BACKGROUND: There have not been any studies to date on trends in digital health literacy (DHL) in Germany. We studied trends in DHL and relevant factors that affect it on the basis of two population-based surveys conducted in 2019/2020 and 2024/2025.
METHODS: Data from the second and third Health Literacy Surveys in Germany (HLS-GER 2, 2019/2020; HLS-GER 3, 2024/2025) were evaluated. Subjective difficulty with digital health information was measured with the HLS19-DIGI survey instrument in two personal cross-sectional surveys of the adult, German-speaking population of Germany.
RESULTS: In H

INTRODUCTION: Proximal hypospadias presents a significant challenge for urethral reconstruction due to the extent of the defect requiring repair. Staged repair with grafting is employed to address this challenge (1). Buccal mucosa and post-auricular skin grafts have been described, but prepuce tends to have lower contraction and failure rates (2, 3). However, the conventional rectangular preputial graft is often of insufficient length for larger defects. In this video, we introduce a novel rainbow technique for preputial skin harvesting in cases of severe proximal hypospadias where an extended

Recent advances in mass spectrometry instrumentation and computational analysis have established label-free data-independent acquisition (DIA) as a powerful tool in quantitative proteomics, offering depth, throughput, and reproducibility. These developments now enable large-scale projects with hundreds or even thousands of samples, opening new applications in systems biology and biomedicine. However, large-scale datasets require special consideration during data preprocessing. Here, we present a practical guide for preprocessing large-scale DIA datasets, illustrated using a public dataset. We

As with any other microscopic technique, in atomic force microscopy (AFM), artifacts and problems can arise. Some of these happen due to improper use of the microscope by the operator, and some are due to particular characteristics of the sample. Some occur depending on the type of instrument or from probe damage. Some of them are artifacts inherent in the technique. Knowledge of these issues is important for correct data acquisition and interpretation, and in many cases, training in AFM is inadequate. In this chapter, we show examples of common artifacts in AFM and describe, where possible, h

Cell's elasticity is an integrative parameter summarizing the biophysical outcome of many known and unknown cellular processes. This includes intracellular signaling, cytoskeletal activity, changes of cell volume and morphology, and many others. Not only intracellular processes defines a cell's elasticity but also environmental factors like their biochemical and biophysical surroundings. Therefore, cell mechanics represents a comprehensive variable of life. A cell in its standard conditions shows variabilities of biochemical and biophysical processes resulting in a certain range of cell's elas

Linking sensor molecules (e.g., antibodies) to an AFM tip converts it into a biosensor by which single target molecules (e.g., antigens) can be detected and localized on the sample surface. Moreover, the mechanism of interaction can be studied by force spectroscopy if purified target molecules are linked to an ultra-flat surface, such as mica or silicon (nitride). Rapid imaging of the binding sites and force spectroscopy studies are greatly facilitated if 6-10 nm long polyethylene glycol (PEG) chains are used as flexible tethers between the sensor molecule and the tip. Here, we describe a set

Native mass spectrometry (nMS) techniques are designed to preserve non-covalent biomolecular complexes for direct observation in an MS instrument. In the past few years, nMS based on NALIM (NAtive LIquid MALDI) has been developed to complement established ESI-based nMS approaches, with the aim to tackle large or poorly soluble protein complexes. It affords a straightforward reading of spectra to monitor oligomer distributions, with applications ranging from the determination of protomers stoichiometry to the presence of peptide or oligonucleotide ligands in complexes. It is well suited to the

Applications of flow cytometry and immunophenotyping offer precise and accurate means for providing information used to both diagnose and monitor disease; they serve as a standard platform for many research endeavors that study discrete populations of biological entities. The proper use of this highly sophisticated technology requires daily and ongoing monitoring of both the instrument and the methodology. Best practices for this begin with quality control (QC) procedures designed to set up and monitor the reagents used, the instrument performance, and the results to ensure that they are worki

Spectral cytometry has rapidly evolved over the past two decades. The development of complete spectral analysis using the flow cytometer as a spectrometer has been actualized due to advances in optics, detectors, and electronics that have made single-cell flow spectroscopy a reality. Polychromatic flow cytometry is a complex process, including the design of antibody panels and instrument compensation. Spectral cytometry has contributed significantly to effectively dealing with the complexity of polychromatic flow cytometry. It has substantial potential to improve primary and clinical research.

Microfluidic tools enable manipulation of microliter-scale volumes within a stable microenvironment. Microfluidics has been widely adopted in various biological fields for, among other applications, controlled delivery of sensory, chemical, and mechanical stimuli to the observed organisms. In this chapter, we describe the design and usage of a custom-made double-layered PDMS microfluidic device, named BUGchip, developed for the purpose of studying plant root growth and metabolic responses in rapidly changing conditions with a high time-scale resolution. We demonstrate the utility of this devic

Mass spectrometry-based proteomics allows the unbiased identification and quantification of proteins and phosphopeptides in biological materials. The nature of walled plant cells requires specific protocols for effective and efficient protein isolation, and, in general, the plant sciences can benefit from more accessible, optimized proteomics workflows. Advances in MS instrumentation now allow the measurement of large numbers of samples, shifting constraints in proteomics toward the accurate, high-throughput preparation of samples. Here, we describe a high-throughput (phospho)proteomics protoc

Proteomics has been revealed as a key set of technologies that provide a detailed description of the molecular processes involved in the development of a specific phenotype. "Omics" technologies can collect an incredible amount of information. Among them, proteomics is an invaluable tool for defining specific biological information by studying the complete set of proteins under specific conditions, the proteome; or specific subsets of proteins, the subproteome. It is a crucial instrument for describing protein post-translational modifications, the functional annotation of the genome, and the d

BACKGROUND: Inadequate access to clinical bacteriology testing in low-resource settings compromises individual patient care and contributes to antimicrobial resistance. Funders, policy makers, and health administrators working to expand bacteriology testing in sub-Saharan Africa require evidence-based guidance. The aim of this study was to assemble a costed consensus list of minimum equipment and consumables required to implement and operate a functional clinical bacteriology laboratory in secondary and tertiary care hospitals in sub-Saharan Africa, to guide funders and health administrators.

Chikungunya virus (CHIKV) is transmitted through the bite of Aedes mosquitoes, specifically A. aegypti and A. albopictus. CHIKV belongs to the alphavirus with a positive-sense ssRNA genome of 11-12 kb size. The virus has been reported from various geographical regions across the globe. Chikungunya fever is an acute febrile illness, which, if left untreated, may develop into chronic arthralgia that may persist for several months or acute encephalitis syndrome. Therefore, early diagnosis of CHIKV is crucial to initiate prompt supportive treatment. Laboratory diagnosis of CHIKV typically relies o

Surveillance of Phytophthora agathidicida , the causal agent of kauri dieback, has included soil baiting followed by loop-mediated isothermal amplification (LAMP)-based detection. This approved assay was reported to detect 1 fg of DNA using OptiGene Isothermal Master Mix on a BioRanger instrument. We evaluated its analytical robustness using the same master mix on an alternative platform (QuantStudio 3), and separately tested an alternative NEB master mix. Both assays yielded a reliable detection limit of 1,000 fg, with variable detection at 100 fg and none below. Diagnostic laboratories shoul

Contemporary headgear assessment techniques apply physical surrogate necks, borrowed from automotive crashworthiness mannequins, despite oft-cited limitations that broadly amount to doubts on their fidelity. Previously, we presented a prototype surrogate neck designed for assessing headgear in direct and oblique head impact leading to simultaneous neck flexion-extension and lateral bending. This study extends the past work by testing the neck in impact-induced axial compression described in previously published cadaver studies. Axial compression is an important impact mode for neck validation

The quality control of commercial Cannabis sativa oil extracts poses analytical challenges due to their complex phytochemical composition, lipid matrix interference, and the risk of adulteration with synthetic cannabinoids. This study evaluated the applicability of two-trace two-dimensional correlation spectroscopy (2T2D-COS) combined with principal component analysis (PCA) for the quality control of full-spectrum cannabis oil extracts, addressing two objectives: detection of MDMB-4en-PINACA adulteration using UV-Vis spectroscopy and discrimination of CBD concentrations using ATR-FT-IR spectro

AP1-AP4 define viability-localized production closure, produced boundaries, admissibility-respecting realization, and existence of a globally coherent viable regime. They do not determine whether a repair relation calibrated in one biological context remains adequate after the context changes. This study develops that second-stage question without introducing a new definition of life or a canonical list of categorical operators. The central construction is a context-indexed family of adequate repair surrogates that separates three claims: local surrogate adequacy in each context, reuse of one
