This study shows that the human hepatoblastoma cell line Hep G2 constitutively expressed a high level of Leukemia Inhibitory Factor (LIF) mRNA in the characteristic major 3.8 and minor 1.8 Kb forms. DNA analysis of the LIF gene from Hep G2 revealed no rearrangements. Production and secretion of significant concentrations of LIF were demonstrated by enzyme-linked immunoabsorbent assay (ELISA) in culture supernatants of Hep G2 cells. Antitumor activity of cisplatin, carboplatin, etoposide, or human hepatocyte growth factor (hHGF), was compared by examining the colony formation ability of four liver cancer cell lines (PLC/PRF/5 and HuH-7, hepatocellular carcinoma; HuH-6 and HepG2, hepatoblastoma). We analyzed the status of retinoblastoma and p53 genes in 10 human hepatoma cell lines. Polyclonal anti-peptide antibodies generated against peptides homologous to COOH-terminal and leucine-zipper domains of the retinoblastoma protein allowed us to identify two cell lines (Hep 3B and FOCUS) with abnormal expression. Rat hepatocytes exhibit bidirectional carrier-mediated transport of reduced glutathione (GSH) across the plasma membrane. Transport of GSH has not been well characterized in human-derived cells. Detection of an abdominal mass in a child is an alarming finding for both parent and practitioner. Diagnostic studies including ultrasonography, computed tomography, and magnetic resonance imaging provide an accurate diagnosis in most cases. The current treatment of Wilms' tumor results in an 85% to 90% survival. Hepatoblastoma and neuroblastoma often present in an advanced stage and require preoperative chemotherapy before resection is attempted. Improved survival for children with these tumors awaits better detection, chemotherapy, and understanding of the biology of their growth.
