Liposome Loaded With Yeast-Derived Nanoparticles for the Enhanced Photothermal-Immunotherapy of Malignant Lymphoma.

Nanoparticles derived from various animals, plants, and microorganisms demonstrate potent application potential in the construction of functionalized pharmaceutic preparations, owing to their special in vivo behavioral and immune-stimulating effects. Yeast cell wall-derived nanoparticles (Ycws) can reverse the tumor immunosuppressive microenvironment via activating various cells related to anti-tumor immunity, while it is difficult to ensure the complete elimination of the treated tumors. Meanwhile, photothermal therapy (PTT) can effectively ablate the irradiated tumors with cell pyroptosis in

Compensatory pathways in tryptophan metabolism and immune regulation following IDO inhibition.

Indoleamine 2,3-dioxygenase 1 (IDO1) is an intracellular heme-dependent enzyme that catalyzes the initial oxidation of L-tryptophan into the kynurenine pathway, thereby linking tryptophan metabolism to immune regulation. IDO1 expression is mainly induced by interferon-γ during immune activation and inflammation, and its activity helps control immune responses through tryptophan depletion and the generation of immunomodulatory metabolites. Because of its central role in immunometabolism, IDO1 has been widely investigated as a therapeutic target, and several strategies have been developed to inh

Impact of dynamic beam delivery of intensity-modulated radiation therapy and volumetric modulated arc therapy on circulating immune cell exposure in glioma.

BACKGROUND AND PURPOSE: To compare how intensity-modulated radiation therapy (IMRT) and volumetric modulated arc therapy (VMAT) affect circulating blood cells (CBC) depletion in glioma patients. MATERIALS AND METHODS: Twenty-eight glioma patients were retrospectively analyzed. Mean clinical target volume (CTV) was 293.8 ± 152.4 cm3 (μ± σ ). For each clinical IMRT or VMAT plan, a corresponding plan using the alternative technique was generated with identical objectives and constraints. A 4D dynamic blood-flow model simulated cerebral blood flow and time-resolved dose delivery to estimate accum

The Galectin Family in Colorectal Cancer: Integrating Molecular Mechanisms with Diagnostic, Prognostic, and Therapeutic Perspectives.

Background: Colorectal cancer (CRC) remains one of the leading causes of cancer-related morbidity and mortality worldwide. Increasing evidence indicates that alterations in glycosylation and glycan-binding proteins, such as galectins, contribute substantially to colorectal carcinogenesis. We review the current knowledge on the role of known human galectins in CRC to provide up-to-date summaries, focusing on their involvement in tumor initiation, progression, invasion, angiogenesis, immune evasion, and treatment resistance, integrating molecular mechanisms with diagnostic, prognostic, and thera

A UK Delphi consensus study on the definition and management of advanced cutaneous squamous cell carcinoma.

BACKGROUND: Advanced cutaneous squamous cell carcinoma (acSCC) encompasses locally advanced and metastatic disease; however, no standardized definition exists. Recognition and categorization, therefore, often rely on multidisciplinary judgment, which may lead to variations in diagnosis and management between centers. This study aimed to establish United Kingdom (UK) consensus on the definition and optimal management of acSCC. METHODS: A modified Delphi methodology was followed. Ten senior clinicians formed a steering group and identified four key topics for consensus statement development sur

Chemokine-Driven Intercellular Crosstalk in the Osteosarcoma Microenvironment After Neoadjuvant Chemotherapy: A Single-Cell RNA Sequencing Study.

Background/Objectives: Osteosarcoma (OS) is an aggressive bone malignancy with a complex tumor microenvironment (TME) that influences therapeutic outcomes and resistance. How neoadjuvant chemotherapy (NACT) reshapes the OS TME at single-cell resolution remains largely undefined. This study aimed to characterize cellular heterogeneity in the OS TME after NACT and identify chemokine-mediated intercellular crosstalk driving chemoresistance. Methods: Single-cell RNA sequencing was performed on surgical specimens from 12 OS patients (6 treatment-naive and 6 post-NACT). After quality control, 77,616

Microfluidic Strategies for Immune Cell Enumeration and Functional Analysis Directly from Whole Blood.

The enumeration and functional analysis of immune cells in whole blood are foundational pillars for disease diagnosis and medical innovation; however, conventional techniques requiring extensive sample preparation often suffer from sample loss, contamination, and phenotypic alterations. Microfluidic technology has emerged as a transformative solution to address these challenges, offering precise spatiotemporal control over cellular microenvironments and enabling integrated analysis directly from whole blood to maximally preserve native cellular states. In this review, we summarize recent advan

Longitudinal dynamics and lineage cross-reactivity of the human antibody response to West Nile virus.

West Nile virus (WNV) is an emerging/re-emerging flavivirus representing one of the major global causes of mosquito-borne encephalitis, yet no approved therapeutics or vaccines currently exist. To characterize the kinetics of WNV-specific immunity, we evaluated the humoral response in a longitudinal cohort of 55 WNV lineage 1-infected blood donors up to 1 year. We demonstrate that WNV infection elicits robust and durable binding and neutralizing antibodies. Using a biphasic decay model, we estimated the half-life of neutralizing antibodies to be 37 days during the rapid decay phase and 472 day

From Glycan Biology to Drug Candidates: An Integrated Sialylation Niche Index and AI-Guided Therapeutic Framework for Head and Neck Squamous Cell Carcinoma.

Background: Head and neck squamous cell carcinoma (HNSCC) responds poorly (<20%) to immune checkpoint blockade. Since sialylation suppresses anti-tumour immunity through Siglec signalling independently of the PD-L1/PD-1 axis, we mapped its prognostic landscape in HNSCC by integrating prognostic modelling, molecular subtyping, and AI-guided drug discovery. Methods: Transcriptomic data from seven GEO cohorts and TCGA-HNSC (n = 501) were analysed with a 1204-gene sialylation compendium. The Sialylation Niche Index (SNI) was built in TCGA-HNSC from 117 machine-learning algorithm combinations (nomi

Postpartum integration of infant vaccines and contraception through gender transformative programming: A cluster randomized trial in India.

Barriers to using contraception in India include norms that promote gender inequality by restricting mobility of women to reach health clinics. We evaluated whether PIVoT (Postpartum Integration of Vaccines and contraception through gender-Transformative programming) improves contraceptive use while enhancing autonomy in contraceptive decision-making. PIVoT delivers the full spectrum of contraceptive methods available along with gender-transformative counseling, linked with community-based infant vaccination. We conducted a cluster randomized trial; 20 Anganwadi Community centers were randomiz

Science still has expertise; what it's lost is authority.

In the years since the pandemic and the political turmoil surrounding the American scientific establishment, much time has been spent discussing why science has lost influence on public policy in the United States. A time when scientific advice about vaccination, nutrition, and environmental regulation was more readily heeded seems a distant memory. Scientific expertise has been decoupled from a sense of authority and standing to dictate policy on matters ranging from public health to climate change. The explanations for this separation include the rise of technologies like the internet and ar

The organization and dynamics of viral factories.

Viral factories (VFs) are dynamic, virus-induced microcompartments that serve as centralized hubs in the host cell for viral genome replication, transcription, and virion assembly. These structures employ unique viral mechanisms for remodeling cellular architecture to create specialized replication organelles and improve the efficiency of viral propagation. VFs exhibit striking structural and functional diversity among RNA and DNA viruses, from reoviruses and poxviruses to the Nucleocytoviricota phylum. Some are enclosed by host-derived membranes, while others exist as biomolecular condensates

Educational background and vaccination beliefs in the Netherlands: A Bayesian Evidence Synthesis from four studies.

BACKGROUND: Although studies repeatedly show that higher educated individuals uphold more positive beliefs about vaccines than their lower educated counterparts, emerging evidence suggests that vaccine skepticism can also be found among highly educated individuals in certain contexts. Given these divergent perspectives on the relation between educational background and vaccine beliefs, the present study investigated (a) the magnitude of educational differences in vaccination attitudes and (b) educational differences in changes in vaccination attitudes over time after the COVID-19 pandemic. Deep Learning-Guided Identification and In Vivo Validation of Compact Cis-Regulatory Elements for the Zebrafish Habenula.

BACKGROUND/OBJECTIVES: Precise genetic access to the zebrafish habenula remains limited by a scarcity of compact, sequence-defined cis-regulatory elements (CREs). Here, we integrated developmental expression mapping, deep-learning predictions on long-range sequences, and in vivo reporter assays to identify compact regulatory sequences driving habenular expression. METHODS: Using a transgenic zebrafish line enriched for habenular reporter expression, we isolated GFP-positive cells from larval brains and profiled their transcriptomes via microarray. A subset of candidate genes enriched in this

Immunogenicity and safety of high-dose versus standard-dose quadrivalent inactivated influenza vaccine in patients living with HIV: a randomized controlled trial.

Influenza remains a major cause of morbidity in people living with HIV (PLWH), and despite recommendations for annual vaccination with the standard-dose (SD) quadrivalent inactivated influenza vaccine (IIV4), immune responses are often suboptimal in this population. To address this gap, we conducted a randomized, single-blinded, controlled trial to compare the immunogenicity and safety of the high-dose (HD) IIV4 (60 μg, Efluelda, Sanofi) with the SD IIV4 (15 μg, Vaxigrip Tetra, Sanofi). Vaccinations were administered between August and December 2023, and seroprotection (SP) was defined as a he
