Salivary Proteins.

The Indirect basophil activation test (iBAT) is a flow cytometry-based assay in which basophils are first passively sensitized with patient-derived IgE and subsequently stimulated with the allergen of interest to assess activation markers on the cell surface. This method was implemented by using the human KU812 cell line to investigate basophil activation mechanisms triggered by tick-derived biomolecules. Unlike the traditional BAT, the iBAT approach avoids variability from non-responder patients and donor-dependent differences, providing a stable and reproducible in vitro model. KU812 cells w

Experimental Food Allergy: From Induction to Anaphylaxis Assessment in a Rat Model.

This chapter describes the experimental methodology used to establish a rat model of oral food allergy. The final protocol has been optimized for the Wistar strain. The model mimics chronic oral allergen re-exposure and allows the study of effector-phase allergic responses. It encompasses all experimental stages, including preliminary procedures, intramuscular sensitization, the use of pro-type 2 immune response adjuvants, allergen re-sensitization, and the final oral challenge, using ovalbumin (OVA). Key outcomes include clinical parameters such as sign scoring, diarrhea classification, and b

Experimental Model to Study Microbiota Perturbation Role in Alpha-Gal Syndrome.

This chapter describes a standardized experimental protocol to induce and analyze galactose-α-1,3-galactose (α-Gal)-specific IgE responses in α-Gal-deficient (α1,3-galactosyltransferase knockout) mice. The absence of endogenous α-Gal in this model enables controlled sensitization to exogenous α-Gal-containing antigens from microbial, arthropod, and mammalian sources. The protocol integrates sequential oral exposure to α-Gal-expressing Escherichia coli O86:B7, capsule-based infestation with Ixodes ricinus nymphs, oral administration of porcine tissue homogenate, and subcutaneous injection of ti

Clinical Protocol for Evaluating Ectoparasite-Induced Allergies with Emphasis on Alpha-Gal Syndrome.

Alpha-gal syndrome (AGS) is an ectoparasite-associated IgE-mediated hypersensitivity in which reactions occur after exposure to galactose-alpha-1,3-galactose (alpha-Gal) in mammalian foods, biologics, medications, and other mammalian-derived products. The delayed onset after oral exposure and the imperfect correlation between sensitization and clinical allergy complicate diagnosis. This chapter provides a practical clinical protocol for evaluating suspected ectoparasite-induced allergy with emphasis on AGS. The protocol integrates structured history taking, tick and other ectoparasite exposure

B-Cell Aware Analysis of Single-Cell Transcriptomics Data.

Single-cell RNA sequencing (scRNA-seq) has facilitated the studies of cellular heterogeneity in many different biological contexts, including how B cells function and mediate immune responses to infection, malignancies, and autoimmunity. scRNA-seq as applied to B cells requires specific bioinformatics strategies, which consider the distinct biology of B cells and biological processes unique to these cells, such as somatic hypermutation (SHM) and class switch recombination (CSR). We present here a protocol that analyses scRNA-seq alignments and count matrices, as well as B-cell receptor sequenc

A theoretical framework for efficient solvent suppression in solid-state NMR: Circumventing solvent-signal refocusing by adiabatic-inversion-based dephasing.

We recently developed a solvent-suppression scheme employing adiabatic-inversion pulses for high-resolution solid-state NMR (SSNMR), termed Solvent suppression of Liquid signal with Adiabatic Pulse (SLAP) [T. Matsunaga et al., J. Biomol. NMR 75, 365 (2021)], and demonstrated its superior performance over the traditional MISSISSIPPI scheme [D.H. Zhou et al., J. Magn. Reson. 192, 162 (2008)]. Here, we present a theoretical framework explaining this advantage. We show that MISSISSIPPI-based suppression is intrinsically limited by solvent-signal refocusing. Theory and simulations revealed that up

Aggregation of Mutant p53 Alters Sialylated and Sulfated Glycan Profiles and Enhances Siglec-9 Ligand Availability in Ovarian Cancer.

PURPOSE: Mutant p53 proteins form aggregates, and the presence of such cytoplasmic p53 aggregates has predicted poor prognosis in several cancers including high-grade serous ovarian cancer (HGSOC). The existence of aberrant expressions of sialylated and sulfated glycans is a major feature of various cancers. At present, the correlation between the presence of cytoplasmic p53 aggregates and the existence of aberrant glycosylation in cancers is unclear. Here, we investigated whether the presence of p53 aggregates may affect sialylated and sulfated glycan profiles in HGSOC. MATERIALS AND METHODS

IL-2 and CD40L can establish and sustain a human B cell-mediated immune regulatory network.

UNLABELLED: B cells can control inflammation, but the factors governing B cell-mediated immune regulatory network remain unclear. We report that IL-2/CD40L enable class-switched memory (CSM) B cells to acquire an immunosuppressive phenotype characterized by the expression of surface TIGIT as well as multiple other inhibitory receptors (PD-1, LAG-3, GITR, CD39, and CTLA-4) and soluble molecules (IL-10, granzyme B, and IL-35). TIGIT + CSM B cells expressing functional CCR9 exhibit significant defects in total immunoglobulin secretion, while preserving IgA2 secretion. Furthermore, IL-2/CD40L-indu

Rabies immunoglobulin utilization and delayed presentation following category III rabies exposure: a 10-year retrospective study in Hangzhou, China.

BACKGROUND: Rabies is an almost invariably fatal, vaccine-preventable zoonosis. Rabies immunoglobulin (RIG) is recommended for Category III exposures, yet real-world RIG utilization remains poorly characterized. This study examined temporal trends and factors associated with RIG utilization and delayed presentation among Category III exposure patients in China. METHODS: A retrospective study was conducted using clinical records of Category III exposure patients presenting to a high-volume rabies prevention clinic in Hangzhou, China, from 2015 to 2024. Calendar year 2018 was treated as a tempo

Stool biomarkers discriminate confirmed necrotizing enterocolitis in preterm infants.

BACKGROUND: Necrotizing enterocolitis (NEC) is a major cause of morbidity and mortality in preterm infants. Early diagnosis remains challenging because clinical signs are nonspecific. We evaluated whether stool biomarkers could discriminate preterm infants with NEC from those without NEC. METHODS: We performed an ancillary analysis within the multicenter ClosNEC cohort of infants born before 32 weeks' gestation. Stool samples were collected at diagnosis in infants with confirmed NEC (Bell stage II or higher) and at comparable postnatal ages in preterm infants without NEC. Fecal calprotectin (

[IgA Nephropathy Update 2025/2026].

Immunoglobulin A nephropathy (IgAN) is the most common form of primary glomerulonephritis and has undergone a therapeutic revolution in just a few years that is unprecedented in nephrology. For the first time, the guidelines define remission as a realistic treatment goal and solidify the so-called "dual-pillar" approach. This educational article summarizes the current recommendations and developments and evaluates the clinical evidence.

Longitudinal basophil activation testing for assessing response to low-dose initiation, slow-escalation peanut oral immunotherapy in patients with high Ara h 2-sIgE.

BACKGROUND: Peanut oral immunotherapy (pOIT) can achieve desensitization, yet patients with high peanut-sIgE may experience more adverse reactions and treatment failure. We evaluated a conservative pOIT protocol in children with high Ara h 2-sIgE (≥40 kU/L) and examined longitudinal basophil activation test (BAT) and sIgE/sIgG4 trajectories as candidate biomarkers. METHODS: Thirty-two peanut-allergic children underwent pOIT, including 16 with high and 16 with lower Ara h 2-sIgE. All received low-dose initiation and slow up-dosing. Clinical outcomes, safety, and immunologic markers were evalua

Hemolin modulates IMD-related immune responses and promotes cellular immunity in Ostrinia furnacalis.

Ostrinia furnacalis is one of the most destructive Lepidopteran pests in Asia and worldwide, reducing yields of corn and other grain crops. Hemolin, an immunoglobulin-like protein with both developmental and immunological functions, has been implicated in protecting insects from pathogen infections. However, the specific regulatory mechanism by which Hemolin modulates innate immunity in O. furnacalis remains unknown. In this study, the full-length Of-Hemolin cDNA sequence was obtained by rapid amplification of cDNA ends and characterized through bioinformatic analysis. Expression levels of Of-

Protective effects of nano-zinc and nano-selenium compared with a toxin binder in broiler chickens fed aflatoxin B1-contaminated diets through antioxidant, immune, and skeletal responses.

Aflatoxin B1 (AFB1) can impair broiler performance, redox balance, and skeletal health. This 42-day study evaluated whether partial replacement of conventional inorganic zinc (Zn) and selenium (Se) sources with nanoforms could mitigate AFB1-associated responses and compared this source-substitution strategy with the use of commercial toxin binder. A total of 792 day-old male Ross 308 broilers were allocated to six treatments (6 replicate pens/treatment, 22 birds/pen; pen served as the experimental unit): Control; AF (0.5 mg AFB1/kg); AF+TB (2 g toxin binder/kg); AF+NZ (40 mg Zn/kg supplied as

Dietary supplementation with coated zinc oxide enhanced growth performance by modulating systemic immunity and microbial composition in weaned pigs.

Dietary zinc oxide (ZnO) is widely adopted in the swine industry to alleviate post-weaning diarrhea and promote the growth performance of weaned pigs. However, its low rate of absorption in the gastrointestinal tract leads to excessive excretion, which causes severe environmental pollution. To overcome this limitation, dietary coated ZnO (CZO) has been studied to enhance zinc absorption and mitigate its environmental influence. In the present study, we investigated the effects of dietary CZO on growth performance, frequency of diarrhea, nutrient digestibility, hematological and immune response

A sensitive colorimetric ELISA to measure allergen-specific salivary SIgA for monitoring sublingual immunity efficiency in pollinosis SLIT.

BACKGROUND: Sublingual immunotherapy (SLIT) is safe and one of the most effective etiological treatment for pollinosis. Allergen-specific salivary secretory immunoglobulin A (SIgA) is a useful marker to objectively evaluate treatment efficacy. However, saliva samples usually exhibit high background noise because of non-specific binding in enzyme-linked immunosorbent assays (ELISAs). We therefore aimed to establish an amplified ELISA to measure allergen-specific salivary SIgA levels to assess the efficacy of SLIT for Japanese cedar pollinosis. METHODS: Biotinyl-tyramide (BT-Tyr) was used to am
