Determination of tumor extent and tissue characterization is provided with standard spin-echo T1- and T2-weighted imaging and is enhanced by the application of advanced sequences such as gradient-echo, fast spin-echo, and fat suppression techniques. Human NAD(P)H:quinone oxidoreductase2 (NQO2) gene, 1336 base pairs (bp) of the 5'-flanking region and 165 bp of the 3'-flanking region, have been sequenced. Enzymatic sulfation of chiral phenolic ethanolamine drugs, e.g. beta-agonists, has been shown to be stereoselective in humans. A case of pleuropulmonary blastoma (childhood variant of pulmonary blastoma) was examined using histological, immunohistochemical, ultrastructural and cytogenetic methods. The tumour consisted of undifferentiated 'blastematous' areas admixed with zones of rhabdomyoblastic and chondroid differentiation and fascicular areas. Hepatoblastoma is the most frequent primary malignant liver neoplasm in childhood; in adults it is extremely rare and only 27 cases have been published. The prognosis of this neoplasm is poor because it is usually discovered late. Surgery, chemotherapy and liver transplantation have been tried with poor results. We present two adult patients who were diagnosed with an epithelial hepatoblastoma. The pathogenesis, histologic features and current management is reviewed.

Aroclor-1254 (Ar-1254) and dichlorodiphenyltrichloroethane (DDT) were compared to phenobarbital (PB) for their ability to promote hepatocellular proliferative lesions to hepatocellular adenomas and carcinomas and to hepatoblastomas in D2B6F1 male mice initiated with N-nitrosodiethylamine (NDEA). Hepatitis B virus (HBV) nucleoprotein complexes were isolated from nuclei of the human hepatoblastoma cell line HepG2.2.15. Five patients with locally advanced or metastatic liver tumours were treated at the Royal Hospital for Sick Children between 1983 and 1992 by preoperative combination chemotherapy and subsequent complete resection of the residual liver tumour. Chemotherapy was generally well tolerated with few significant adverse effects. Tumour resection was accomplished by lobectomy in three cases and an extended lobectomy in the remaining two. All five children are currently well and free of disease at a mean follow-up of 36 months.

A 5'-flanking region (-2024 to +61) of the guinea pig liver transglutaminase gene and some 5'-deletion mutants were tested for promoter activity in human hepatoblastoma HepG2 cells treated with interleukin-6 (IL-6) by an assay of the transient expression of the chloramphenicol acetyltransferase reporter gene. Confluent monolayers of the human hepatoblastoma-derived cell line, HepG2, were incubated in serum-free medium. This medium was harvested and concentrated. Catalytic activity and immunoblotting of concentrated medium revealed the presence of a functional cholinesterase. This cell line may be an useful model for investigating the regulation and physiological role of the enzyme secreted by liver.
