The effect of uracil on the metabolism of 5-fluorouracil (5-FU) in vitro was studied. 5-FU was mainly phosphorylated in intact Yoshida sarcoma cells, whereas it was mainly degraded in liver slices. Uracil inhibited degradation of 5-FU much more than its phosphorylation; incubation of 2,500 microM of uracil with 2.5 microM of 5-FU (molar ratio, 1,000:1) inhibited the degradation of 5-FU by 70%, but did not affect its phosphorylation. The antimetastatic effect of Triton WR 1339 (TWR), a nonionic detergent, was evaluated in the rats bearing ascites tumor (Yoshida sarcoma or AH 66F). TWR did not inhibit the growth of primary tumor (ascites production). However, a marked inhibition of metastasis was observed in the lung and liver of rats treated with TWR. The results obtained suggest that TWR exerts antimetastatic effect before the entry of tumor cells into vascular channels. With other tests, it was found that TWR inhibits the release of tumor cells from tumor graft.

Two sarcomas that appeared as distinct solitary nodules in the walls of ovarian mucinous cystic tumors are described. One of the tumors was a fibrosarcoma associated with a mucinous cystadenoma; the other was an undifferentiated sarcoma in a mucinous cystadenocarcinoma. Both patients died within 1 1/2 years of their operations with distant metastasis; one of them had had metastatic sarcoma in the para-aortic lymph nodes at the time of exploration. These two cases are the only well-documented examples of this unusual combination of tumor types.

Using the indirect immunoperoxidase method, alpha 1-fetoprotein (AFP) can be demonstrated in paraffin sections of tumours by light-microscopy. Among 160 tumour preparations it was demonstrated in five of six endodermal sinus tumors, 19 of 40 teratomas, the three hepatoblastomas and 9 of 16 primary liver-cell carcinomas. The carcinogenicity of aflatoxin B1 (AFB1) has been under evaluation in nonhuman primates for the past 13 years. A total of 47 Old World monkeys, chiefly rhesus and cynomolgus, have received AFB1 i.p. (0.125 to 0.25 mg/kg) and/or p.o. (0.1 to 0.8 mg/kg) for 2 months or longer, and 12 are currently alive and without evidence of tumor. Subcutaneous injection of 1,2-dimethylhydrazine into female CBA mice once a week resulted in the development of tumours of the colon, anal region, uterus and liver. In 12-13-month-old mice treated with DMH an earlier appearance (week 8) of uterine sarcomas and more rapid increase in the incidence of tumours of the anal region were noted as compared to 3-month-old mice. In pregnant females treated with DMH a statistically significant decrease in the uterine sarcoma incidence was observed (10.3% versus 48.3% in nonpregnant). Pregnancy exerted no effect on the incidence of tumours at other sites.

A 4 1/2-year-old girl is described in whom the portal vein is absent and the venous return from the gastrointestinal tract, spleen, and pancreas is through the left renal vein into the inferior vena cava. She had a hepatoblastoma, which was treated surgically by right hepatic lobectomy. Regeneration occurred in the remaining left lobe within 3 months after surgery. This is the second such case documented in the literature.

The incidence of primary liver cell carcinoma was investigated in a prospective study over 6 yr and 5 mo in 403 clinically unselected patients derived from a homogeneous population by means of serial determination of alpha 1-fetoprotein (AFP) by radioimmunoassay.
