Chikungunya Virus Infection in Paraffin-Embedded Tissue: Analysis of Histological Alterations and Viral Detection by Immunohistochemistry.

Histopathological analysis of tissues infected with Chikungunya virus (CHIKV), including biopsy and autopsy specimens, can provide valuable insights into the pathogenesis of atypical and fatal cases. By examining tissue architecture and cellular alterations under the microscope, it is possible to identify patterns of injury, inflammation, and cellular degeneration. These morphological findings provide evidence of how the pathogen interacts with host cells and tissues. Moreover, immunohistochemistry may be performed in tissue sections for the detection of viral antigens. Importantly, this techn

Integrative Transcriptomic Network Modeling Coupled with Patient-Derived Validation for Circulating lncRNA Biomarker Discovery in NAFLD: A Comprehensive Workflow.

.: Nonalcoholic fatty liver disease (NAFLD) is a significant global health concern, impacting roughly 25% of people and leading to chronic liver conditions. It involves excess fat accumulation in the liver without significant alcohol intake and can develop into nonalcoholic steatohepatitis (NASH), fibrosis, or cirrhosis. While liver biopsy remains the gold standard for diagnosis, its invasive nature and associated risks restrict its routine use. Noninvasive biomarkers, such as serum ALT, AST, and various composite scores, are available; however, their clinical usefulness is often limited by va

In Silico Single-Cell Frame work for Modeling Intestinal Stem and Transit-Amplifying Progenitor Cells Dynamics.

Single-cell RNA sequencing (scRNA-seq) has revolutionized the ability to resolve cellular heterogeneity within complex tissues, enabling the identification of discrete cell states. Here, we present an in silico analytical pipeline designed to characterize intestinal stem cells (ISC), transit-amplifying (TA) progenitors, and BEST4⁺ enterocyte precursors from human scRNA-seq datasets, with a focus on inflammatory contexts such as inflammatory bowel disease (IBD). The pipeline integrates dataset acquisition, quality control, normalization, dimensionality reduction, unsupervised clustering, and ce

Mechanisms underlying rheumatic heart disease immune-mediated pathogenesis: T-cell responses, cytokine networks, and immune dysregulation as drivers of valvular injury.

Acute rheumatic fever is an immune-mediated sequela of Streptococcus pyogenes (group A Streptococcus) infection, presenting with fever, arthritis, chorea, rash, and, crucially, carditis. Without effective secondary prophylaxis and care, acute rheumatic fever can progress to chronic rheumatic heart disease, a leading yet under-recognised cause of cardiovascular morbidity and death that disproportionately affects children and young adults aged 5-24 years in low-resource settings. Robust evidence implicates the role of antigenic mimicry-particularly between streptococcal M protein and cardiac or

Labeling and Detection of Peptidoglycan in Chlamydia and Chlamydia-Related Organisms Utilizing D-Alanine Analogs.

The incorporation and visualization of modified D-amino acids into the peptidoglycan (PG) of actively replicating microbes have made a tremendous impact on the field of bacterial physiology. Here, we describe the optimization of this technique for labeling PG in obligate, intracellular Chlamydia species as well as Chlamydia-like organisms. Fluorescent D-amino acids (FDAAs) are D-alanine analogs that readily incorporate into the stem peptides of living bacteria and can be visualized rapidly with a fluorescence microscope without the need for laborious postfixation procedures. By comparison, D-a

Quantification of Human T Cell Responses to Chlamydia trachomatis: Intracellular Cytokine Staining (ICS) and Activation-Induced Marker (AIM) Assays.

High-resolution characterization of antigen-specific T cell responses is essential for understanding cellular immunity to Chlamydia trachomatis infection in humans. This chapter describes two complementary flow cytometry-based approaches for measuring human antigen-specific T cell responses: intracellular cytokine staining (ICS) and activation-induced marker (AIM) staining. Both assays enable single-cell-level analysis following antigen stimulation but differ in their readouts. ICS quantifies functional cytokine production in response to short- or long-term stimulation, whereas the AIM assay i

Mitochondrial Genome Characteristics of Rhodobryum laxelimbatum and Comparative Analysis of RNA Editing and Codon Usage Bias With the Chloroplast Genome.

Rhodobryum laxelimbatum is a traditional medicinal plant belonging to the genus Rhodobryum. To date, studies on this genus have focused primarily on its geographical distribution, chemical constituents, and biological activities, whereas genomic data within the genus are limited. The mitochondrial genome of R. laxelimbatum was sequenced, assembled, and annotated. Genomic structural features and simple sequence repeats (SSRs) were comprehensively characterized. Comparative analyses of RNA editing patterns and codon usage bias were performed using the mitochondrial and chloroplast genomes of R.

Phytochemical Investigation of Stachys hissarica: Antioxidant Activity, Flavonoid Characterization, and In Silico Analysis.

Evaluating Stachys hissarica extracts via DPPH, FRAP, and ABTS assays revealed that the aqueous methanolic fraction exhibited strong, ascorbic acid-comparable antioxidant activity. This high potency confirms that hydrophilic phenolic compounds are the primary drivers of its radical-scavenging effect. HPLC-PDA/ESI-MS/MS analysis of the aqueous methanolic extract led to the tentative annotation of 35 metabolites, including hydroxycinnamic acid derivatives, flavonoid glycosides, acetylated and methoxylated flavones, consistent with the phytochemical profile of the Stachys genus. The antioxidant a

Consistency in Mate Preferences by Female Guppies Using Male Chemical Cues.

Females often choose mates using multimodal sensory signals from males. However, in many species, it is common for studies to concentrate on mate preferences for specific sensory signals. The guppy (Poecilia reticulata) is a model species used to study female mate choice. Mate preference studies in female guppies have concentrated on male visual cues, such as color spot characteristics and courtship activity. Although female chemical cues are known to affect mating preferences in male guppies, knowledge of the effects of male chemical cues on female mating preferences is limited. In this study

Effects of High-Altitude Environments on Plumage Lightness (L-value) in Poultry: Melanin-Mediated Mechanisms and Multifactorial Interaction Effects.

High-altitude environments expose poultry to low temperature, hypoxia, and strong UV radiation, but how combined environmental gradients shape plumage lightness remains insufficiently resolved. Using Qiangshan cloud chickens from Wenchuan, we analyzed 150 dorsal plumage samples collected along 900-2800 m. Plumage lightness (L-value) was measured by colorimetry, and eumelanin content was quantified by alkaline dissolution spectrophotometry. Five environmental variables (UV index, altitude, mean annual temperature, annual precipitation, and forest coverage) were integrated from field monitoring,

The NuRD complex shapes RNA polymerase III activity at highly expressed tRNA gene clusters, tuning the dynamic range of cellular tRNA pools.

RNA polymerase III (Pol III) transcription produces RNAs involved in core biological processes and is therefore coupled to fluctuating demands for protein synthesis and cell growth. However, the mechanisms underlying dynamic Pol III regulation remain incompletely understood. To address this gap, we investigated protein-protein interactions shared across multiple Pol III subunits to characterize the macromolecular interactome of Pol III, with special attention directed toward potential regulatory factors. Our survey uncovers interactions between Pol III and the NuRD (nucleosome remodeling and d

Demographic Conscious Analysis: A New Paradigm for Health Research.

Race and ethnicity are frequently misused in research as biological determinants, reinforcing essentialist assumptions and embedding structural factors into clinical guidelines and tools. Disparities in health outcomes increasingly reflect lived experiences and structural inequities, yet standard analytic approaches have not meaningfully evolved. We introduce demographic conscious analysis (DCA), a methodological framework replacing the use of social demographics as predictor variables. Race/ethnicity is a "social demographic" variable that acts as a proxy for causative variables. The social d

Socioeconomic level influences the early motor repertoire of infants at biological risk in a low- and middle-income country setting.

BACKGROUND: Early neurodevelopment is shaped by biological and environmental factors. Prematurity, low birth weight, and socioeconomic adversity are linked to a higher risk of developmental impairments. The General Movements Assessment (GMA) is a reliable tool for early identification of infants at risk for neurodevelopmental impairment. OBJECTIVES: To describe the early motor repertoire of infants followed in a neonatal follow-up clinic using the GMA and to investigate how clinical and socioeconomic factors influence motor outcomes during the first five months of life. A secondary objective

A novel hydrogen persulfide donor HPD1 alleviated LPS-induced neuroinflammation by modulating glial polarization through TLR4-MAPK/NF-κB pathway.

The biological role of hydrogen persulfide (H2S2) has remained elusive due to the scarcity of controllable donors, despite its importance as a reactive sulfur species whose dyshomeostasis drives neuroinflammation. Herein, we employed HPD1, a novel esterase-responsive H2S2 donor, to investigate its anti-inflammatory effects and underlying mechanisms in lipopolysaccharide (LPS)-stimulated glial cells. HPD1 exhibited superior cytoprotective and anti-inflammatory efficacy over inorganic sodium disulfide (Na2S2) and sodium hydrosulfide (NaHS) in BV2 microglia, consistent with its superior and susta

The other side of the coin: health-related applications of some metabolites produced by Pseudomonas aeruginosa.

Pseudomonas aeruginosa is an opportunistic pathogen with high intrinsic and acquired antibiotic resistant levels, so the World Health Organization has defined this bacterium as a priority pathogen for research. In the case of Mexico, different strains belonging to a sequence-type (ST309) that is considered as high-risk due to its wide geographical distribution in different countries, and its high levels of antimicrobial resistance have been isolated, so the research with this bacterium is an active field. In a contrasting way, it has been recognized for decades, that some metabolites produce b

Hierarchical regulation of oxygen-sensing through coupled transducers in Arabidopsis thaliana.

The sensing of molecular oxygen is an essential component of development and environmental response in both animals and plants. Few substrates of the plant oxygen-sensing PLANT CYSTEINE OXIDASE (PCO) N-degron pathway have been identified. Here we show that HYPOXIA TRANSDUCER1 (HYT1, previously named HYPOXIA-RESPONSIVE UNKNOWN PROTEIN, HUP40), an angiosperm-specific tail anchored-domain protein, is a substrate of this pathway through a Cys-2 dependent N-degron. HYT1 is stabilised in response to acute hypoxia in root tissues and is associated with the endoplasmic reticulum and nucleus. Genetic e

Lactate-albumin ratio as an early predictor of mortality in critically ill children: A prospective cohort study.

Early risk stratification in critically ill children relies on severity scores such as PRISM-III and PELOD-2, requiring multiple variables. The lactate-albumin ratio (LAR), a two-parameter marker, shows prognostic value in adult and paediatric critical illness. This study evaluated serial LAR for predicting PICU mortality and compared its performance at admission against PRISM-III and PELOD-2. This prospective cohort study enrolled 335 of 563 screened children aged 1 month to less than 13 years admitted to a tertiary-care PICU in South India over 12 months. Serum lactate and albumin were measu
