Reduced T3 deiodination by the human hepatoblastoma cell line HepG2 caused by deficient T3 sulfation.

Type I deiodination of T3 sulfate occurs at a Vmax that is 30-fold higher as compared to T3, both in rat and in human liver homogenates. We now present data showing lack of T3 deiodination by a human liver derived hepatoblastoma cell line, HepG2, caused by deficient T3 sulfation. Cellular entry of T3 was assessed by its nuclear binding after whole cell incubation. In spite of the presence of type I deiodinase, as confirmed by T4 and rT3 deiodination in homogenates, no deiodination of T3 could be detected. Since HepG2 cell homogenates also deiodinated chemically synthesized T3 sulfate (T3S) and

Trends in patterns of treatment of childhood cancer in Los Angeles County.

To assess the proportion of children with cancer who have been managed by mainstream pediatric cancer programs, population-based cancer incidence data for Los Angeles County (LAC) children (under 20 years of age) for the years 1972 through 1987 were linked with patient records of children registered with the two national cooperative pediatric oncology groups, Children's Cancer Study Group and Pediatric Oncology Group. The proportion of children with cancer who were registered by cooperative groups increased markedly over time: 9% of LAC children younger than 15 years of age who were diagnosed

[The modulation of the phenotype of the human hepatoblastoma cell line HepG2 under the action of dimethyl sulfoxide].

Phenotype modulation of the human hepatoblastoma cells HepG2 treated with dimethyl sulfoxide (DMSO) was studied using indirect immunoperoxidase method with monoclonal antibodies against antigens with characteristic localization in the adult liver: cytokeratin 18, membrane antigen AGS5 and embryo-specific alpha-fetoprotein (AFP). Untreated HepG2 cells were found to contain all these antigens. However, their localization was different from that in the adult human liver. Instead of submembrane layer characteristic of liver parenchyma cells in situ, cytokeratin in HepG2 cells formed a cytoplasmic

The mechanism of hepatic cellular injury in sepsis: an in vitro study of the implications of cytokines and neutrophils in its pathogenesis.

Neutrophil-mediated injury to hepatocytes was evaluated in vitro. A new in vitro coculture system of neutrophils and a human hepatoblastoma cell line (HuH-6), instead of normal hepatocytes, was established. Recombinant human tumor necrosis factor (TNF) activated neutrophils to release neutrophil elastase and showed the significant cytotoxicity for HuH-6 cells, which was determined by measuring the release of the cytoplasmic enzyme, lactate dehydrogenase (LDH), from HuH-6 cells. The concentration of neutrophil elastase from zymosan-primed/TNF (1.0 ng/ml)-stimulated neutrophils cocultured with H

Regulation of ferritin and transferrin receptor expression by iron in human hepatocyte cultures.

HepG2 cell cultures and human hepatocyte primary cultures were used to develop appropriate hepatocytic in vitro models of iron load in order to further understand the pathophysiological mechanisms occurring in the liver of patients with hemochromatosis. The first step of this study was to obtain an efficient iron supply in conditions of minimal toxicity. It was demonstrated that iron complexed to citrate entered efficiently into HepG2 cells and human hepatocytes. This iron load was obtained with minimal toxicity in both culture models as evaluated by the intracellular LDH activity and the tota

Interleukin-6 inhibits corticosteroid-binding globulin synthesis by human hepatoblastoma-derived (Hep G2) cells.

Corticosteroid-binding globulin (CBG) belongs to the superfamily of serine proteinase inhibitors which include alpha 1-antitrypsin, alpha 1-antichymotrypsin, and T4-binding globulin. Interleukin-6 (IL-6), the main mediator of the acute phase phenomenon, increases alpha 1-antitrypsin and alpha 1-antichymotrypsin synthesis and decreases T4-binding globulin synthesis by human hepatoblastoma-derived (Hep G2) cells. This effect is predominantly at a transcriptional level. When Hep G2 cells were exposed to different concentrations of IL-6 for variable time intervals, IL-6 caused a dose- and time-dep

Heat shock gene expression and development of translational thermotolerance in human hepatoblastoma cells.

Synthesis of heat shock proteins is a conserved cellular response to disparate environmental stresses, including circulatory shock/resuscitation. Heat shock proteins are believed to play a cytoprotective role within the cell. To understand the regulation of the heat shock response, the kinetics of expression of the major inducible heat shock protein (hsp-72) was studied in a simple model: human hepatoblastoma (HepG2) cells in culture under thermal stress. At both mild (41 degrees C) and moderate (43 degrees C) heat shock temperatures, new synthesis of hsp-72 was proportional to the steady-stat

Hypercalcemia complicating childhood malignancies.

BACKGROUND: Hypercalcemia complicating malignancy is a frequent complication in adults, but little has been published about the pathogenesis or the true incidence of hypercalcemia in children with cancer. METHODS: Hypercalcemia developing in childhood malignancies was studied retrospectively at St. Jude Children's Research Hospital to determine its incidence, the timing of its presentation, and its response to therapy. RESULTS: Over a 29-year period, 25 children (median age, 9.5 years) had been diagnosed and treated for hypercalcemia that occurred during the course of their malignancy. These

Melanin-containing hepatoblastoma with endocrine differentiation. An immunohistochemical and ultrastructural study.

BACKGROUND: The authors previously have reported that hepatoblastomas may exhibit endocrine differentiation. This report describes a hepatoblastoma in which a melanocytic component was present in addition to endocrine differentiation. METHODS: The tumor, which arose in a 15-month-old girl, was subjected to conventional histologic, histochemical, immunohistochemical, and electron microscopic investigation. RESULTS: The tumor had fetal and embryonal epithelial areas and osteoid. The presence of melanin could be suspected, even on the basis of gross examination. The melanin was found predominan

Human thyroxine-binding globulin gene: complete sequence and transcriptional regulation.

T4-binding globulin (TBG) is a glycoprotein of hepatic origin which transports thyroid hormone in serum. To characterize the human TBG (hTBG) gene, we studied its genomic organization, promoter activity, and regulation. To this purpose, we isolated from liver a complete hTBG cDNA clone containing the 5'-untranslated region and localized the transcription start site (TSS). The analysis of genomic clones revealed that the hTBG gene consists of five exons and that its exon-intron organization is similar to that of other members of the serine protease inhibitor family. The first exon (exon 0) is a

Effect of X protein on transactivation of hepatitis B virus promoters and on viral replication.

The X gene product of hepatitis B virus (HBV) transactivates a wide variety of promoters, including four promoters on the HBV genome (Rossner, 1992, J. Med. Virol. 36, 101-117). We compared their transactivation efficiencies and investigated whether the spatial organization of the promoters with respect to other cis-acting elements might influence their activities. Eight reporter plasmid constructs containing the bacterial chloramphenicol acetyltransferase (CAT) gene were designed such that four had the isolated HBV promoters linked to the CAT gene. In the other four, the CAT gene was inserted

Cytokine regulation of low density lipoprotein receptor gene transcription in HepG2 cells.

Elevated plasma levels of cytokines have been demonstrated in inflammatory, malignant, and infectious diseases. These disease states are often associated with abnormal lipid metabolism and reductions in plasma cholesterol levels. To determine if inflammatory cytokines could influence hepatic lipid metabolism, we evaluated low density lipoprotein (LDL) receptor function and gene expression in cytokine stimulated HepG2 cells, a hepatoblastoma-derived cell line which shares many functional similarities with normal hepatocytes. Tumor necrosis factor-alpha (TNF) and interleukin-1 beta (IL-1) increa

Production of alpha-fetoprotein by human submandibular gland.

Alpha-fetoprotein (AFP) is a 590 amino acid polypeptide that was initially defined as an embryonal serum globulin. The yolk sac endoderm, fetal liver and fetal gut were shown to be the main sites of AFP synthesis in the embryo (Gitlin and Boesman, J. Clin. Invest. 46: 1010-1016, 1967). AFP synthesis is still continued in human adults (Ruoslahti and Seppala, Int. J. Cancer 8: 374-383, 1971) although the physiological level of serum AFP is lower than 10 ng/ml. AFP was also demonstrated in certain tumors and in various diseases or conditions such as yolk sac tumor, hepatoma, hepatoblastoma, acute

Mutation at codon 249 of p53 gene in a human hepatoblastoma.

G-to-T transversion at codon 249 of the p53 gene has been shown to be specifically associated with human hepatocellular carcinomas, particularly that subset associated with exposure to the chemical hepatocarcinogen aflatoxin B1. We surveyed 47 North American adult hepatocellular carcinomas and three childhood liver tumors for codon 249 mutation. We report here a case of childhood hepatoblastoma in a patient, without known exposure to aflatoxin B1 or hepatitis B or C virus, whose tumor had a mutation at codon 249 involving G-to-T transversion.

Regulation of the human asialoglycoprotein receptor by cAMP.

The mechanism of regulated expression of the asialoglycoprotein receptor (ASGR) by cAMP was investigated in the human hepatoblastoma cell line, HepG2. Incubation of HepG2 cells with the cell-permeant 8-bromo-cAMP or induction of intracellular cAMP with forskolin reduced receptor expression in confluent HepG2 cultures. Immunoblot analysis established that this reduction of receptor activity was due to a reduction of expression of both ASGR subunit polypeptides H1 and H2. Estimates of the steady-state levels of H1- and H2-related mRNA by Northern blot analysis indicated that reduced ASGR express
