By means of two-dimensional lectin affinity electrophoresis of human alpha-fetoprotein (AFP) from different sources, AFP bands separated with erythroagglutinating phytohemagglutinin (E-PHA) were further characterized with other lectins of known oligosaccharide specificities. During the initial 4 h of treatment, copper and zinc similarly activated the rates of transcription and mRNA accumulation from the two human metallothionein (MT) genes, viz., MTI-G and MTII-A, in the hepatoblastoma cell line HepG2. The levels of copper-induced MT mRNAs remained at a plateau for up to 15 h. In contrast, the levels of zinc-induced MT mRNAs gradually declined after about 4 h, despite substantial transcription. The decrease in the zinc-induced MT mRNA half-life is probably due to a posttranscriptional event(s).

Four patients presented with large intra-abdominal masses. Three patients were diagnosed as having hepatoblastomas of the right hepatic lobe, one of the left lobe. Initially these tumors were unresectable, and after biopsy each patient was treated with a standardized chemotherapy protocol. All tumors decreased remarkably in size and were resected successfully. The role of insulin-like growth factor-II (IGF-II) in the hypoglycemia associated with nonislet cell tumors is controversial. In this study we have addressed this question by measuring the IGF-II mRNA levels in extracts of these tumors. The effect of various factors and substrates on the growth of a human hepatoblastoma cell line, HuH-6, which was inoculated at low density in a serum-free medium was examined. Several supplements were required to enhance cell growth of HuH-6. These included cholera toxin (CT), glucagon (Glu) and selenium (Se). Type IV collagen (C-IV) provided the most conductive environment tested for cell growth. These results suggest that CT, Glu, Se, and C-IV are important stimulators for the continuous growth of HuH-6 in a serum-free medium at low density.

A tumor model was developed in DBA/2 mice for studying the progression of the mastocytoma P 815 X 2 tumor. Tumor cells obtained from ascites were grown in vitro. The number of cells derived from a single clone was increased by in vitro culture. Cells were then injected either i.v. or i.p. into DBA/2 mice. The feasibility of 2-deoxy-2-[18F]fluoro-D-galactose ([ 18F]FdGal) for imaging galactose metabolism in tumors with positron emission tomography (PET), was investigated using two hepatomas, Yoshida sarcoma, or glioma in rats, and mouse mammary carcinoma.
